摘要
目的基于网络药理学技术探讨柴胡加龙骨牡蛎汤治疗抑郁症的多靶标作用机制。方法采用网络预测和分子对接技术,从药物组成成分入手,借助中药系统药理学分析平台(TCMSP)检索柴胡和龙骨牡蛎汤所含化学成分的相关信息。通过生物利用度和类药性筛选潜在药效成分。同时从疾病入手,登录TTD、OMIM疾病数据库,检索与"depression"相关的已知靶基因或靶蛋白。通过Perl语言快速准确获取两部分数据的交集,并通过UniProt数据库确定与抑郁症相关靶蛋白的调控基因。通过可视化工具绘制潜在药效成分-靶基因、靶蛋白-疾病的网络示意图。最后通过R语言和DAVID数据库进行功能富集分析、聚焦核心通路、阐述柴胡加龙骨牡蛎汤治疗抑郁症的生物学过程。结果通过上述过程绘制作用网络,24个成分包括Stigmasterol、kaempferol、acacetin等,4个靶标包括MAOA、SLC6A2、ADRB2、DRD2。GO功能富集分析得到条目2个(P<0.05),生物过程条目regulation of sodium ion transport、分子功能条目integral component of plasma membrane。结论柴胡加龙骨牡蛎汤主要通过MAOA、SLC6A2、ADRB2、DRD2靶标的调节达到治疗抑郁症作用的目的。
Objective To explore the multi-target mechanism of Chaihu plus Longgu Muli Decoction in the treatment of depression based on network pharmacology technology.Methods Network prediction and molecular docking technology were used to retrieve the information of chemical components in Chaihu plus Longgu Muli Decoction by TCMSP.Potential pharmacodynamic components were screened by bioavailability and pharmacokinetics.Whilestarting from the disease,we searched TTD and OMIM disease databases to get known target genes or proteins related to depression.Through Perl language,we can quickly and accurately obtain the intersection of the two parts of data,and identify the regulatory genes of depression-related target proteins through UniProt database.Visualization tools were used to map the network of potential pharmacodynamic components-target genes and target proteins-diseases.Finally,the biological process of Chaihu plus Longgu Muli Decoction in the treatment of depression was elaborated by functional enrichment analysis and focusing on the core pathway through R language and DAVID database.Results The action network was drawn through the above process,including 24 components including Stigmasterol,kaempferol,acacetin and so on.Four targets included MAOA,SLC6 A2,ADRB2 and DRD2.GO functional enrichment analysis yielded two entries(P<0.05),including regulation of sodium transport for biological process entries and integration component of plasma membrane for molecular function entries.Conclusion Chaihu plus Longgu Muli Decoction can treat depression mainly by regulating the targets of MAOA,SLC6 A2,ADRB2 and DRD2.
