连作对北沙参土壤养分、酶活性及微生物群落多样性的影响 被引量:8

Effects of Continuous Cropping on Soil Nutrients, Enzyme Activities and Microbial Community Diversity of Glehnia littoralis

摘要 目的:检测未连作(初茬)与连作(重茬)北沙参土壤中养分、pH值、酶活性和微生物群落变化,并对上述因素的相关性进行分析,探究北沙参发生连作障碍的原因。方法:以对照、初茬、重茬北沙参土壤为材料,测定土壤中的养分、pH值及酶活力,结合高通量测序技术,分析土壤中微生物群落的变化。结果:重茬土壤中全氮、有机质含量不断升高,全磷和全钾变化较小,pH值呈上升趋势。重茬土壤中微生物群落发生明显变化,变形菌门数量增加,酸杆菌门和放线菌门数量则呈减少趋势;而真菌中无法区分的群落明显增多。重茬土壤中土壤脲酶、硝酸还原酶、多酚氧化酶、蔗糖酶与酸性磷酸酶的活性较初茬增高明显。RDA分析表明土壤的养分、pH值及酶活性对土壤中真菌和细菌群落的变化有显著影响,相关性分析表明土壤全氮与有机质之间,全磷与全钾之间,全氮、有机质与部分土壤酶之间有着显著的相关性。结论:重茬导致北沙参土壤中的养分、pH值和酶活力发生了变化,而上述变化又导致土壤中微生物群落发生改变,这些因素之间的相互作用和相互影响,可能是导致北沙参连作障碍的主要原因。 Objective:To detect the changes of soil nutrients,pH,enzyme activities and microbial community in uncontinuous cropping(initial crop)and continuous cropping(repeated crop)of Glehnia littoralis,and to analyze the correlation of the above factors and explore the causes of continuous cropping disorder of Glehnia littoralis.Methods:Soil nutrients,pH and enzyme activity were measured in the control,initial and repeated cropping soil of Glehnia littoralis,and the changes of microbial community in the soil were analyzed by high-throughput sequencing technology.Results:In the repeated cropping soil,the contents of total nitrogen and organic matter increased continuously,while the changes of total phosphorus and total potassium were small,and the pH value showed an upward trend,the microbial community changed significantly,the number of proteobacteria increased,while that of acidobacteria and actinobacteria decreased,the number of undifferentiated fungal communities in fungi increased significantly.The activities of soil urease,nitrate reductase,polyphenol oxidase,sucrase and acid phosphatase in the repeated cropping soil were significantly higher than those in the primary cropping soil.RDA analysis showed that soil nutrients,pH and enzyme activities had significant effects on the changes of soil fungi and bacterial communities.Correlation analysis showed that there were significant correlations between total nitrogen and organic matter,total phosphorus and total potassium,total nitrogen,organic matter and some soil enzymes.Conclusion:The changes of nutrient,pH and enzyme activity in the soil cause by repeated cropping,these changes in turn lead to changes in soil microbial community.The interaction and influence among these factors may be the main cause of continuous cropping obstacle of Glehnia littoralis.
