摘要
目的:探究中药复方活血灵预防骨折后深静脉血栓(DVT)作用机制。方法:60只1月龄Wistar大鼠随机平均分成空白组,对照组,阳性药物组,中药HXL低、中、高剂量组,采用创伤性造模方法制备大鼠下肢DVT模型;各组在造模前1周灌胃,每天1次,于造模后第7天取各组大鼠静脉组织做HE切片观察血管形态改变;取用各组大鼠腹腔血液检测血流变及相关凝血指标,并采用ELISA检测组织中IL-1、IL-6、TNF-α水平改变;取用发生血栓处静脉组织用生化法检测ROS水平、SOD活力、MDA含量。结果:HE染色显示,空白组无血栓形成,对照组为完全性血栓,阳性药物组、中药HXL低、中、高剂量组有不完全血栓形成及少量炎症细胞浸润;各组与对照组凝血指标比较,差异有统计学意义(P<0.05),阳性药物组APTT、PT、TT、D-dimer值与中药HXL高剂量组比较,差异有统计学意义(P<0.05),中药HXL高剂量组对凝血指标改善优于阳性药物组;各组与对照组炎症因子比较,差异有统计学意义(P<0.05),阳性药物组IL-1、TNF-α、IL-6水平与中药HXL高剂量组比较,差异有统计学意义(P<0.05),中药HXL高剂量组对炎症因子改善优于阳性药物组;各组与对照组的氧化应激指标对比,差异有统计学意义(P<0.05),阳性药物组ROS、SOD、GSH-Px、MDA值与中药HXL高剂量组比较,差异有统计学意义(P<0.05)。结论:复方活血灵可能通过抗炎、抗氧化作用来预防骨折后DVT。
Objective:To explore the mechanism of the prevention of deep vein thrombosis(DVT)after fracture by Compound Huoxueling.Methods:Sixty one-month-old Wistar rats were randomly divided into blank group,control group,positive drug group,traditional Chinese medicine Huoxueling low,medium and high dose group.The changes of blood flow and related coagulation parameters were observed in each group in the first week of the model,and the changes of the blood flow and the related coagulation indexes were taken from the abdominal blood of each group,and the levels of IL-1,IL-6 and TNF-αin the tissues were detected by ELISA.The level of ROS,the activity of SOD and the content of MDA were detected by biochemical method.Results:HE staining showed no thrombus formation in blank group.The control group was complete thrombus,positive drug group,traditional Chinese medicine Huoxueling low,middle and high dose group had incomplete thrombus formation and small amount of inflammatory cell infiltration,and the difference of blood coagulation index in group and control group was statistically significant(P<0.05).The difference of APTT,PT,TT and D-dimer in the positive drug group was statistically significant than traditional Chinese medicine Huoxueling high dose group(P<0.05).The improvement of coagulation index of traditional Chinese medicine Huoxueling high dose group was better than positive drug group.Inflammatory factors in control group was significant than other groups(P<0.05).The levels of IL-1,TNF-α,and IL-6 in the positive drug group were significantly higher than that of the high-dose group(P<0.05).The improvement of inflammatory index of traditional Chinese medicine Huoxueling high dose group was better than the positive drug group.The oxidative stress index of control group was significant with other groups(P<0.05).The values of ROS,SOD,GSH-Px and MDA in the positive drug group were significantly higher than those of the high-dose group(P<0.05).Conclusion:Compound Huoxueling may prevent DVT after fracture through anti-infl
