目的:研究激活蛋白-1(AP-1)对机械通气性肺损伤小鼠炎症反应、细胞凋亡和基质金属蛋白酶-9(MMP-9)/组织基质金属蛋白酶抑制因子-1(TIMP-1)平衡的影响。方法:选择2~3个月龄健康昆明小鼠30只,雌雄不限,分为假手术组、模型组和干预组3组,每... 目的:研究激活蛋白-1(AP-1)对机械通气性肺损伤小鼠炎症反应、细胞凋亡和基质金属蛋白酶-9(MMP-9)/组织基质金属蛋白酶抑制因子-1(TIMP-1)平衡的影响。方法:选择2~3个月龄健康昆明小鼠30只,雌雄不限,分为假手术组、模型组和干预组3组,每组10只。假手术组行气管切开保留自主呼吸,模型组和干预组行机械通气6 h,干预组连接呼吸机后立即腹腔注射AP-1单克隆抗体(20μg)。ELISA法检测血浆白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)水平,取腹主动脉血检测血氧分压(PaO2),测定肺湿干重比值(W/D),HE染色观察肺组织病理学改变并进行肺损伤评分,TUNEL法检测肺组织中细胞凋亡指数,Western blot法检测肺组织中AP-1、MMP-9和TIMP-1蛋白的相对表达量。结果:与假手术组比较,模型组和干预组血浆IL-6和TNF-α水平升高,PaO2降低,肺W/D增加,肺组织病理学改变明显,肺损伤评分、细胞凋亡指数、AP-1和MMP-9蛋白相对表达量升高,TIMP-1蛋白相对表达量降低;与模型组比较,干预组血浆IL-6和TNF-α水平下降,PaO2升高,肺W/D降低,肺组织病理学改变减轻,肺损伤评分、细胞凋亡指数、AP-1和MMP-9蛋白相对表达量降低,TIMP-1蛋白相对表达量升高(P<0.05)。结论:外源性抑制AP-1表达可改善机械通气性肺损伤程度,减轻炎症反应和细胞凋亡,调节MMP-9/TIMP-1平衡。 展开更多

目的探讨TGF-β1、TIMP-1及CollagenⅢ在盆腔器官脱垂患者阴道壁中的表达及意义。方法选取2020年3月至2020年12月在昆明医科大学第一附属医院妇科住院部手术的盆腔器官脱垂(pelvic organ prolapse,POP)患者28例为实验组,其中按POP-Q分... 目的探讨TGF-β1、TIMP-1及CollagenⅢ在盆腔器官脱垂患者阴道壁中的表达及意义。方法选取2020年3月至2020年12月在昆明医科大学第一附属医院妇科住院部手术的盆腔器官脱垂(pelvic organ prolapse,POP)患者28例为实验组,其中按POP-Q分期法将其分为:Ⅱ度7例,Ⅲ度16例,IV度5例。选择同期因妇科良性病变行全子宫和(或)双附件切除且无POP患者30例为对照组。通过免疫组化的方法检测阴道壁组织中TGF-β1、TIMP-1及CollagenⅢ的表达。结果 POP组的TGF-β1、TIMP-1及CollagenⅢ的IRS得分明显低于对照组,差异有统计学意义(P <0.05);POP组Ⅱ度、Ⅲ度及Ⅳ度患者的TGF-β1、TIMP-1及CollagenⅢ的IRS得分随着分度的增加呈逐渐降低的趋势,差异均有统计学意义(P <0.05)。POP组患者阴道壁组织中CollagenⅢ与TIMP-1、TGF-β1表达呈正相关(r=0.381,0.519,P <0.05);TGF-β1表达与TIMP-1表达呈正相关(r=0.270,P <0.05)。结论 POP患者阴道壁组织中CollagenⅢ、TIMP-1和TGF-β1呈低表达,且与病情程度有一定的关系,可能参与POP的发生、发展;POP患者阴道壁组织中CollagenⅢ、TIMP-1、TGF-β1间存在一定相关性,三者之间存在一定的协同作用,共同参与疾病的发生发展。 展开更多

Imbalance of matrix metalloproteinase-9 and matrix metalloproteinase tissue inhibitor-1 may contribute to hemorrhage in cerebellar arteriovenous malformations

In this study, we determined the expression levels of matrix metalloproteinase-2 and -9 and matrix metalloproteinase tissue inhibitor-1 and -2 in brain tissues and blood plasma of patients undergoing surgery for cerebellar arteriovenous malformations or primary epilepsy (control group). Immunohistochemistry and enzyme-linked immunosorbent assay revealed that the expression of matrix metalloproteinase-9 and matrix metalloproteinase tissue inhibitor-1 was significantly higher in patients with cerebellar arteriovenous malformations than in patients with primary epilepsy. The ratio of matrix metalloproteinase-9 to matrix metalloproteinase tissue inhibitor-1 was significantly higher in patients with hemorrhagic cerebellar arteriovenous malformations compared with those with non-hemorrhagic malformations. Matrix metalloproteinase-2 and matrix metalloproteinase tissue inhibitor-2 levels were not significantly changed. These findings indicate that an imbalance of matrix metalloproteinase-9 and matrix metalloproteinase tissue inhibitor-I, resulting in a relative overabundance of matrix metalloproteinase-9, might be the underlying mechanism of hemorrhage of cerebellar arteriovenous malformations. 展开更多

目的通过构建尿酸性肾病动物模型,检测给药后KIM-1、NGAL、TIMP-1、MCP-1等肾损伤标志物,探讨苗药痛风停汤对尿酸性肾病的保护作用。方法将70只SD大鼠随机分为空白组、模型组、苗药痛风停高、中、低剂量组、别嘌醇组和苯溴马隆组,构建... 目的通过构建尿酸性肾病动物模型,检测给药后KIM-1、NGAL、TIMP-1、MCP-1等肾损伤标志物,探讨苗药痛风停汤对尿酸性肾病的保护作用。方法将70只SD大鼠随机分为空白组、模型组、苗药痛风停高、中、低剂量组、别嘌醇组和苯溴马隆组,构建尿酸性肾病动物模型,造模后各组灌胃给药治疗28天,末次给药后收集大鼠血清及肾脏标本。生化检测大鼠血清尿酸、尿素氮、肌酐含量,用免疫组化法、Western Blot检测肾组织的KIM-1、NGAL、TIMP-1的表达、RT-PCR法检测肾脏组织KIM-1、MCP-1mRNA的表达。结果生化结果:别嘌醇、高剂量、中剂量、苯溴马隆组组间比较,其BUN、SCR、UA含量差异无统计学差异(P>0.05)。免疫组化结果:(1)痛风停高、中、低剂量、别嘌醇组KIM-1平均光密度比苯溴马隆组明显升高,差异具有统计学意义(P<0.01);(2)痛风停高、中剂量、别嘌醇组NGAL、TIMP-1平均光密度与苯溴马隆组相比,无明显差异(P>0.05)。RT-PCR结果与别嘌醇及苯溴马隆组相比,痛风停高剂量组KIM-1、MCP-1mRNA表达量与其无明显差异(P>0.05)。Western Blot结果(1)与别嘌醇组相比,痛风停高剂量组KIM-1、NGAL、TIMP-1蛋白表达均与其无明显差异(P>0.05);(2)与苯溴马隆组对比,痛风停高剂量组NGAL蛋白表达与其无明显差异(P>0.05);其余各组KIM-1、NGAL、TIMP-1蛋白表达量升高,差异具有统计学意义(P<0.05)。结论苗药痛风停汤可显著降低尿酸性肾病大鼠血肌酐、尿素氮及尿酸水平,并可通过降低肾损伤因子KIM-1、NGAL、TIMP-1、MCP-1的表达,从而实现保护肾脏功能作用。 展开更多

长期抗病毒治疗的慢性乙型肝炎患者血清TIMP-1和M-CSF水平变化 被引量:1

目的 探讨长期抗病毒治疗的慢性乙型肝炎(CHB)患者血清组织基质金属蛋白酶抑制物-1(TIMP-1)和巨噬细胞集落刺激因子(M-CSF)水平变化。方法 纳入2017年4月—2019年2月合肥市第三人民医院收治的CHB患者148例为研究对象,连续抗病毒治疗2年... 目的 探讨长期抗病毒治疗的慢性乙型肝炎(CHB)患者血清组织基质金属蛋白酶抑制物-1(TIMP-1)和巨噬细胞集落刺激因子(M-CSF)水平变化。方法 纳入2017年4月—2019年2月合肥市第三人民医院收治的CHB患者148例为研究对象,连续抗病毒治疗2年。观察治疗前后血清TIMP-1和M-CSF水平变化。统计CHB患者肝硬化发生情况,并依据是否发生肝硬化分为发生组和未发生组。对比发生组和未发生组患者的临床资料。logistic多因素回归分析影响CHB患者肝硬化发生的因素。制作受试者工作特征曲线(ROC),以曲线下面积(AUC)评价血清TIMP-1、M-CSF变化对CHB患者肝硬化发生的预测效能。结果 与治疗前1 d相比,治疗6个月后CHB患者血清TIMP-1和M-CSF均降低(P<0.05),ΔTIMP-1、ΔM-CSF分别为(56.6±8.3)μg/L、(62.5±9.2)ng/L。治疗2年后肝硬化发生率为24.32%。发生组丙氨酸氨基转移酶(ALT)、肝门静脉内径、肝脏弹性值(LSM)、TIMP-1,治疗6个月后TIMP-1、M-CSF均高于未发生组(P<0.05),ΔTIMP-1、ΔM-CSF则低于未发生组(P<0.05)。logistic回归分析结果显示,LSM、ΔTIMP-1及ΔM-CSF均为影响CHB患者肝硬化发生的危险因素(OR=2.732、3.040、3.330,P<0.05)。ROC分析显示,血清ΔTIMP-1、ΔM-CSF预测CHB患者肝硬化发生的最佳截断点分别为50.9μg/L、61.4 ng/L,灵敏度分别为80.6%、72.2%;特异度分别为75.9%、80.4%,AUC分别为0.848、0.809。结论 血清TIMP-1、M-CSF水平变化预测CHB患者肝硬化发生的效能较高,可作为临床评估CHB患者是否发生肝硬化的重要参考指标。 展开更多

目的:探讨氨基胍对肺纤维化(PF)大鼠基质金属蛋白酶-13(MMP-13)和组织基质金属蛋白酶抑制物-1(TIMP-1)表达的影响及其对转化生长因子-β1/信号转导分子(TGF-β1/Smads)信号通路的调控机制。方法:SD大鼠随机分为4组:正常组、模型组、实... 目的:探讨氨基胍对肺纤维化(PF)大鼠基质金属蛋白酶-13(MMP-13)和组织基质金属蛋白酶抑制物-1(TIMP-1)表达的影响及其对转化生长因子-β1/信号转导分子(TGF-β1/Smads)信号通路的调控机制。方法:SD大鼠随机分为4组:正常组、模型组、实验组、地塞米松组(对照组),每组30只。模型组、实验组、对照组大鼠气管滴注博莱霉素构建PF模型,正常组大鼠滴注等剂量等浓度氯化钠溶液。滴注完毕后,实验组、对照组大鼠每日分别腹腔注射1 ml氨基胍溶液(20 mg/kg)和1 ml地塞米松溶液(20 mg/kg),正常组、模型组分别腹腔注射等剂量生理盐水。处死大鼠,qRT-PCR、免疫组化、Western blot检测各组大鼠肺组织病理学及纤维化改变、TGF-β1、Smad2和Smad7 mRNA表达、MMP-13和TIMP-1表达、TGF-β1、p-Smad2、pSmad7蛋白表达。结果:与正常组相比,模型组肺组织TGF-β1和Smad2 mRNA表达、TGF-β1、p-Smad2和TIMP-1表达明显升高,Smad7 mRNA和p-Smad7蛋白表达、MMP-13表达明显降低;与模型组相比,实验组和对照组大鼠肺组织TGF-β1和Smad2mRNA表达、TGF-β1、p-Smad2、TIMP-1表达明显降低,Smad7 mRNA和p-Smad7蛋白表达、MMP-13表达明显升高(P<0.05)。结论:氨基胍可保护博来霉素诱导的PF大鼠肺组织,其抗纤维化作用可能通过调控TGF-β1/Smads信号通路实现。 展开更多

目的检测油酸制备大鼠急性肺损伤模型血浆及肺组织基质金属蛋白酶9(matrix metalloproteinases9,MMP-9)及组织基质金属蛋白酶抑制因子-1(tissue matrix metalloproteinase inhibitor-1,TIMP-1)含量,分析MMP-9及TIMP-1的表达特点及临床... 目的检测油酸制备大鼠急性肺损伤模型血浆及肺组织基质金属蛋白酶9(matrix metalloproteinases9,MMP-9)及组织基质金属蛋白酶抑制因子-1(tissue matrix metalloproteinase inhibitor-1,TIMP-1)含量,分析MMP-9及TIMP-1的表达特点及临床意义。方法雄性SD大鼠24只分为3组:空白对照组(n=8)、油酸造模2h组(n=8)、油酸造模6h组(n=8)。空白对照组大鼠尾静脉注射0.9%氯化钠注射液(0.1mL/kg)6h后观察。油酸造模组大鼠尾静脉缓慢注射油酸(OA,0.1mL/kg)2h、6h后观察,以复制油酸诱导大鼠急性肺损伤模型。观察及检测指标为光镜下肺组织病理形态学变化、动脉血氧分压(PaO2)、肺湿/干质量比(W/D)、血浆和肺组织MMP-9和TIMP-1含量。结果①光镜下,与空白对照组比较,油酸造模组大鼠明显可见肺间质及肺泡水肿,肺泡腔内有浆液性渗出物及大量炎性反应细胞、红细胞渗出,部分肺间隔增厚,呈均匀红染的"透明膜"样改变;②与空白对照组比较,2h及6h时,油酸造模组PaO2明显降低(P<0.001),W/D明显升高(P<0.05);③与空白对照组比较,2h及6h时,油酸造模组血浆和肺匀浆MMP-9含量明显升高(P<0.001),TIMP-1含量明显降低(P<0.05,P<0.01或P<0.001);2h与6h比较,油酸造模组血浆和肺匀浆MMP-9及TIMP-1含量无明显变化(P>0.05);④2h及6h时,血浆MMP-9与肺匀浆MMP-9含量呈高度正相关(r=0.999,P<0.001或r=0.885,P<0.05),血浆TIMP-1与肺匀浆TIMP-1含量呈高度正相关(r=0.850,P<0.001或r=0.800,P<0.05)。结论油酸制备急性肺损伤大鼠模型血浆、肺匀浆MMP-9含量明显升高,TIMP-1含量明显降低,两者随发病时间延长无明显变化。血浆MMP-9、TIMP-1与肺匀浆MMP-9、TIMP-1含量高度正相关,检测血浆MMP-9、TIMP-1有助于急性肺损伤的病理诊断和治疗,从而为临床急性肺损伤的诊断、治疗及预后提供简单、可行、准确的检测方法和途径。 展开更多

目的观察中成药冠心舒通胶囊对高脂饮食喂养ApoE-/-小鼠主动脉粥样硬化斑块病理变化、人组织型基质金属蛋白酶抑制剂1(TIMP-1)、基质金属蛋白酶9(MMP-9)表达的影响,探讨冠心舒通胶囊对粥样斑块稳定性的影响以及对相关机制进行初步研究... 目的观察中成药冠心舒通胶囊对高脂饮食喂养ApoE-/-小鼠主动脉粥样硬化斑块病理变化、人组织型基质金属蛋白酶抑制剂1(TIMP-1)、基质金属蛋白酶9(MMP-9)表达的影响,探讨冠心舒通胶囊对粥样斑块稳定性的影响以及对相关机制进行初步研究。方法将8周龄雄性ApoE-/-小鼠30只给予高脂喂养,12周时随机分为:模型组、冠心舒通胶囊高剂量[1.8 g/(kg·天)]组和冠心舒通胶囊低剂量[0.6 g/(kg·天)]组,喂养12周后处死,取主动脉中段,大体标本油红O染色观察斑块面积,冰冻切片后HE染色观察斑块厚度,冰冻切片免疫荧光法检测斑块内TIMP-1、MMP-9表达情况。结果与模型组相比,冠心舒通胶囊组平均斑块浸润面积减小(P<0.05),弥漫程度减轻,斑块厚度降低,MMP-9表达减少,而且冠心舒通胶囊高剂量组与低剂量组相比,各项统计指标亦有统计学意义(P<0.05),TIMP-1表达未见明显变化。结论冠心舒通胶囊可以对抗ApoE-/-小鼠动脉粥样硬化斑块的形成和进展,以及通过降低斑块内MMP-9表达,从而抑制胶原纤维分解,稳定易损粥样斑块。 展开更多

Objective: To investigate the effects of Biejia Ruangan Tablet (复方鳖甲软肝片, BRT)- containing serum on the expression of matrix metalloproteinase (MMP-9) and tissue inhibitor of metalloproteinase (TIMP-1) in cultured renal interstitial fibroblasts. Methods: Different BRT-containing sera were prepared by gastric gavages to rats with the high-dose (7 g/kg), mid-dose (3.5 g/kg), and low-dose (1.75 g/kg) BRT respectively. The expression of extracellular matrix in NRK-49F cells was induced by treatment with human transforming growth factor-β1 (recombined human TGF-β 1), and BRT-containing serum. Western blotting and Northern blotting were used to measure type I and III procollagen, MMP-9, and TIMP-1. Results: The high dose BRT-containing serum could decrease the type Ⅰ and Ⅲ procollagen gene expression which boosted by TGF- 13 1, at the same time cut down TIMP-1 protein and gene expression which increased by TGF- β1 (P〈0.05). Treatment of cells with recombined human TGF-β 1 had no significant effect on MMP-9 expression and BRT- containing serum also had no effect on MMP-9 expression. Conclusions: High dose BRT has anti-fibrosis effects in NRK-49F cells, as indicated by its inhibition of type Ⅰ and Ⅲ procollagen and TIMP-1 expression. 展开更多

Activity of matrix metalloproteinase-9 increases following cerebral ischemia/reperfusion, and is associated with cerebral microvascular permeability, blood-brain barrier destruction, inflammatory cell infiltration and brain edema. Matrix metalloproteinase-9 also likely participates in thrombolysis. A rat model of middle cerebral artery infarction was established by injecting autologous blood clots into the internal carotid artery. At 3 hours following model induction, urokinase was injected into the caudal vein. Decreased neurological severity score, reduced infarct volume, and increased expression of matrix metalloproteinase-9 and tissue inhibitor of metalloproteinase-1 were observed in the cerebral cortex 24 hours after urokinase thrombolysis. These results suggest that urokinase can suppress damage in the acute-early stage of cerebral infarction. 展开更多

AIM: To compare matrix metalloproteinase (MMP)-9 and tissue inhibitor of metalloproteinase (TIMP)-1 in gastric ulcer (GU) and chronic superficial gastritis (CSG). METHODS: This study enrolled 63 patients with GU and 25 patients with CSG. During upper gastroduodenal endoscopy, we took samples of gastric mucosa from the antrum and ulcer site from patients with GU, and samples of antral mucosa from patients with CSG. Mucosal biopsy tissues were cultured for 24 h, and the culture supernatant was measured for levels of MMP-9 and TIMP-1. After receiving eradication therapy for Helicobacter pylori (H. pylori ) and 8 wk proton-pump inhibitor therapy for GU, follow-up endoscopy examination was performed after 6 mo and whenever severe symptoms occurred. RESULTS: Levels of MMP-9 and TIMP-1 at the ulcer site or in the antrum were significantly higher in GU than CSG patients. MMP-9 levels at the ulcer site were significantly higher than in the antrum in GU patients, and had a significantly positive correlation with TIMP-1. MMP-9 levels were significantly higher in H. pylori -positive than H. pylori -negative GU and CSG patients. Levels of MMP-9 or TIMP-1 at the ulcer site were associated with the histological severity of activity and inflammation. About 57 GU patients were followed up, and seven had GU recurrence. H. pyloriinfection and MMP-9 levels were risk factors for the recurrence of GU adjusted for age and sex by multiple logistic regression analysis. CONCLUSION: MMP-9 may perform an important function in gastric ulcer formation and recurrence. 展开更多

Small hairpin RNA (shRNA) was used to silence the HIF1α gene in human retinal pigment epithelial cells (RPE) under hypoxia in order to observe the effect of gene silencing on the expression of matrix metalloproteinase tissue inhibitor 1 (TIMP1). By using chemical hypoxic inducer CoCl2 to mimic RPE hypoxic environment, shRNA against the targeting region of HIF1α mRNA sequence was synthesized by a method of in vitro transcription, and the HIF1α was interfered in RPE cultured under hypoxia (induced by 150 μmol/L CoCl2 ). RT-PCR was employed to detect the expression of HIF1α and TIMP1. The expression levels of HIF1α and TIMP1 were measured by using Western blotting. The results showed that after the RPE were transfected with specific shRNA against HIF1α mRNA, RT PCR revealed that under hypoxia, the efficacy of HIF1α gene silencing in RPE was 83.4 %. Western blotting revealed that the expression levels of HIF1α protein was dramatically dropped. In addition, RT-PCR results demonstrated that the expression of TIMP1 mRNA was decreased by 28.9 %, and the expression levels of TIMP1 protein were also significantly reduced by Western blotting. It was suggested that shRNA targeted against HIF1α mRNA could effectively silence the HIF1α gene, subsequently effectively inhibit the hypoxia-induced up-regulation of TIMP1. 展开更多

BACKGROUND: Matrix metalloproteinase-9 (MMP-9) can degrade collagen IV (the main structural ingredient of basilar membrane), and it also plays an important role in tumor vascularization, tumor cell progression, formation of metastatic focus, etc. Tissue inhibitor of metalloproteinase-1 (T1MP-1) can bind with MMP-9 to form 1 : 1 compound and inhibit its activity, and can negatively regulate the tumor progression and metastasis. OBJECTIVE: To analyze the relationship of MMP-9 and T1MP-1 expressions with the pathological grade, metastasis and prognosis of malignant peripheral nerve sheath tumor (MPNST). DESIGN: An observational comparative experiment. SETTING: Heze Medical College. PARTICIPANTS: Fifty-eight surgical pathological samples, which were clearly diagnosed to be MPNST, were collected from the pathological laboratory archives in the Department of Pathology, Heze Municipal Hospital from January 1988 to December 2003. The MPNST pathological types were common tumor in 53 cases, malignant triton tumor in 2 cases, epithelial MPNST in 2 cases and MPNST with gland differentiation in 1 case. The pathological grade was grade 1 in 11 cases, grade 2 in 24 cases and grade 3 in 23 cases. Besides, the resected tumor samples of 20 patients with benign peripheral nerve tumor (10 cases of nerve sheath tumor and 10 cases of neurofibromatosis) and the normal peripheral nerves (by-products of some surgeries) of 5 patients were also collected. The samples were used with the approval of the patients. Rat-anti-human MMP-9, TIMP-1 monoclonal antibody and S-P kit were purchased from Fuzhou Maixin Biotechnology, Co.,Ltd. METHODS: The documented paraffin blocks were again prepared to sections of 5 lJ m. The expressions of MMP-9 and TIMP-1 in the samples were detected with mmunohistochemical S-P method. The objective of this study was to investigate the correlations between papillary thyroid cancer and peripheral blood levels of matrix metalloproteinase-2, matrix metalloproteinase-9, tissue inhibitor of metalloproteinase-1, and tissue inhibitor of metall0Proteinase-2. Methods The correlations were studied bY detecting the levels of matrix metalloproteinase-2, matrix metalloproteinase-9, tissue inhibitor of metalloProteinase-1, and tissue inhibitor of metalloproteinase-2 by enzyme-linked immunosorbant assay and reverse-transcription polymerase chain reaction in the peripheral blood of 30 patients with papillary thyroid carcinoma, 27 patients with benign thyroid disease, and 25 hea !hy vo!unteers. Results The leve!s of matrix metalloproteinase-2, matrix metalloproteinase-9, tissue inhibitor of metalloproteinase-1, and tissue inhibitor of metalloproteinase-2 in the peripheral blood of patients with papillary thyroid carcinoma were significantly higher than those in the peripheral blood of patients with benign thyroid disease and healthy volunteers (P 〈0.05). However, there were no significant differences between patients with benign thyroid disease and healthy volunteers (P 〉0.05). The accuracy of detection by both enzyme-linked immunosorbant assay and reverse-transcription polymerase chain reaction in the papillary thyroid cancer group was 83.33%. Conclusions The levels of matrix metalloproteinase-2, matrix metalloproteinase-9, tissue inhibitor of metalloproteinase-1, and tissue inhibitor of metalloproteinase-2 in the peripheral blood are helpful in identifying thyroid carcinoma and aid in preoperative assessment. 展开更多
