[目的]评价乳腺癌患者血清可溶性T细胞免疫球蛋白及黏蛋白分子-3(souble T cell immunoglobulin and mucin-domain containing molecule-3,sTim-3)及其配体半乳糖凝集素-9(galactose lectin-9,Galectin-9)、癌胚抗原相关细胞黏附分子1(c...[目的]评价乳腺癌患者血清可溶性T细胞免疫球蛋白及黏蛋白分子-3(souble T cell immunoglobulin and mucin-domain containing molecule-3,sTim-3)及其配体半乳糖凝集素-9(galactose lectin-9,Galectin-9)、癌胚抗原相关细胞黏附分子1(carcino-embryonic antigen related cellular adhesion molecule-1,CEACAM-1)表达水平,并探讨其临床意义。[方法]收集2019年7月至2020年10月武汉市中西医结合医院甲乳外科住院的乳腺癌患者90例血清,同期选择选择60名年龄相应匹配的健康体检者血清为对照组。酶联免疫吸附法检测乳腺癌患者和对照组血清sTim-3、Galectin-9、CEACAM-1表达水平。ROC曲线分析血清sTim-3、Galectin-9对乳腺癌的诊断学效能。[结果]乳腺癌患者血清s Tim-3[2.59(1.78~4.03)ng/ml]和Galectin-9[2.20(1.55~3.05)ng/ml]表达水平显著性高于对照组sTim-3[1.79(1.19~2.45)ng/ml]和Galectin-9[1.59(1.18~2.15)ng/ml](P<0.05),乳腺癌患者血清CEACAM-1水平与对照组相比无显著性差异(P=0.622)。乳腺癌患者血清s Tim-3表达水平与年龄、肿瘤大小、临床分期、腋窝淋巴结转移显著性相关(P<0.05),与组织学分级、Ki-67表达无显著性相关(P>0.05)。Galectin-9表达水平与临床病理学特征无显著性相关(P>0.05)。乳腺癌患者血清CEACAM-1表达水平随组织学分级的升高而增高(P=0.025),与其他临床病理学特征无显著性相关(P>0.05)。sTim-3和Galectin-9诊断乳腺癌的曲线下面积分别为0.741(95%CI:0.666~0.816),0.692(95%CI:0.599~0.785)。[结论]乳腺癌患者血清sTim-3及其配体Galectin-9表达水平显著性上调,血清sTim-3与多个临床病理学特征相关,对乳腺癌的诊断具有一定价值。CEACAM-1在乳腺癌患者血清中表达未见升高,但与组织学分级存在一定关联,其与乳腺癌的关系有待进一步研究。展开更多

The role of Clostridium butyricum (C. butyricum) in reducing intestinal low-grade inflammation via immune pathways has been well defined. However, the detailed mechanisms of the effects of C. butyricum on intestinal mucosal immunity, especially on immune cells of the lamina propria, remain unclear. Dendritic cells (DCs), which are important immune cells, secrete proinflammatory cytokines (IL-1β, IL-6, and others) and express T cell immunoglobulin and mucin domain-3 (TIM3), promoting proliferation and activation of DCs, and mediating Th1 and Th17 inflammatory responses. AIM To investigate the role of DCs in the development of IBS in a rat model and to understand the regulation of DCs after C. butyricum intervention. METHODS An IBS animal model was established using C57BL/6 mice, and C. butyricum was continuously administered via the intragastric route to simulate different intestinal immune states. Intestinal visceral hypersensitivity and histopathology were assessed using the abdominal withdrawal reflex (AWR) test and hematoxylin & eosin (H&E) staining, respectively. The expression of proinflammatory cytokines (IL-1β and IL-6) and TIM3 was analyzed by Western blot analysis and real-time PCR. Flow cytometry was applied to analyze the quantity, function, and membrane molecule TIM3 of the lamina propria dendritic cells (LPDCs). The regulatory effect of C. butyricum was verified in bone marrowderived dendritic cells by in vitro experiments. RESULTS The secretion of proinflammatory cytokines (IL-1β and IL-6) in mice with IBS was significantly increased compared with that of the control group, which suggested that the intestinal mucosa in mice with IBS was in a low-grade inflammatory state. 目的探讨T细胞免疫球蛋白和黏蛋白结构域分子3(T cell immunoglobulin and mucin domain protein 3,Tim-3)在晚期血吸虫病患者外周血CD56+自然杀伤细胞(NK细胞)表面的表达及其与肝纤维化指标的关系。方法采用流式细胞术检测28例临床晚...目的探讨T细胞免疫球蛋白和黏蛋白结构域分子3(T cell immunoglobulin and mucin domain protein 3,Tim-3)在晚期血吸虫病患者外周血CD56+自然杀伤细胞(NK细胞)表面的表达及其与肝纤维化指标的关系。方法采用流式细胞术检测28例临床晚期血吸虫病患者和30例健康者CD56+NK细胞表面Tim-3的表达,应用全自动化学发光免疫分析仪检测肝纤维化血清学三型前胶原N端肽(PIIIP N-P)、层黏连蛋白(LN)、IV型胶原(CIV)和透明质酸(HA)等4项指标,ELISA检测血清中γ-干扰素(IFN-γ)和白细胞介素-4(IL-4)水平,Fibroscan仪检测肝硬度值(LSM)反映肝纤维化程度。统计分析Tim-3表达水平与IL-4和IFN-γ水平、肝脏纤维化程度以及肝纤维化血清指标的相关性。结果流式细胞分析结果显示,晚期血吸虫病患者外周血CD56+NK细胞表面Tim-3表达水平为(62.3±11.4)%,高于健康对照组的(52.1±6.5)%(P<0.01)。晚期血吸虫病患者和健康对照组的PIIIP N-P水平分别为(86.5±29.5)ng/ml和(22.0±7.8)ng/ml,LN分别为(49.3±21.5)ng/ml和(20.4±6.3)ng/ml,CIV分别为(67.5±22.3)ng/ml和(22.0±3.9)ng/ml,HA分别为(645.9±483.1)ng/ml和(54.7±27.7)ng/ml,两组各指标差异均具有统计学意义(P<0.05)。晚期血吸虫病患者和健康对照组IFN-γ水平分别为(93.9±20.1)ng/L和(107.7±24.6)ng/L,IL-4分别为(46.6±11.8)ng/L和(28.9±8.9)ng/L,两组差异均具有统计学意义(P<0.05)。Spearman非参数相关分析结果显示患者CD56+NK细胞表达的Tim-3水平与患者肝纤维化指标LSM、PIIIP N-P、LN、CIV和IL-4水平呈正相关(r=0.528~0.834,P<0.01),与IFN-γ水平呈负相关(r=-0.501,P<0.01),而与HA水平无明显相关性(r=0.352,P>0.05)。结论晚期血吸虫病患者外周血CD56+NK细胞Tim-3表达呈上调,并与患者肝纤维化指标LSM、PIIIP N-P、LN、CIV和IL-4水平呈正相关。展开更多

Thirty-one pregnant women with RSA and 27 norm...The relationship between T cell immunoglobulin domain and mucin domain protein 3(Tim-3)/Galectin(Gal)-9 pathway and recurrent spontaneous abortion(RSA) was studied. Thirty-one pregnant women with RSA and 27 normal early gravidas were investigated to detect the levels of Tim-3 and Gal-9 in villi and deciduas by Western blotting. Meanwhile, the concentration of interleukin(IL)-4 and IL-12 in peripheral blood plasma was determined by ELISA in 25 healthy fertile non-pregnant controls, the normal early gravidas and pregnant women with RSA mentioned above, respectively. It was found that the relative expression levels of Tim-3 and Gal-9 in villi and deciduas were significantly increased in pregnant women with RSA as compared with those in the normal early gravidas. The concentration of IL-4 in peripheral blood plasma of pregnant women with RSA was lower than that of the normal early gravidas(P〈0.05) and healthy fertile non-pregnant controls(P〈0.05), but that of IL-2 in pregnant women with RSA was significantly higher than that of the normal early gravidas(P〈0.05) and healthy fertile non-pregnant controls(P〈0.05). It was suggested that the overexpression of Tim-3/Gal-9 pathway may be related to the pathogenesis of RSA.展开更多
