全血中24种元素的直接稀释-电感耦合等离子体质谱测定法

摘要 目的建立全血中24种元素的直接稀释-电感耦合等离子体质谱同时测定方法。方法以0.1%Triton X-100+0.5%硝酸为稀释剂,将全血样品稀释20倍,Sc、Y、ln、Ge、Bi溶液为在线内标,采取碰撞反应池(KED)模式,配合大流量雾化器,用电感耦合等离子体质谱仪对24种元素同时进行测定。结果Li、Be、V、Cr、Mn、Co、Ni、As、Se、Sr、Mo、Cd、Sn、Sb、Te、Tl(0.1~10.0μg/L),Mg、Ca(0.05~5.0 mg/L),Al、Cu(5.0~60.0μg/L),Fe(0.01~2.0 mg/L),Zn(10.0~500μg/L),Ba、Pb(0.5~25.0μg/L)在线性范围内,所得的回归方程均呈较好的线性关系,r≥0.9990。方法的检测限为0.008μg/L~0.007 mg/L,定量下限为0.025μg/L~0.023 mg/L。加标回收率为81.9%~110.0%;批内精密度为0.8%~7.9%,批间精密度为1.3%~9.2%。结论该方法灵敏度高、前处理简单、操作方便、线性范围宽,精密度和准确度满足生物样品测定要求,可用于人群大批量样品的生物监测和中毒样品的快速筛查。 Objective To establish the determination of 24 elements in whole blood by direct dilution-inductively coupled plasma mass spectrometry.Methods Diluting the whole blood sample 20 times by 0.1%Triton X-100+0.5%nitric acid as diluent,inductively coupled plasma mass spectrometer with a high-flow nebulizer in collision reaction cell(KED)mode were used to measure 24 elements using Sc,Y,ln,Ge,Bi solutions as online internal standards.Results The regression equations showed a good linear relationship with r≥0.9990,for Li,Be,V,Cr,Mn,Co,Ni,As,Se,Sr,Mo,Cd,Sn,Sb,Te,Tl in the range of 0.1-10.0μg/L,Mg,Ca in the range of 0.05-5.0 mg/L,Al,Cu in the range of 5.0-60.0μg/L,Fe in the range of 0.01-2.0 mg/L,Zn in the range of 10.0-500μg/L,Ba,Pb in the range of 0.5-25.0μg/L.The detection limits of method were 0.008μg/L-0.007 mg/L.The quantification limits of method were 0.025μg/L-0.023 mg/L。The recovery rates were 81.9%-110.0%;The intra-batch precisions were 0.8%-7.9%,and the inter-batch precisions were 1.3%-9.2%.The measurement results of quality control sample are all within the uncertainty range.Conclusion The method has the advantages of high sensitivity,simple preprocessing,easy operation,and wide linear range.The precision and accuracy meet the requirements for determination of biological sample,and can be used for biological monitoring of large batches of samples in the population and rapid screening of poisoned samples.
