摘要
目的探讨牛磺酸对大鼠视网膜缺血再灌注损伤的保护作用及机制,为牛磺酸对视网膜缺血再灌注损伤的治疗方面的研究提供理论依据。方法将36只Wistar大鼠随机分为3组:正常组(A组)、缺血再灌注组(B组)、牛磺酸治疗组(C组)。通过升高眼内压造成视网膜缺血1h后,再恢复正常眼内压而建立视网膜缺血再灌注损伤动物模型,并在术前3d连续皮下注射牛磺酸200mg/(kg.d)。采用原子吸收光谱法测定视网膜组织细胞内钙离子的变化,酶化学法检测视网膜组织中MDA含量、SOD活性、GSH含量的改变。结果B组视网膜组织中细胞内钙离子含量和MDA含量明显高于A组(P<0.01),而SOD活性、GSH含量低于A组(P<0.01)。C组视网膜组织细胞内钙离子和MDA含量低于B组(P<0.01),而SOD活性、GSH含量高于B组(P<0.05)。结论大鼠视网膜缺血再灌注损伤后组织钙超载,自由基产生增多;牛磺酸能够减轻钙超载、抑制视网膜自由基的产生和增强氧自由基清除,提高视网膜的抗损伤和抗氧化能力,有望临床用于视网膜缺血再灌注损伤保护的治疗。
Objective To investigate the therapeutical effect and mechanisms of taurine on retina ischemia-reperfusion injury,then to provide the possible theoretical evidence.Methods Thirty-six Wistar rats were randomly divided into three groups,normal control(group A),ischemia-reperfusion(group B)and taurine-treated(group C).Retinal ischemia-reperfusion model was induced by increasing intraocular pressure for 60 min,then recovering to normal intraocular pressure.The rats in Group C were subcutaneously injected with taurine three days before establishing the model.Atomic absorption spectrum method was taken to evaluate the intracellular calcium changes of retinal tissues.The content change of MDA,GSH and the activity change of SOD of retina tissues was detected by enzyme chemistry method.Results The intracellular calcium and MDA contents of retinal tissues of group A were less than those of group B(P<0.01).The activity of SOD and content of GSH of retinal tissues of group B were less than those of group A(P<0.01).The intracellular calcium and MDA contents of retinal tissues of group C were less than those of group B(P<0.01).The activity of SOD and content of GSH of retinal tissues of group C were more than those of group B(P<0.05).Conclusion The intracellular calcium and free radical increase after the retina ischemia-reperfusion injury.Taurine could inhibit the increasing of the intracellular calcium and free radical,enhance the cleaning of free radical and prompt the capability of anti-injury and anti-oxidation of the retina,so could be applied to the treatment of retina ischemia-reperfusion injury.
