用醇提-酸醇水解法提取了龙葵内的澳洲茄胺,对其进行了定性、定量分析;以澳洲茄胺为参照品,表征了甾体类生物碱在龙葵中所占质量分数,考察了龙葵果实成长期内甾体类生物碱的形成、变化趋势。发现甾体类生物碱主要富集在龙葵未成熟果实中... 用醇提-酸醇水解法提取了龙葵内的澳洲茄胺,对其进行了定性、定量分析;以澳洲茄胺为参照品,表征了甾体类生物碱在龙葵中所占质量分数,考察了龙葵果实成长期内甾体类生物碱的形成、变化趋势。发现甾体类生物碱主要富集在龙葵未成熟果实中,检测出其在未成熟果实内的质量分数最高达到3.52%;茎叶所含甾体类生物碱的质量分数低于0.5%。果实内甾体类生物碱的变化在其生长中经历了增加、减少两个时段,在结果一个月后,果实内的甾体类生物碱质量分数达到最大,高达2.5%以上;果实成熟时甾体类生物碱质量分数反而最小,低于1%;阳光照射程度的不同会改变龙葵果实的生长期,从而对甾体类生物碱的质量分数变化产生影响。因此,龙葵的采摘及其生物碱入药,需着重考虑生物碱在该植物不同部位的分布及生长周期、阳光照射等因素。 展开更多

目的建立龙葵中澳洲茄胺的高效液相色谱(HPLC)检测方法。方法采用甲醇酸水超声提取龙葵生物碱,在回流条件下对生物碱苷进行酸性水解,得到相应的澳洲茄胺。然后进行高效液相色谱检测,色谱条件:色谱柱为Diamonsil C18色谱柱(250mm&... 目的建立龙葵中澳洲茄胺的高效液相色谱(HPLC)检测方法。方法采用甲醇酸水超声提取龙葵生物碱,在回流条件下对生物碱苷进行酸性水解,得到相应的澳洲茄胺。然后进行高效液相色谱检测,色谱条件:色谱柱为Diamonsil C18色谱柱(250mm×4.6mm),乙腈-0.05%七氟丁酸(30:70)体系为流动相,流速为1.0ml/min,检测波长为210nm。结果龙葵生物碱获得了良好的基线分离;澳洲茄胺的线性范围为102~612μg·ml-1(r=0.9999);加样回收率超过96.0%,RSD为3.35%(n=6)。结论该方法简单快速,准确可靠,可作为龙葵药材及其制剂的质量控制方法。该方法也为龙葵质量标准的制定及进一步的药效学研究提供了可靠的数据和方法学平台。 展开更多

Through basic and clinical research, non-invasive treatments like Curaderm have been developed, leading to improved quality of life for patients. Excipients, previously considered inactive ingredients, play a crucial role in enhancing the performance of topical formulations. The development of Curaderm emphasizes the importance of understanding the interactions between active ingredients, excipients, and the biological system to create effective and affordable pharmaceutical formulations. The systematic approach taken in the development of Curaderm, starting from the observation of the anticancer activity of natural solasodine glycosides and progressing through toxicological and efficacy studies in cell culture, animals, and humans, has provided insights into the pharmacokinetics and pharmacodynamics of solasodine glycosides. It is crucial to determine these pharmacological parameters within the skin’s biological system for maximal effectiveness and cost-effectiveness of a skin cancer treatment. Solasodine Glycosides: A Topical Therapy for Actinic Keratosis. A Single-Blind, Randomized, Placebo-Controlled, Parallel Group Study with Curaderm<sup>BEC5</sup> 被引量:3

This communicatio... Background: Untreated actinic keratosis can advance to squamous cell carcinoma, which in turn is associated with a risk of metastasis. Current treatments for actinic keratosis have many shortcomings. This communication describes the efficacy and safety of a topical cream therapy, CuradermBEC5, containing solasodine glycosides (0.005%) for actinic keratosis.Methods: Randomly assigned patients with actinic keratosis on the face, trunk or extremities received so-lasodine glycosides cream (CuradermBEC5) or placebo (vehicle) that was self-applied to the lesions and covered with an occlusive dressing (micropore) twice daily for 3 consecutive days. Complete clearance and local reactions were as-sessed at 56 days with follow-up periods of 6 months and 1 year. Results: The rate of complete clearance at day 56 was higher with solasodine glycosides than with placebo (92% vs. 38%, P 0.001). The absolute success rates after 1 year follow-up were 82% for solasodine glycosides and 18% for placebo. No differences in local reactions were obtained when solasodine glycosides and placebo were compared. Local reactions in both groups peaked at days 2 and 3 with local pain as the major event. The pain associated with treatments lasted approximately 10 minutes after application of solasodine glycosides and placebo. Complete reepithelialization occurred two weeks after treatment. Adverse events were generally mild to moderate in intensity and resolved without sequelae. Conclusions: Solasodine glycosides cream applied topically twice daily with a dressing for 3 days is effective for the treatment of actinic keratoses. 展开更多

Background: Psoriasis is a chronic disease that can have significant effects on quality of life. Aim: To test whether the antineoplastic, antipathogenic plant derived secondary metabolites, solasodine glycosides, can treat psoriasis. Case Presentation: We report a case of a 54-year-old French-Vietnamese male who presented with diagnosed erythematous scaly annular recalcitrant psoriasis scattered throughout his body. Method: After failing conventional treatment regimens for over 10 years the patient received a trial with a topical cream formulation PsorendBEC, containing solasodine glycosides, for his psoriasis. Result: Topical applications of PsorendBEC twice daily resulted in complete resolution of cutaneous lesions after 4 weeks of treatment with no recurrence post 1 year after therapy. Conclusion: Topical PsorendBEC therapy rapidly removes recalcitrant psoriasis with no apparent side effects. 展开更多

Solasodine rhamnosides produced in plants as secondary metabolites, are safe and effective when treating a variety of cancers, including non-melanoma skin cancers. They are cytotoxic against multi-drug resistant tumor cells, stimulate lasting immunity against cancer, are not mutagenic and display anti-mutagenic properties. These antineoplastics, through cellular specific receptor-mediated actions, directly induce apoptosis by triggering extrinsic and intrinsic apoptotic pathways in cancer cells but not normal cells. CuradermBEC5 contains solasodine rhamnosides and is a topical formulation for the treatment of keratoses and non-melanoma skin cancers. The mode of action, together with the selectivity towards cancer cells, with CuradermBEC5 therapy, results in outstanding beneficial outcomes. This study shows graphically and pictorially that CuradermBEC5 seeks and destroys basal cell carcinoma whilst normal skin cells replace the dead cancer cells during therapy, emanating into impressive cosmetic end results. The clinical observations with CuradermBEC5 therapy reveal that initially the lesion size increases over four-fold due to the interaction of CuradermBEC5 with deeper and more lateral tumor cells, followed by a decrease in size, ultimately, resulting in complete elimination of the basal cell carcinoma. 展开更多

目的探讨澳洲茄胺对人骨肉瘤移植瘤的抑制作用及其机制。方法该研究于2018年1月至2019年3月完成。MG63细胞株购于中科院上海细胞库。将0.2 mL人骨肉瘤MG63细胞悬液(共2×106个细胞)注射于裸鼠右侧腋窝皮下,建立人骨肉瘤移植瘤模型,... 目的探讨澳洲茄胺对人骨肉瘤移植瘤的抑制作用及其机制。方法该研究于2018年1月至2019年3月完成。MG63细胞株购于中科院上海细胞库。将0.2 mL人骨肉瘤MG63细胞悬液(共2×106个细胞)注射于裸鼠右侧腋窝皮下,建立人骨肉瘤移植瘤模型,实验分为4组,即对照组和澳洲茄胺组(10 mg/kg、20 mg/kg、40 mg/kg),隔日腹腔注射给药1次,连续4周。测量移植瘤的质量和体积,原位末端标记(TUNEL)染色法检测细胞的凋亡,蛋白质印迹法检测凋亡相关蛋白、血管生成相关因子和磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/AKT)信号通路的表达。结果与对照组相比,澳洲茄胺组的移植瘤质量(0.45±0.12)g和在21 d(510.62±95.78)mm~3和28 d(579.56±92.04)mm~3的体积降低,且以40 mg/kg组的抑制效应最明显(P<0.05)。与对照组相比,澳洲茄胺组的原位末端标记的异硫氰酸荧光素(TUNEL-FITC)的荧光强度升高,且以40 mg/kg组最明显。与对照组相比,澳洲茄胺组移植瘤中B细胞淋巴瘤/白血病-2相关X蛋白(Bax)(3.55±0.39)、活化的半胱氨酸天冬氨酸蛋白酶-3(Cleaved caspase-3)(3.94±0.33)、活化的半胱氨酸天冬氨酸蛋白酶-9(Cleaved caspase-9)(3.59±0.31)、凝血酶敏感蛋白1(TSP-1)(3.70±0.41)蛋白的表达量上调,B细胞淋巴瘤/白血病-2(Bcl-2)(0.31±0.15)、血管内皮生长因子(VEGF)(0.25±0.12)、磷酸化磷脂酰肌醇3-激酶(p-PI3K)(0.39±0.12)和磷酸化蛋白激酶B(p-AKT)(0.33±0.11)蛋白的表达量下调,且以40 mg/kg组的效应最明显(P<0.05)。结论澳洲茄胺可以抑制人骨肉瘤细胞皮下移植瘤的生长,诱导细胞凋亡,下调VEGF的表达,上调TSP-1的表达,其作用机制可能与PI3K/AKT信号通路的抑制有关。 展开更多

目的研究澳洲茄胺(solasodine)对人骨肉瘤细胞的增殖、凋亡及迁移的影响,探讨澳洲茄胺抑制人骨肉瘤细胞生长的作用机制。方法以澳洲茄胺浓度为梯度和澳洲茄胺处理时间为梯度,采用MTT法检测澳洲茄胺对人骨肉瘤细胞MG-63的增殖影响,并通... 目的研究澳洲茄胺(solasodine)对人骨肉瘤细胞的增殖、凋亡及迁移的影响,探讨澳洲茄胺抑制人骨肉瘤细胞生长的作用机制。方法以澳洲茄胺浓度为梯度和澳洲茄胺处理时间为梯度,采用MTT法检测澳洲茄胺对人骨肉瘤细胞MG-63的增殖影响,并通过趋势线计算处理48 h时澳洲茄胺对MG-63的半抑制浓度;将半抑制浓度澳洲茄胺处理过的MG-63设为处理组,未处理的MG-63设为对照组,用流式分析技术检测2组细胞凋亡情况,用Transwell检测2组细胞的侵袭转移情况,并用Western blot检测2组细胞中与调亡、侵袭、迁移相关信号通路AKT/GSK-3β/β-catenin相关蛋白的表达情况。结果MTT检测结果显示,澳洲茄胺明显抑制人骨肉瘤细胞MG-63的增殖,并呈现浓度依赖性。澳洲茄胺处理48 h时,澳洲茄胺对MG-63的半抑制浓度为54.03μmol/L。流式分析结果显示,澳洲茄胺处理组细胞凋亡率显著高于对照组(P<0.01)。Transwell结果显示,澳洲茄胺处理组细胞迁移能力与侵袭能力均明显低于对照组(P<0.01)。Western blot结果显示,与对照组相比,MG-63细胞经澳洲茄胺处理后pAKT、pβ-cateninze显著减少(P<0.01),pGSK-3β显著增加(P<0.01)。结论澳洲茄胺介导人骨肉瘤细胞AKT/GSK-3β/β-catenin信号通路,促进人骨肉瘤细胞凋亡,抑制人骨肉瘤细胞增殖和侵袭转移。 展开更多

First glycosylation before N-cyclization significantly facilitated synthesis of the desired molecule.
