摘要
目的侵袭是恶性肿瘤转移发生最初的一步,黏附、蛋白水解及细胞游走能力是影响侵袭的3个主要因素。文中旨在通过扫描电镜观察膀胱肿瘤细胞在体外侵袭实验中的侵袭过程。方法采用大鼠ESH肉瘤细胞外基质提取Matri-gel构建侵袭小室,BIU-87-m3常规体外侵袭7 h,扫描电镜观察侵袭膜的室腔面及背面,了解肿瘤细胞侵袭情况。结果侵袭膜室腔面观察到膀胱肿瘤细胞贴附于Matrigel表面生长,向聚碳酸酯膜上的小孔伸出伪足及正在穿过Matrigel及聚碳酸酯膜上小孔;背面观察到膀胱肿瘤细胞穿过侵袭膜后贴附于侵袭膜背面生长及穿过侵袭膜的伪足。结论从形态学上证实了肿瘤细胞侵袭特性的存在及传统体外侵袭实验设计的科学性和体外检测肿瘤细胞侵袭力的可行性。
Objective Invasion is the initial step of malignant tumor metastasis.Adhesion,proteolysis and transmigration ability of cells are three main influencing factors of invasion.The purpose of this study was to observe the invasion process of bladder tumor cells in vitro under the scanning electron microscope.Methods The invasion chamber was constructed using Matrigel and the bladder tumor cells of BIU-87-m3 were subjected to invasion for 7 hours in vitro.Then the cavosurface and dorsal surface of the constructed membranes were observed under the scanning electron microscope,and the invasion of the tumor cells was analyzed.ResultsBladder tumor cells were seen growing on the cavosurface,stretching out their pseudopodia towards and going through the microbore of the polycarbonate adipose membrane.On the dorsal surface,the bladder tumor cells penetrated the constructed membrane,adhered there and grew,with their pseudopodia stretching out from the cavosurface.Conclusion The characteristic of tumor invasion was morphologically confirmed.The invasion experiment in vitro is scientific and feasible for the determination of the invasiveness of tumor cells.
