摘要
目的 建立细胞间粘附分子 1 (ICAM 1 )抑制剂的高通量筛选方法。方法 用脂多糖 (LPS)激活原代培养的人脐带内皮细胞 ,用酶联免疫法 (ELISA)测定内皮细胞激活后粘附分子ICAM 1的表达。用MTT法检测所筛化合物的毒性作用。结果 LPS时间和剂量依赖性地激活内皮细胞表达ICAM 1 ,以质量浓度为 1mg·L-1 和刺激时间为2 4h为佳。在对 2 0 0 0个化合物的初筛中 ,发现 30个化合物可抑制ICAM 1的表达 ,入围率为 1 5 % ,其中 2 4个化合物有细胞毒作用。结论 ELISA方法成本低、重复性好 ,效率高 ,适合ICAM
Aim To develop a high throughput screening assay to identify inhibitors of intercellular adhesion molecule-1 (ICAM-1) expression in human umbilical vein endothelial cells (HUVEC). Methods ICAM-1 expression in lipopolysaccharide-stimulated endothelial cells was measured by ELISA. The cytotoxicity of the compounds was measured by MTT.
