基于层次分析法和正交设计法优选盆消灌肠液的提取工艺 被引量:2

摘要 目的:确定指标权重,优选盆消灌肠液的最佳渗漉提取工艺。方法:建立高效液相色谱法(HPLC),同时测定处方药味中绿原酸、咖啡酸、芍药苷、阿魏酸、丹酚酸B 5个指标性成分的含量,以各指标性成分的含量和干膏率计算综合评分,采用层次分析法(AHP)确定各指标权重系数,结合正交试验结果优化盆消灌肠液提取工艺。结果:建立了HPLC同时测定绿原酸、咖啡酸、芍药苷、阿魏酸、丹酚酸B 5种成分含量的方法;5种成分分别在0.1516~1.5160μg、0.0516~0.5160μg、0.9624~9.6240μg、0.0160~0.1600μg、0.5012~5.0120μg内呈良好线性范围(R^(2)为0.9993~1.000),加样回收率为96.86%~98.67%,RSD均<3.0%;绿原酸、咖啡酸、芍药苷、阿魏酸、丹酚酸B、干膏率的权重系数分别为0.2564、0.2564、0.0943、0.0943、0.2564、0.0422;按照权重系数综合评价确定的盆消灌肠液最佳渗漉提取工艺条件为:处方量药材粉碎成粗粉,加8倍量70%乙醇,浸渍48h,以3mL/min的速度渗漉,收集渗漉液。结论:AHP法可用于盆消灌肠液渗漉提取工艺多指标权重的建立,优选的渗漉提取工艺经验证,合理、稳定、重复性好,研究可为盆消灌肠液工业化生产工艺提供参考依据。 Objective:To determine the weight of indexes and optimize the extraction process of the compound of Penxiao enema solution.Methods:Chlorogenic acid,Caffeic acid,Paeoniflorin,Ferulic acid and Salvianolic acid B in the prescription herbs were simultaneously determined by HPLC.The content of each index component and extraction rate were taken as the evaluation index,and the weight coefficient of each index was determined by AHP.and ethanol-extracting process was optimized by orthogonal test.Results:The HPLC method was established for the simultaneous determination of Chlorogenic acid,Caffeic acid,Paeoniflorin,Ferulic acid and Salvianolic acid B.The excellent linearity was obtained with correlation coefficients of 0.1516~1.5160、0.0516~0.5160、0.9624~9.6240、0.0160~0.1600、0.5012~5.0120μg,R^(2)of 0.9993~1.000,and the average recoveries of the five compouds were 96.86%~98.67%and RSD was less than 3.0%.The weighting coefficients of Chlorogenic acid,Caffeic acid,Paeoniflorin,Ferulic acid and Salvianolic acid B and extraction rate were 0.2564,0.2564,0.0943,0.0943,0.2564,0.0422,respectively.According to the comprehensive evaluation of the weight coefficient,the optimum extraction conditions were as follows,The medicinal materials were crushed into coarse powder,added with 8 times of 70%ethanol,soaked for 48 hours,percolated at the speed of 3 mL/min,and the percolate was collected.Conclusion:The weight coefficient determined by AHP was objective and feasible.The optimized extraction process is reasonable,stable and reproducible.
