摘要
研究了影响玉米花药培养的3个因素,结果表明:高温高压灭菌和过滤灭菌对于胚状体产生和绿色植株分化没有实质性差异,而将蔗糖分开灭菌,则可显著提高胚状体产生的数目.在8种不同碳水化合物的比较中,以庶糖和棉子糖培养效果最好,显著高于其它糖类,两者可以相互代替诱导培养期间蔗糖浓度变化,显著地改变着胚状体的产生和绿色植株的分化,以0~14大,180g/L,第15~21天,90g/L处理培养效果最好,高出传统方法3倍多。
Three factors which affect the anther culture of maize(Zea nlavs L)werestudied.The results showed:there was no diffrence between autoclavedsterilizationand filtersterilization of medium on maize anther culture response,but it could increasethe number of embrvo-like-structures and green plants if sucrose was autoclavedseparatly.By comparison with 8 kinds of carbohydrates,the best resuIts came fromsucrose or raffinose,which were much higher than the others;Raffinose can replacesucrose if it is necessary.The changes ofconcentration of sucrose during the induction pe-riod varied the number of embryo-like-structures andgreen plants significandy,thetreatment:from lst day to l4th day,sucrose:180 g/L;15th day to 2lst day,sucrose:90g/L produced the best result,which was over three times more efficientthaii the traditionaI treatment(from lst day to 2lst day,sucrose:90g/L),thismethod can increase the efficiencv of maize anther cuIture substantiallv。
