人骨髓间充质干细胞的培养及细胞增殖测定 被引量:3

摘要 目的分离培养并鉴定人骨髓间充质干细胞。方法成人骨髓取自非血液系统疾病、非肿瘤及乙肝表面抗原阴性的胸外科手术中摘取的肋骨,采用密度梯度离心法和贴壁筛选法进行培养;免疫细胞化学法鉴定细胞膜抗原。结果人骨髓间充质干细胞(MSCs)的CD44表达阳性;早、中、晚3代传代培养具有以下共性特征:传代培养潜伏期约为24~36h;传代培养对数增殖期约为4~6d;对数增殖期结束后至接种后第9d,MSCs生长逐渐缓慢,进入平台期。结论培养的细胞不是骨髓中的造血干细胞或骨髓基质中的成纤维细胞,是处于未分化阶段的骨髓间充质干细胞。 Objective To culture fractionally and identify human bone marrow mesenchymal stem cells(MSCs).Methods Human bone marrow was obtained from ribs without blood diseases,tumors,and hepatitis B,and was cultured by density gradient centrifugation and adherence screening method.The cellular membrane antigen was identified with immunocytochemical method.Results The expression of CD44 in MSCs was positive;the serial subcultivation of the early,middle and late period had the following similarities:the eclipse period of serial subcultivation was about 24~36 h; the log multiplication period was 4~6 days; form the ending of the log multiplication period to 9 days after inoculation ,MSCs grew slowly and went to a platform period.Conclusion The cultured cells are neither hemopoietic stem cells nor fibroblasts but the MSCs that have not differentiated.
