The improved peri-implant bone response demonstrated by platform switching may be the result of reduced amounts of metal ions released to the surrounding tissues. The aim of this study was to compare the levels of metal ions released from platform-matched and platform-switched implant-abutment couples as a result of accelerated corrosion. Thirty-six titanium alloy (Ti-6AI-4V) and cobalt-chrome alloy abutments were coupled with titanium cylinders forming either platform-switched or platform-matched groups (n = 6). In addition, 18 unconnected samples served as controls. The specimens were subjected to accelerated corrosion by static immersion in 1% lactic acid for 1 week. The amount of metal ions ion of each test tube was measured using inductively coupled plasma mass spectrometry. Scanning electron microscope (SEM) images and energy dispersive spectroscopy X-ray analyses were performed pre- and post-immersion to assess corrosion at the interface. The platform-matched groups demonstrated higher ion release for vanadium, aluminium, cobalt, chrome, and molybdenum compared with the platform-switched groups (P〈 0.05). Titanium was the highest element to be released regardless of abutment size or connection (P〈0.05). SEM images showed pitting corrosion prominent on the outer borders of the implant and abutment platform surfaces. In conclusion, implant-abutment couples underwent an active corrosion process resulting in metal ions release into the surrounding environment. The highest amount of metal ions released was recorded for the platform-matched groups, suggesting that platform-switching concept has a positive effect in reducing the levels of metal ion release from the implant-abutment couples. 展开更多

This study evaluated the adhesion of zirconia core ceramics with their corresponding veneering ceramics, having different thermal expansion coefficients (TECs), when zirconia ceramics were coloured at green stage. Zirconia blocks (N=240; 6 mm x 7 mm x 7 mm) were manufactured from two materials namely, ICE Zirconia (Group 1) and Prettau Zirconia (Group 2). In their green stage, they were randomly divided into two groups. Half of the specimens were coloured with colouring liquid (shade A2), Three different veneering ceramics with different TEC (ICE Ceramic, GC Initial Zr and IPS e.max Ceram) were fired on both coloured and non-coloured zirconia cores. Specimens of high noble alloys (Esteticor Plus) veneered with ceramic (VM 13) (n= 16) acted as the control group. Core-veneer interface of the specimens were subjected to shear force in the Universal Testing Machine (0.5 mm-min-1). Neither the zirconia core material (P=0.318) nor colouring (P=0.188) significantly affected the results (three-way analysis of variance, Tukey's test). But the results were significantly affected by the veneering ceramic (P=0.000). Control group exhibited significantly higher mean bond strength values (45.7__.8) MPa than all other tested groups ((27.1__.4.1)-(39.7__.4.7) and (27.4__.5.6)-(35.9___4.7) MPa with and without colouring, respectively) (P^0.001). While in zirconia-veneer test groups, predominantly mixed type of failures were observed with the veneering ceramic covering ~ 1/3 of the substrate surface, in the metal-ceramic group, veneering ceramic was left adhered 1/3 of the metal surface. Colouring zirconia did not impair adhesion of veneering ceramic, but veneering ceramic had a significant influence on the core-veneer adhesion. Metal-ceramic adhesion was more reliable than all zirconia-veneer ceramics tested. 展开更多

In vitro manipulation of induced pluripotent stem cells(iPSCs) by environmental factors is of great interest for three-dimensional(3D) tissue/organ induction. The effects of mechanical force depend on many factors, including force and cell type. However, information on such effects in i PSCs is lacking. The aim of this study was to identify a molecular mechanism in i PSCs responding to intermittent compressive force(ICF) by analyzing the global gene expression profile. Embryoid bodies of mouse i PSCs, attached on a tissue culture plate in 3D form, were subjected to ICF in serum-free culture medium for 24 h. Gene ontology analyses for RNA sequencing data demonstrated that genes differentially regulated by ICF were mainly associated with metabolic processes, membrane and protein binding. Topology-based analysis demonstrated that ICF induced genes in cell cycle categories and downregulated genes associated with metabolic processes. The Kyoto Encyclopedia of Genes and Genomes database revealed differentially regulated genes related to the p53 signaling pathway and cell cycle. q PCR analysis demonstrated significant upregulation of Ccnd1, Cdk6 and Ccng1. Flow cytometry showed that ICF induced cell cycle and proliferation, while reducing the number of apoptotic cells. ICF also upregulated transforming growth factor β1(Tgfb1) at both m RNA and protein levels, and pretreatment with a TGF-β inhibitor(SB431542) prior to ICF abolished ICF-induced Ccnd1 and Cdk6 expression. Taken together, these findings show that TGF-β signaling in i PSCs enhances proliferation and decreases apoptosis in response to ICF, that could give rise to an efficient protocol to manipulate i PSCs for organoid fabrication. 展开更多

Introduction and Objective: The aim of this research was to find out the interaction of anthropometric measurements particularly body mass and oral health status of Turkish young adult nationals regionally, depending on the effects of diet (food and fluid consumption), smoking and tooth brushing habits, which were determined by using the body mass index (BMI) and dental health index (DMFT). Material and Methods: The BMI and DMFT of 202 cases, with an age average 22.5 years, were recorded through physical and clinical examinations. Statistical evaluation of the data was checked by Cronbach’s Alpha Reliability Scale and performed by analysis of variance, Somer’s d and Pearson Chi-Square statistics, Poisson Regression and Odds Ratio analysis. Results: The results showed that the mean of BMI was at the optimum level (21.52) for all cases. The DMFT was higher for underweight females and for overweight males. The mean of DMFT according to sexes seemingly had no significant difference;however, females had 8.6% more DMFT than males. When the risk factors of dental caries were evaluated, the DMFT was found as 19.3% more for cases consuming high carbohydrate foods, 4.5% more for high beverage consumption and 18% more for smokers. Conclusions: In conclusion, tooth brushing 3 or more times a day is necessary to decrease the DMFT and tooth brushing 3 times a day as opposed to 1 or 2 times shows significant difference regarding oral health. 展开更多

The aim of this study was to compare the bone resorption differences between implant placement sites (IPS) and non-implant placement sites (NIPS) after autogenous block bone grafts in the anterior maxilla. Fourteen patients (58 edentulous sites) with alveolar atrophy in the anterior maxilla were treated with autogenous block bone grafts. CBCT examinations were performed at 1 month before surgery (T0), immediately after surgery (T1), 3 to 4 months after surgery (T2), 6 to 7 months after surgery before implant placement (T3), 12 to 13 months after surgery (T4), and the longest follow-up point (T5). Alveolar crestal and basal bone width (ACBW, ABBW), and alveolar bone height (ABH) were measured and divided into IPS (30 sites) and NIPS (28 sites). All results were compared by the Wilcoxon Signed Rank test. The bone resorption changes for both groups were the same. For these three parameters, ACBW didn’t change significantly from T2 to T3 and T4 to T5, ABBW didn’t change at every period from T2 to T5, and ABH didn’t change from T4 to T5. The bone resorption volume of ACBW and ABH in NIPS were more than in IPS after implant placement surgery, while the volume of ABBW was similar in both groups. At T5, the bone resorption percentages of ACBW, ABBW, and ABH were 25.57%, 16.85% and 43.84% in IPS, and 33.55%, 15.92% and 46.44% in NIPS. A more rapid loss of alveolar crest in NIPS resulted from implant placement surgery, and this reminded us of the importance of immediate implant placement. 展开更多
