摘要
目的探讨长链非编码RNA(long non-coding RNA,lncRNA)同源盒基因反义转录RNA(homeobox transcript antisense RNA,HOTAIR)在骨肉瘤组织中的表达及其与侵袭和转移的相关性。方法应用实时荧光逆转录定量聚合酶链反应(real-time reverse transcription quantitative polymerase chain reaction,RT-qPCR)技术检测40例骨肉瘤患者肿瘤组织及相对应癌旁组织中HOTAIR mRNA表达水平,并分析其与骨肉瘤患者临床病理特征的相关性。将U-20S细胞分为对照组、siR-NC组和siR-HOTAIR组,用Lipofectamine法分别将siR-NC和siR-HOTAIR转染到siR-NC组和siR-HOTAIR组骨肉瘤U-20S细胞。48 h后检测细胞增殖率、侵袭能力和迁移能力的变化,同时检测U-20S细胞中细胞周期蛋白E(cyclin E)、周期蛋白依赖性激酶2(cyclin-dependent kinases 2,CDK2)、金属基质蛋白酶2(matrix metalloproteinase 2,MMP-2)、E-钙粘附素(E-cadherin)和神经钙附粘蛋白(N-cadherin)水平。结果HOTAIR mRNA在肿瘤组织中的表达量显著高于癌旁组织,差异具有统计学意义(P<0.05);以均值为标准,15例(37.50%)患者肿瘤组织HOTAIR呈现低表达,25例(62.50%)患者肿瘤组织HOTAIR呈现高表达;HOTAIR表达水平与年龄、性别、肿瘤直径无相关性(P>0.05);HOTAIR表达水平与淋巴结转移和远处转移相关(P<0.05)。对照组和siR-NC组细胞增殖率、侵袭细胞数、迁移细胞数、cyclin E、CDK2、MMP-2、N-cadherin、E-cadherin水平比较差异无统计学意义(P>0.05);转染siR-HOTAIR后,与对照组和siR-NC组比较,siR-HOTAIR组U-20S细胞增殖率、侵袭细胞数、迁移细胞数、cyclin E、CDK2、MMP-2和N-cadherin明显降低,E-cadherin明显增高,差异具有统计学意义(P<0.05)。结论HOTAIR在骨肉瘤组织中高表达,并与肿瘤的侵袭相关,抑制HOTAIR的表达可抑制骨肉瘤U-20S细胞的侵袭和转移。
Objective To investigate the expression of long non-coding RNA(lncRNA) homeobox transcript antisense RNA(HOTAIR) in osteosarcoma tissue and its correlation with invasion and metastasis.Methods Real-time reverse transcription quantitative polymerase chain reaction (RT-qPCR) technology was used to detect the expression level of HOTAIR mRNA in tumor tissues and corresponding adjacent tissues of 40 osteosarcoma cases,and its correlation with the clinicopathological characteristics of osteosarcoma patients were analyzed.U-20 S cells were divided into control group,siR-NC group and siR-HOTAIR group.siR-NC and siR-HOTAIR were transfected into osteosarcoma U-20 S cells in siR-NC group and siR-HOTAIR group by Lipofectamine method respectively.After 48 hours,the cell proliferation rate,invasion ability and migration ability were detected.At the same time,the levels of cyclin E,cyclin-dependent kinases 2(CDK2)and matrix metalloproteinase 2(MMP-2),E-cadherin and N-cadherin in U-20 S cells were detected.Results The expression of HOTAIR mRNA in tumor tissues was significantly higher than that in adjacent tissues(P<0.05).According to the mean value,there were 15 patients(37.50%)with low HOTAIR expression in tumor tissue,and 25 cases(62.50%)with high HOTAIR expression in tumor tissue.The expression level of HOTAIR was not correlated with age,gender and tumor diameter(P>0.05).The expression level of HOTAIR was correlated with lymph node metastasis and distant metastasis(P<0.05).The cell proliferation rate,number of invasive cells,number of migrating cells,cyclin E,CDK2,MMP-2,N-cadherin,E-cadherin levels were not significantly different between the control group and the siR-NC group(P>0.05).After transfection with siR-HOTAIR,compared with the control group and the siR-NC group,the proliferation rate,the number of invasive cells,the number of migrating cells,cyclin E,CDK2,MMP-2 and N-cadherin expression in U-20 S cell were decreased,and E-cadherin increased(P<0.05).Conclusion HOTAIR is highly expressed in osteosarcoma tissues,and i
