摘要
目的建立了高效液相色谱法-荧光检测器测定食品中De含量的方法,探讨样品测定的影响因素。方法样品用甲醇+水(80+20,v/v)超声提取30 m in,离心定容后过0.45μm滤膜,经200×4.6mm 5μm的C18柱分离,以甲醇+水作流动相,梯度洗脱,荧光检测器进行测定,检测器激发波长Ex=366 nm、发射波长Em=417 nm。结果方法线性范围为0.5μg/m l-30μg/m l,回归曲线为Y=13924X-8625,r=0.9995。样品的加标回收率为90.2%-108.4%,RSD在0.93%-5.2%之间。结论该方法前处理简单,干扰少,准确快速,可作为测定保健食品中大豆异黄酮含量的方法。
objective To establish a method for the determination of soybean isoflavones in health foods by HPLC.Methods After sonicated for 30 min and filtered through the 0.45 μm filter,the samples were separated on a C18 reversed-phase colum(200×4.6 mm 5 μm) using methanol +wateras the mobile phase,with gradient elution and detected by fluorescence detector. Ex=366nm,Em=417nm.Results The content was linear over the range 0.5 μg/mL-30 μg/mL.Y=13924Y-8625,r=0.9995.The recovery of the added sample De was 90.2%-108.4%,RSD was between 0.93%-5.2%.Conclusion With its accuracy,sensitivity,reproducibility as well as the simplicity of sample preparation,the method was feasible for the determination of soybean isoflavones in health foods.
