The effects of chemical activation on birth efficiency of cloned pigs were studied by investigating the developmental process from porcine oocyte activation to birth of cloned pigs.Three different activation methods w...The effects of chemical activation on birth efficiency of cloned pigs were studied by investigating the developmental process from porcine oocyte activation to birth of cloned pigs.Three different activation methods were used:(i) Electroporation(Ele);(ii) Ele followed by incubation with 6-dimethylaminopurine(6-DMAP);and(iii) Ele followed by a treatment with cycloheximide(CHX).In experiment 1,the rates of cleavage,developmental rates and cell number of porcine parthenogenetic(PA) embryos were investigated in the three treatment groups.In experiment 2,NT embryos produced by the three different activation treatments were compared for the rates of cleavage,development and cell number.Finally,the effects of Ele and Ele+CHX activation methods on birth efficiency of cloned pigs were compared.The activated oocytes treated by combination activation generally showed a higher(P<0.05) blastocyst rate and produced more expanded blastocysts than oocytes activated with Ele.The rates of cleavage and total cell number of parthenotes were not significantly different.Parthenogenetic embryos activated with 6-DMAP developed into blastocyst and expanded blastocyst stages at a significantly(P<0.05) higher rate than those treated with Ele,but the developmental capability was dramatically decreased in NT embryos.With the CHX activation method,the NT embryo blastocyst rate was substantially(P<0.05) increased although the production of expanded blastocysts was not significantly different from that by the other two methods.The birth rate of cloned pigs increased in the CHX group,though the rate was not significantly different from Ele.The effects of season on developmental rate of the porcine PA embryos and birth rate of cloned pigs were also examined in our study.Porcine oocytes collected in the spring had higher developmental capabilities than those collected in the winter.However,no difference in birth rate of the cloned pigs was found between the oocytes collected in the two seasons.The results obtained from PA and NT embryos,following 展开更多
In recent years, the demand for goat products has been growing due to the fact that goat milk has a number of advantages over cow milk, for example, it is low in lactose, and is considered less allergenic and easier t...In recent years, the demand for goat products has been growing due to the fact that goat milk has a number of advantages over cow milk, for example, it is low in lactose, and is considered less allergenic and easier to digest. To increase production during both breeding and non-breeding seasons and reduce the price of dairy products, it is necessary to effectively use reproductive management and assisted reproductive technologies. In vitro embryo production makes it possible to obtain a large number of eggs from goats, which for some reason are unable to conceive, but have genetic value. Afterward in vitro produced embryos can be transferred into recipient goats of other less genetically valuable breeds, such as the Ukrainian local breed. Therefore, the aim of the present study was to investigate the effectiveness of transfers of in vitro produced embryos of Saanen goats into surrogate sires of the Ukrainian local breed in different seasons. All manipulations with animals were carried out following ethical standards (Strasbourg, 1986). Six Saanen goats were selected as the oocyte donors. After the hormonal stimulation oocytes were retrieved by laparoscopic ovum pick-up. In vitro produced embryos were transferred laparotomically into 24 recipients of Ukrainian local breed. Fifty days after embryo transfers, pregnancies were determined by ultrasound diagnostics. Although the embryo development rate in the breeding season was 20% higher than in the non-breeding season, there was no difference in pregnancy and kidding rates between seasons. In conclusion, the transfer of in vitro produced Saanen goat embryos to recipients of the Ukrainian local breed gives the opportunity to achieve pregnancy and kidding regardless of the breeding season, which will enable a faster and more efficient increase in the livestock of highly productive goats in Ukraine in the post-war period.展开更多

Three climatic zones(mid-temperate zone,warm temperate zone,subtropical zone) were designed,and embryo transfers had been carried out in spring,summer,autumn and winter from 2009 to 2011,respectively. The total number of transplant recipient cattle was 22 208. The results showed that the best seasons for embryo transfers varied with different climatic zones. In mid-temperate zone,summer and autumn were better while summer was the best,and the rate of pregnant was 50. 67%(the number of pregnant cattle was 8 005). In warm temperate zone,spring and autumn were better while autumn was the best,and the rate of pregnant was 54. 99 %(the number of pregnant cattle was 551). In subtropical zone,spring and winter were better while winter was the best,and the rate of pregnant was 56. 94 %(the number of pregnant cattle was 328). The seasonal average temperatures and relative humidity of the best seasons in different climatic zones were concentrated on the same region. In mid-temperate zone,the mean temperature ranged between 22. 4 ℃ and 24. 2 ℃,and the mean relative humidity ranged from 44% to 55. 3%. In warm zone,the mean temperature ranges were between 14. 2 ℃ and 16. 2 ℃,and the mean relative humidity ranges were from 59. 3% to 71. 6%. In sub-tropical zone,the mean temperature ranges were between 3. 26 ℃ and 7. 73 ℃,and the mean relative humidity ranges were from 72% to 80. 6%. Therefore,the optimized conditions of temperature and humidity of season in different zones could be simulated. Therefore, to examine the thermal environment of infants, experiments and simulations using a human body thermal model may be feasible by clarifying the human body coefficient values of infants. When focusing on the heat transfer area of the infant’s body coefficient values, the body surface area and solar radiation area factor have only been clarified as the standard. The essential basic data for examining the thermal environment of infants have not been sufficiently obtained. Therefore, the purpose of this study was to determine the clothing area factor for infants’ clothing in a typical season. The clothing area factor of infants clothed in summer, mid-season, and winter clothing was measured using a photographic method. The clothing area factor was 1.22, 1.42, and 1.90 for summer, mid-season, and winter clothing, respectively, indicating a significant seasonal difference. It was also found that the clothing area factor was significantly greater for infants than for adults.展开更多
