IFN-γ、IL-10对HL-60细胞表达B细胞活化因子的影响

IFN-γ and IL-10 upregulate the expression of B-cell activating factor in human HL-60 cells

摘要 B细胞活化因子(BAFF)的异常表达与骨髓瘤、淋巴瘤的发病相关,为探索常见炎性细胞因子是否能影响人骨髓白血病细胞HL-60中BAFF的表达,以细胞因子IFN-γ、IL-10和IL-4刺激体外培养的HL-60细胞1~3 d后,采用流式细胞术、ELISA和荧光定量RT-PCR方法检测BAFF的核酸和蛋白表达含量的变化。结果显示,细胞因子IFN-γ、IL-10干预后,HL-60细胞表面BAFF蛋白表达水平、细胞上清液中BAFF蛋白含量、细胞内BAFF mRNA的含量均能明显升高(P<0.05),其中在INF-γ、IL-10干预的第1天,HL-60细胞内BAFF mRNA的含量为最高;在干预的第2天,HL-60细胞的上清液中BAFF蛋白含量及细胞表面BAFF蛋白表达水平均达最大值;而IL-4干预HL-60细胞后,仅对细胞上清液中BAFF蛋白水平稍有降低(P<0.05),细胞表面BAFF蛋白表达水平和细胞内BAFF mRNA的含量均无显著性差异。研究提示,INF-γ、IL-10可能是通过促进HL-60细胞对BAFF核酸的表达来上调BAFF蛋白的合成与分泌,而IL-4则仅能一定程度的降低HL-60细胞对BAFF蛋白的分泌...
