重组人生长激素联合氟尿嘧啶对人胃癌裸小鼠皮下移植瘤作用的实验研究 被引量:13

摘要 目的:探讨不同剂量重组人生长激素(rhGH)对人胃癌裸小鼠皮下移植瘤的作用,及联合化疗(5-FU)的治疗效果。方法:建立人胃癌MKN-45裸小鼠皮下移植瘤模型,将荷瘤裸小鼠随机分为对照组,常规剂量GH组,小剂量GH组,5-FU组,常规剂量GH/5-FU组,小剂量GH/5-FU组,治疗10天,观察治疗效果。结果:治疗结束时两GH组的裸小鼠体质量明显大于对照组(P<0·05),两GH/5-FU组的裸小鼠体质量明显大于5-FU组(P<0·05),两GH/5-FU组与对照组比较没有统计学差异(P>0·05),两GH/5-FU组之间、两GH组之间比较无统计学差异(P>0·05)。与对照组相比,两GH组皮下移植瘤的体积和质量没有显著增大(P>0·05),5-FU组和两GH/5-FU组明显减少(P<0·05);5-FU组与两GH/5-FU组比较无显著性差异(P>0·05)。与对照组相比,两GH组的G0/G1期、S期、G2/M期肿瘤细胞百分比及增殖指数PI无显著性差异(P>0·05);与5-FU组相比,两GH/5-FU组的G0/G1期、S期、G2/M期肿瘤细胞百分比及增殖指数PI无显著性差异(P>0·05)。不同的GH剂量对实验结果无明显影响。化疗药物5-FU引起明显的白细胞减少(P<0·05),以及腹泻等胃肠道反应,对脏器及脏器功能无明显影响。结论:rhGH应用于荷瘤动物,显著增加机体质量,未明显促进肿瘤生长,对机体也无不良影响。联合化疗时,rhGH不影响化疗的进行,并可改善化疗时的机体状况。 Objective:To invest the effect of different doses of recombinant human growth hormone (rhGH)on stomach neo- plasms implanted in nude mice, and its efficacy in combining with chemotherapy(flurouracil,5-FU). Methods:Human stomach neo- plasm MKN-45 model was established in nude mice. The nude mice were divided into six groups:control group, moderate dose of rhGH group, low dose of rhGH group,5-FU group, moderate dose of rhGH/5-FU group, low dose of rhGH/5-FU group. The treatment results of each group were observed for ten days. Results : After therapy, the body mass of two rhGH groups was significantly increased compared with control group( P 〈 0. 05 ), the body mass of two rhGH/5-FU groups was significantly increased compared with 5-FU group (P 〈0. 05),but it was no significant difference between rhGH/5-FU groups and control group( P 〉0.05 ). The average tumor mass and volume of rhGH groups were not significantly increased compared with control group( P 〉0. 05 ), but obviously reduced in 5-FU group and rhGh/5-FU groups( P 〈0. 05 ). There was no significant difference between any rhGh/5-FU group and 5-FU group( P 〉0. 05 ). After treatment, the percentages of S, G0/G1 , G2/M phasees and proliferation indices ( PI )were not significantly changed in rhGH groups compared with control group (P 〉0. 05 ), and as well as rhGH/5-FU groups compared with 5-FU group (P 〉 0. 05 ). The difference caused by dose of rhGH was not significant. Conclusion : rhGH enhances body mass, but does not stimulate tumor growth, and has no adverse effects on tumor bearing nude mice. Combined with Flurouracil, rhGH does not influence the efficacy of chemotherapy. Its effect on cell cycle and chemotherapy may depend on expression of growth hormone receptor in tumor cells.
