鞘氨醇调节人胰腺癌细胞株JF305生长和钙代谢 被引量:1

GROWTH REGULATING OF HUMAN PANCREAS CARCINOMA CELL JF305 BY SPHINGOSINE AND THE METABOLIZATION OF CALCIUM

摘要 目的 :观察鞘氨醇对人胰腺癌细胞株 JF30 5的增殖作用 ;观察鞘氨醇对人胰腺细胞株 JF30 5内Ca2 + 浓度的影响 ,探讨鞘氨醇的细胞信使作用。方法 :细胞培养后 ,分别加入不同浓度的鞘氨醇 ,应用四甲基偶氮唑盐 MTT比色法来观察细胞增殖程度 ;应用 Fura- 2 / AM荧光比色测定 [Ca2 + ]i 结果 :鞘氨醇促进人胰腺癌细胞株JF30 5的生长 ,促进 JF30 5内 Ca2 +的生成 ,3min内达峰值 ,并在 0 .5 μ m ol/ L~ 2 .0 μ mol/ L 时呈剂量依赖性。结论 :鞘氨醇能促进人胰腺癌细胞株 JF30 5增殖 ;鞘氨醇的信使作用是通过刺激 Ca2 +转运的和 Ca2 Objective: To observe the growth promoting role of sphingosine (SPH) on human pancreas carcinoma cell strains JF305 and to study the role of sphingosine in signal transduction by measuring the concentrations of intracellular free calcium.Methods:JF305 cells were in medium,then were treated with SPH of different concentrations.Proliferation and viability of cell was measured by the Mosmann's method (MTT).Intracellular Ca 2+ level were measured using Fura 2/AM by a F 2000 flurorescence spectrophotomet...
