电针对阿尔茨海默病大鼠纹状体神经细胞超微结构的影响 被引量:6

Study on the influence of Electro-acupuncture on neural ultrastructural change in the corpora striatum of rats with Alzheimer's disease

摘要 目的探讨电针治疗对Aβ所导致的大鼠脑内神经元损伤发挥保护作用的机制。方法将凝聚态的Aβ1-40立体定向注入大鼠双侧M eynert核造成痴呆模型,通过电针足三里、太溪、百会穴进行治疗,透射电镜观察电针对AD大鼠纹状体神经细胞超微结构的影响。结果模型组大鼠纹状体可见一些神经细胞变性,胞膜皱缩,细胞体积缩小,细胞浆和细胞器密度增高,核染色体固缩,核膜间隙增宽,其内细胞器可见内质网扩张,线粒体肿胀、空泡样变。电针组大鼠纹状体神经元细胞核基本正常,细胞器可见线粒体轻度肿胀。结论电针治疗可抑制注射Aβ导致的大鼠纹状体的神经细胞损伤变性,促进神经细胞的修复,改善神经细胞功能,从而对AD模型大鼠的学习记忆障碍起到调节改善作用。 Objective To explore the mechanism of the neuroprotective effects of electro-acupuncture(EA) on treating rat's cerebral neuron injury induced by injection Aβ.Methods The model of Alzheimer disease was established by stereotaxis injecting agglutinated Aβ1-40 into rat's bilateral Meynert basal nuclei.The rat with Alzheimer disease was treated by EA at the points of'Zusanli'(ST 36)、'Taixi'(KI 3) and 'Baihui'(DU 20).And the neural ultrastructural change in the corpus striatum of rats were observed by transmission electron microscope.Results In the corpus striatum of the rats in model group,we observed some degenerated nerve cells with cell membrane shrinked,the volume of the cell contracted,electron density in cytoplasm and organelle densified,nuclear membrane space widened,endoplasmic reticulum of inter organelle expanded and mitochondrion swelled.However,in the corpus striatum of the rats in EA group,cell nucleus was almost normal and mitochondrion in organelle swelled slightly.There weren't any above-mentioned changes in the nerve cells in the corpus striatum of the rats in normal group and sham-operated group.The cell membrane was integral,electron density in ground substance of cytoplasm was well-distributed,biomembrane was integral and the structure was clear.There weren't obvious pathological changes in the nerve cells of the rats in these two groups.Conclusion EA therapy can restrain the nerve cells degeneration in corpus striatum in rats caused by injection of Aβ,promote the reparation of nerve cells and ameliorate the function of nerve cells,so as to regulate and improve the learning disorder and dysmnesia of the rat with Alzheimer's disease.
