摘要
目的建立可同时测定抗衰老化妆品中4种儿茶素类成分表儿茶素(EC)、表儿茶素没食子酸酯(ECG)、表没食子儿茶素(EGC)、表没食子儿茶素没食子酸酯(EGCG)含量的高效液相色谱(HPLC)法。方法水剂、油剂化妆品用质量分数80%甲醇超声提取;膏霜乳液、凝胶化妆品采用稀释剂提取,并经澄清剂净化。经C_(18)色谱柱(4.6 mm×250 mm,5.0μm)分离,流动相为0.05%三氟乙酸水溶液-甲醇,梯度洗脱,柱温为35℃,采用二级阵列管检测器(DAD)测定4种儿茶素类成分。结果4种化合物在0.47~123.75mg/L线性范围内,所得回归方程的线性关系良好,r>0.999。该方法的检出限为2~30μg/g,对水剂、油剂、膏霜乳液、凝胶的平均回收率分别为85.5%~106.6%,82.1%~108.0%,86.7%~102.1%,85.2%~108.9%,RSD分别为0.13%~3.87%,0.15%~3.74%,0.30%~6.10%,0.19%~4.43%。结论该方法高效、灵敏、准确,适用于抗衰老化妆品中表儿茶素、表儿茶素没食子酸酯、表没食子儿茶素、表没食子儿茶素没食子酸酯4种成分的检测。
Objective To establish a method for the simultaneous determination of epicatechin(EC),(-)-epicatechin gallate(ECG),(-)-epigallocatechin(EGC)and epigallocatechin gallate(EGCG)in anti-aging cosmetics by high performance liquid chromatography(HPLC).Methods Toner and oil cosmetic samples were extracted with 80%(V/V)methanol while cream samples and gel samples were extracted by thinner and precipitated by precipitator.Then all the samples were separated on a C_(18)column(4.6 mm×250 mm,5.0μm).Under the optimum conditions of HPLC instrument,with gradient elution of 0.05%TFA and methanol and column temperature of 35℃,the four substances in samples were determined by photo-diode-arraydetector(DAD).Results In the linear range of 0.47-123.75 mg/L,the regression equation of four substances showed good linear correlation(r>0.999).The limits of detection were 2-30μg/g.The recovery rates were 85.5%-106.6%for toner cosmetic,82.1%-108.0%for oil cosmetic,86.7%-102.1%for cream cosmetic and 85.2%-108.9%for gel cosmetic respectively,and the RSDs were 0.13%-3.87%(toner),0.15%-3.74%(oil),0.30%-6.10%(cream)and 0.19%-4.43%(gel)respectively.Conclusion This method is efficient,sensitive and accurate,and is applicable to the simultaneous determination of epicatechin,(-)-epicatechin gallate,(-)-epigallocatechin and epigallocatechin gallate in anti-aging cosmetics.
