摘要
目的观察不同温度刺激K562细胞热休克蛋白gp96的基因转录及蛋白表达的影响。方法将培养获得的K562细胞分别进行40℃、45℃、50℃恒温刺激30min,采用逆转录-聚合酶链反应技术检测gp96mRNA的表达和采用western bloting技术检测gp96蛋白的表达。结果各组gp96mRNA的转录结果为:对照组0.61±0.15,40℃组0.93±0.09,45℃组1.32±0.11,50℃组1.57±0.07,各组之间可见显著性差异(P<0.05)。各组细胞内表达gp96蛋白的量分别为,对照组2.15±0.18,40℃组2.94±0.12,45℃组3.45±0.24,50℃组5.21±0.09。各组间差异有统计学意义(P<0.05)。结论温热刺激可提高K562细胞内gp96mRNA的转录水平,并且随着温度的不断提高,gp96的表达快速增高。
Objective To observe the expression of gp96 and the transcriptional levels of gp96 mRNA in K562 cell line after the heat stimulation.Methods After the 40℃,45℃,50℃ constant temperature stimulation for 30 minutes respectively,the transcriptional levels of gp96mRNA were detected by RT-PCR technique and the expression of gp96 were detected by western blotting technique.Results The results of the gp96 mRNA transcription: Control:0.61 ±0.15,40℃ :0.93±0.09,45℃:1.32±0.11,50℃:1.57 ± 0.07.There were significant differences among every group ( P <0.05).The results of the protein expression of gp96:Control:2.15 ± 0.18,40℃: 2.94± 0.12,45℃ : 3.45± 0.24,50℃: 5.21 ± 0.09.There were significant differences among every group ( P <0.05).Conclusion Heat stimulation can elevate the transcriptional level of gp96mRNA.With the temperature stepping up gradually,the expression of gp96 increases quickly.
