OBJECTIVE:To investigate the effects of Hippeastrum hybridum(HH)as a free radical scavenger,and an inhibitor of the two enzymes i-e Alpha-amylase(α-amylase)and acetylcholinesterase(AChE).METHODS:In this study,HH plan...OBJECTIVE:To investigate the effects of Hippeastrum hybridum(HH)as a free radical scavenger,and an inhibitor of the two enzymes i-e Alpha-amylase(α-amylase)and acetylcholinesterase(AChE).METHODS:In this study,HH plant was preliminary analyzed for phytochemical screening and then tested for its antioxidant,anti-α-amylase,and anti-AChE efficiency via standard procedures.RESULTS:Phytochemical analysis shows the existence of different compounds;while Coumarins and quinones were absent.The total phenolic,flavonoid,and tannins content were found to be(78.52±0.69)mg GAE/g,(2.01±0.04)mg RUE/g,and(58.12±0.23)mg TAE/g of plant extract respectively.28.02%±0.02%alkaloid and 2.02%±0.05%saponins were present in the HH extract.The HH extract showed the anti-oxidant property with IC50(50%inhibition)of(151.01±0.13)(HH),(79.01±0.04)(Ascorbic acid)for ferric reducing,(91.48±0.13)(HH),(48.02±0.11)(Ascorbic acid)against Ammonium molybdenum,(156.02±0.31)(HH),(52.38±0.21)(Ascorbic acid)against DPPH,136.01±0.21(HH),52.02±0.31(Ascorbic acid)against H_(2)O_(2),and 154.12±0.03(HH),(40.05±0.15)(Ascorbic acid)μg/mL against ABTS respectively.Statistical analysis indicated that HH caused a competitive type of inhibition ofα-amylase(V_(max)remained constant and Km increases from 10.65 to84.37%)while Glucophage caused the un-competitive type of inhibition i-e both Km and V_(max)decreased from40.49 to 69.15%and 38.86 to 69.61%respectively.The K_(i),(inhibition constant);K_(I),(dissociation constant),K_(m),(Michaelis-Menten constant),and IC50 were found to be62,364,68.1,and 38.08±0.22 for HH and 12,101.05,195,34.01±0.21 for Glucophage.Similarly,HH causes an anon-competitive type of inhibition of AChE i-e Km remains constant while V_(max)decreases from 60.5%to74.1%.The calculated K_(i),K_(I),K_(m),and IC50 were found to be 32,36.2,0.05,and 18.117±0.018.CONCLUSION:From the current results,it is concluded that HH extract contains bioactive compounds,and could be a good alternative to controlling oxidants,Alzheimer‘s and Type-�展开更多
Background:Winter climate change including frequent freeze-thaw episodes and shallow snow cover will have major impacts on the spring regrowth of perennial crops.Non-bloating perennial forage legume species including ...Background:Winter climate change including frequent freeze-thaw episodes and shallow snow cover will have major impacts on the spring regrowth of perennial crops.Non-bloating perennial forage legume species including sainfoin,birdsfoot trefoil,red clover,and alsike clover have been bred for their adaptation to harsh winter conditions.In parallel,the selection of cold-tolerant rhizobial strains could allow earlier symbiotic nitrogen(N)fixation to hasten spring regrowth of legumes.Methods:To identify strains forming nodules rapidly and showing high N-fixing potential,60 rhizobial strains in association with four temperate legume species were evaluated over 11 weeks under spring soil temperatures for kinetics of nodule formation,nitrogenase activity,and host yield.Results:Strains differed in their capacity to form efficient nodules on legume hosts over time.Strains showing higher nitrogenase activity were arctic strain N10 with sainfoin and strain L2 with birdsfoot trefoil.For clovers,nitrogenase activity was similar for control and inoculated plants,likely due to formation of effective nodules in controls by endophyte rhizobia present in seeds.Conclusions:Selection based on nodulation kinetics at low temperature,nitrogenase activity,and yield was effective to identify performant rhizobial strains for legume crops.The use of cold-tolerant strains could help mitigate winter climatic changes.展开更多
In the present study,genetic diversity and genetic relationship of 11materials including red clover ( Trifolium pretense L.),white clover ( T.repen L.) and alsike clover ( T.hybridum L.) and leaf-type variants of whit...In the present study,genetic diversity and genetic relationship of 11materials including red clover ( Trifolium pretense L.),white clover ( T.repen L.) and alsike clover ( T.hybridum L.) and leaf-type variants of white clover were investigated by SRAP (sequence related amplified polymorphism).Forty SRAP primers were screened,and 792 reliable bands were amplified,among which 426 (53.8%) were polymorphic.The number of polymorphic bands per pair of primers ranged from 3 to 38 with an average of 10.6.SRAP cluster analysis showed that the similarity coefficients between white clover materials were from 0.465 to 0.997 with an average of 0.812,indicating that there was certain genetic diversity among white clover.Specific bands appeared in white clover variants,indicating that there were certain differences in DNA sequence between normal while clover plants and their leaf-type variants.The similarity coefficients between red clover materials were from 0.457 to 0.827 with an average of 0.597,indicating that the different red clover germplasms had genetic diversity at the DNA molecular level.The results also showed that the genetic relationship of the alsike clover with the red clover was closer than that with the white clover.This study shows that SRAP technique can be effectively used for the analysis on interspecific and intraspecific relationship,germplasm resource identification and genetic diversity of clover.展开更多
[Objective]The paper was to study the physiological mechanism of Oncidium hybridum‘Gower Ramsey’infected by Erwinia carotovora pv.carotovora.[Method]O.hybridum‘Gower Ramsey’was inoculated with E.carotovora by need...[Objective]The paper was to study the physiological mechanism of Oncidium hybridum‘Gower Ramsey’infected by Erwinia carotovora pv.carotovora.[Method]O.hybridum‘Gower Ramsey’was inoculated with E.carotovora by needle-punching method.The leaves inoculated with sterile water were used as control,and samples were collected at 12,24,36 and 48 h post inoculations to determine the physiological indexes,respectively.[Result]The contents of MDA,flavonoid,lignin,soluble protein and the activities of POD,SOD,CAT,PPO and PAL in O.hybridum leaves first increased then decreased.At 12 h post inoculation,the activities of SOD and CAT increased significantly by 40.49%and 37.77%(P<0.01,the same below),respectively;the contents of flavonoid increased significantly by 21.56%(P<0.05,the same below);and the activities of PAL,POD and PPO increased significantly by 42.29%,81.8%and 37.98%,respectively.The contents of MDA and lignin increased significantly by 57.98%and 58.72%at 24 h,respectively;the content of soluble protein increased by 87.71%at 24 h,and 61.28%at 36 h,while the content of Pro increased at 24 h(P>0.05,the same below),and increased significantly by 23.66%at 36 h;the content of soluble sugar content had no significant difference compared with the control.Additionally,the chlorophyll a and carotenoid contents tended to decrease dramatically,while chlorophyll b increased significantly by 95.33%at 24 h,then decreased.The initial fluorescence parameter of Fm/Fv and Y(II)decreased significantly by 47.14%and 54.61%,respectively,while the Y(NO)increased significantly by 33.61%,compared with the control.[Conclusion]The contents of Pro,MDA,lignin,soluble protein and the activities of POD,SOD,CAT,PPO and PAL in O.hybridum leaves are closely related to the infection of E.carotovora,which provide new clues for revealing the infection mechanism of E.carotovora in O.hybridum.展开更多
The treatment and disposal of municipal sewage sludge(MSS)is an urgent problem to be resolved in many countries.Safely using the nutrients within MSS to increase crop yield and enhance the fertility of poor soil could...The treatment and disposal of municipal sewage sludge(MSS)is an urgent problem to be resolved in many countries.Safely using the nutrients within MSS to increase crop yield and enhance the fertility of poor soil could contribute to achieving sustainable development.An indirect use of MSS in ditches alongside Pennisetum hybridum plants was studied in field plots for 30 months and the contents of heavy metals and macronutrients were monitored in soil,sludge and plant samples.We found that the yield of P.hybridum was significantly increased by 2.39 to 2.80 times and the treated plants had higher N content compared with no sludge.In addition,the organic matter(OM)and N contents in the planted soil increased significantly compared with the initial soil.The OM content in the planted soil of the MSS treatment was 2.9 to 5.2 times higher than that with no sludge,and N increased by 2.0 to 3.8 times.However,MSS had no significant effect on the N,P and K contents in the soil at the bottom of the MSS ditch,and the content of heavy metals(Cd,Pb,Cu and Zn)were also within the safe range.Moreover,the moisture content and phytotoxicity of MSS after this indirect use were reduced and the heavy metal contents changed little,which is favorable to the further disposal of recovered MSS.Therefore,this indirect use of MSS is beneficial to agricultural production,soil quality and environmental sustainability.展开更多
