摘要
目的分析暴露于香烟烟雾下的雄性SD大鼠肝组织生化水平及氧化应激指标的变化,并观察大鼠肝脏组织病理改变,探讨香烟烟雾暴露对大鼠肝脏毒性的影响。方法选用清洁级SD雄性大鼠40只,分成低、中、高剂量组(即每日10、20、30支香烟)及正常对照组,每组10只,以呼吸道静式染毒方法连续染毒4周。测定各组大鼠肝组织中谷丙转氨酶(ALT)、谷草转氨酶(AST)、血清总胆红素(TBIL)、肝组织丙二醛(MDA)、超氧化物歧化酶(SOD),并通过HE染色观察大鼠肝脏病理改变。结果随着染毒剂量的增加,大鼠体重及肝脏重量有下降趋势,差异有统计学意义(P<0.05)。染毒组大鼠肝组织中AST、ALT活力及二者比值均较正常对照组升高,差异有统计学意义(P<0.05);染毒组大鼠血清TBIL较正常对照组有升高趋势,但差异无统计学意义(P>0.05)。染毒组大鼠肝组织MDA含量较正常对照组升高,SOD活力较正常对照组下降,差异均有统计意义(P<0.05)。染毒组大鼠的肝脏较正常对照组均有不同程度的病理改变。结论香烟烟雾暴露可抑制大鼠体重增长及肝脏发育,且对肝脏组织生化指标及氧化应激水平产生一定影响。
Objective To understand the hepatotoxicity of cigarette smoke exposure to rats,effects of cigarette smoke exposure on liver toxicity in male SD rats by analyzing the changes of biochemical levels and oxidative stress indexes in liver tissues of rats exposed to cigarette smoke and observing the pathological changes in liver tissues of rats.Methods A total of 40 clean grade male SD rats were randomly divided into four dose groups,10 in each,the control,high,moderate and low exposure groups,and the static method of cigarette smoke exposure was used to treat the rats at the doses of 0,10,20 and 30 cigarettes per day respectively,for four consecutive weeks.The levels of ALT,AST,MDA,SOD in liver and TBIL in serum of rats in each group were determined.The pathological examination of liver was done.Results With the increase of cigarette smoke exposure doses,the weight and mass of liver of rats showed decreasing trend(P<0.05).The activities of AST and ALT in the liver tissues of rats in the exposure groups and the ratio of them were all increased significantly compared with those in the control group(P<0.05).Increased levels of serum TBIL were seen in the exposure groups compared with the control group,the difference was not significant(P>0.05).The level of MDA activity in liver tissues in the exposure groups was higher than that of the control group,with significant difference(P<0.05),and the level of SOD activity in the exposure groups was significantly lower than that of the control group(P<0.05).Significant pathological changes of liver in the exposure groups were observed.Conclusion Cigarette smoke exposure can inhibit weight gain and liver development in rats,and has an adverse effect on liver function and oxidative stress in rats.
