卵巢切除大鼠血清甲状旁腺素、降钙素变化与骨质量降低 被引量:8

Changes of parathyroid hormone and calcitonin contents in serum and deterioration of bone quality in ovariectomized rats

摘要 本文以OVX大鼠为动物模型,观察了去势15天、30天和60天大鼠血清甲状旁腺激素(PTH)和降钙素(CT)含量,第6胸椎(T6),第3腰椎(L3)和股骨矿盐含量及骨力学参数的变化。实验结果表明,去势15天、30天和60天大鼠血清CT显著降低,去势30天和60天大鼠血清PTH显著降低。去势15天大鼠T6横截面积显著降低,去势60天大鼠T6类骨质含量显著增加。去势15天和30天大鼠L3矿盐含量显著降低,30天骨压缩强度显著降低,60天最大应力、杨氏模量显著降低,横截面积显著增加。去势15天和30天大鼠股骨弯曲应力、张应力显著降低,弹性能密度在30天显著降低,去势60天股骨近1/3部位矿盐含量显著降低。实验结果提示,OVX大鼠CT降低是引起骨质丢失、骨力学性能低下的一个重要因素。松质骨骨丢失早于皮质骨,但力学性能的降低晚于皮质骨,OVX大鼠骨有机质减少是非胶原蛋白减少。p<0.01从表1可知,与对照组比较,去势15天大鼠T6骨横截面积显著降低,去势30天大鼠股骨干横截面积显著增加,去势60天大鼠L3截面积显著增加。去势30天和60天大鼠血清PTH显著降低。去势15天、30天和60天大鼠血清CT均显著降低。2.2两? The aim of the study was to determine the effect of ovariectomy on serum contents of parathyroid hormone (PTH) and calcitonin (CT), and aslo hone mass and biomechanical competence. A total of 42 Sprague-Dawley rats, 2 months old, were randomized to six groups. Of them three groups were ovarietomized (OVX) and followed for 15, 30 and 60 days after operation. The other three groups were sham-operated and followed for the same periods. At death, the sixth thoracic vertebra (T6), the third lumbar vertebra (L3) and left femur were obtained from each rat. The results revealed that, compared with controls, serum CT contents in OVX rats were significantly decreased on days 15, 30 and 60,and serum PTH content was sinificantly decreased on days 30 and 60. The cross-sectional area of T6 was significantly decreased on day 15 and osteoid content was significantly increased on day 60. The ash content of L3 had a significant decrease on days 15 and 30, compressive strength of L3 had a significant decrease on day 30. The maximum stress, Young's moduluswere significantly decreased and cross-sectional area of L3 was significantly increased on day 60 Bending stress and tension stress of femur significantly decreased on days 15 and 30, elastic energy density had a significant decrease on day 30. The ash content ware significantly decreased only in proximal 1/3 of the femur shaft on day 60. The study has showed that CT decrease in OVX rats may be a factor in reduction of bone quality. Bone mineral loss of trabecular bone is earlier than that of cortical bone, but the decrease in biomechanical competence of trabecular bone is later than that of cortical bone. The decrease of bone matrix in OVX rats is the decrease in noncollagenous proteins.
