摘要
目的评估阿仑膦酸钠(alendronate,ALN)对骨质疏松症的治疗作用。方法将Wis-tar雌性大鼠50只随机分成5组:正常组、模型组、ALN组、己烯雌酚组和对照组。除正常组做假性手术外,其余各组均切除双侧卵巢。治疗3个月后,作组织形态计量、骨密度测定和扫描电镜观察。结果形态学分析显示,ALN组的平均骨小梁宽度、骨形成表面和骨矿沉积率均比己烯雌酚组显著增加(P<0.001);两组的骨密度变化差异无显著性(P>0.05)。扫描电镜显示,ALN组及己烯雌酚组治疗后骨小梁宽度增加,断点减少,Howship陷窝变浅。结论ALN是一种值得选用的治疗骨质疏松的药物,它不但抑制骨吸收,且促进骨形成。
Objective To evaluate the effect of alendronate in osteoporosis. Methods Fifty female Wistar rats were allocated randomly into five groups: one group of normal, one group of model, two groups treated respectively with alendronate (ALN) or stilbestrol and a control. Every group, except the group of normal received a shamoperation, were ovarietomized bilaterally. The parameters to be assayed were histomorphometry, bone density and scanning electron microscopy (SEM) after treated for three months. Results Morphologic analysis revealed that ALN increased mean trabecular thickness, formation surface and mineral deposition rate in comparison with stilbestrol (P<0001), but no significant differences in bone mineral density were seen between both of the treatment groups (P>005).SEM showed that the trabecular bone widened, its broken points lessened and its Howship's lacunae shallowed in the groups after treatment. Conclusions These data indicate that ALN may provide a valuable alternative to current therapy for the treatment of osteoporosis, it not only depresses bone resorption, but also promotes bone formation.
