Notopterygium incisum(QH) has been used for the treatment of rheumatoid arthritis(RA), and volatile oils may be its mainly bioactive constituents. The present study was designed to analyze the volatile compounds in QH and to determine the anti-arthritic capacity of Notopterygium volatile oils and the potential mechanism of action. The volatile compounds analysis was conducted by GC-MS. The anti-arthritic capacity test of the volatile oils was conducted on adjuvant-induced arthritis(AIA) rats. The anti-inflammatory property was tested in NO release model in RAW 264.7 cells. Endothelial cells were used to evaluate the anti-proliferative and anti-tube formative effects. 70 compounds were analyzed by GC-MS in the volatile oils. Notopterygium volatile oils weakened the rat AIA in a dose-dependent manner(2, 4, and 8 g crude drug/kg). The NO production by RAW 264.7 was decreased by more than 50% in Notopterygium volatile oils(5, 15, and 45 μg·mL^(-1)) pretreated groups. Notopterygium volatile oils also inhibited EAhy926 cell proliferation and further delayed EAhy926 cell capillary tube formation in a concentration-dependent manner. The anti-NO productive, anti-proliferative, and anti-tube formative effects of Notopterygium volatile oils strongly suggested that the therapeutic effect of QH in AIA might be related to the potent anti-inflammatory and anti-angiogenic capacities of the volatile oils. 展开更多

Figs (Moracea: Ficus) and fig wasps (Hymenoptera: Chlocloids: Agaonideae) depend on each other to complete their reproduction. Monoecious fig species and their pollinating wasps are in conflict over the use of fig ovaries which can either produce one seed or one wasp. From observation on Ficus virens Ait., we showed that female flowers with outer layer of ovaries (near to the wall of syconium) had no significant difference from that with inner and interval layer of ovaries (near to the syconium cavity), in which most seeds and wasps were produced. This meant that fig tree provided the same potential resource for seed and wasps production. Observation indicated that there was usually only one foundress in syconium at female flower phase and no com- petition pollinators. Measurement of the style length of female flowers and the ovipositor of pollinators indicated that most ovaries could be reached by pollinator’s ovipositor. However, at the male flower phase, production of seeds was significantly more than that of wasps including non-pollinating wasps but there was no significant difference between seed and pollinating wasp production when without non-pollinating wasps produced. This result indicated that non-pollinating wasps competed ovaries not with seeds but with pollinating wasps for ovipositing. Bagged experiment showed that the sampling fig species was not self-sterile which was important for figs and wasps to survive bad season. Seed production in self-pollinated figs was not significantly different from total wasps in- cluding non-pollinating ones. 目的探讨双歧杆菌脂磷壁酸(LTA)对小鼠腹腔巨噬细胞体外吞噬功能和NO释放的影响。方法将无菌收集培养的小鼠腹腔巨噬细胞分为RPMI 1640完全培养液组和地塞米松处理组(终浓度为100μg/ml),分别向每组培养孔内加入不同终质量浓度的LTA,于... 目的探讨双歧杆菌脂磷壁酸(LTA)对小鼠腹腔巨噬细胞体外吞噬功能和NO释放的影响。方法将无菌收集培养的小鼠腹腔巨噬细胞分为RPMI 1640完全培养液组和地塞米松处理组(终浓度为100μg/ml),分别向每组培养孔内加入不同终质量浓度的LTA,于培养4 h和24 h后,分别用中性红法和Griess Reagent试剂盒检测并分析LTA对巨噬细胞体外吞噬和NO释放的影响。结果终浓度为5、10、20、40μg/ml的LTA对小鼠腹腔巨噬细胞体外吞噬功能和NO的释放均有明显的促进作用(P<0.05),并且可以有效拮抗地塞米松的抑制效应(P<0.05)。LTA的上述作用与其浓度呈一定的梯度关系。结论LTA不但可以显著激活小鼠腹腔巨噬细胞的体外吞噬功能和促进NO的释放,还能有效拮抗地塞米松对巨噬细胞的体外抑制作用。 展开更多

Effects of structural modification of anti-inflammatory steroidal antedrug on pro-inflammatory mediators and inhibitory cytokines in human alveolar epithelial cells 被引量:1

The anti-inflammatory effects of the new ster-oidal antedrug, 21-acetyloxy-9α-fluoro-11β-hy-droxyl-3, 20-dioxo-1, 4-pregnadieno-[16α, 17α-d] isoxazoline (FP-ISO-21AC), on nitric oxide (NO) and interleukin 8 (IL-8) production, were inves-tigated together with its parent steroid predni-solone (PRED). PRED is one of the anti-in-flammatory steroids but has systemic side ef-fects which limit the use of it. PRED was modi-fied with ‘antedrug concept’ to create safer drugs that attack problems such as inflamma-tion, then quickly become inactive before they can cause systemic side effect. We had a test about the effect of the modified anti-inflamma-tory steroidal antedrug on anti-inflammatory activity. The present study evaluated their ability to inhibit cytokine-induced NO and IL-8 produc-tion in human alveolar epithelial cells. We also investigated their ability to enhance the expres-sion of inhibitory cytokine receptor, interleukin 22 receptor (IL-22R) in human alveolar epithelial cells. Our results showed that FP-ISO-21AC sh- owed higher ability to inhibit the cytokine - in-duced production of NO than PRED. Exogenous IL-22 was added to the media of both human alveolar epithelial cells (A549) and human lung fibroblast (HLF-1). In the presence of the ex-ogenous inhibitory cytokine IL-22, further re-duction of NO production was observed in A549 cells, which express IL-22R, but not in HLF1, which does not express IL-22R. These data suggested that the steroidal antedrugs en-hanced the expression of IL-22R. FP-ISO- 21AC showed higher potency than PRED to restore the expression of IL-22R. FP-ISO-21AC further reduced NO production to 27% and PRED further reduced NO production to 39%. In con-clusion, a synthesized steroidal antedrug FP- ISO-21AC showed higher anti-inflammatory ef-fects than PRED by inhibiting the expression of pro-inflammatory mediator NO and stimulating the expression of IL-22R. 展开更多

Their structures were elucidated on the basis of spectroscopic evidences and hydrolysis products. Bisdesmosidic saponin(3–9) showed modest suppression of NO production with the inhibition ratios in the range of 51.3%– 64.6% at 50 μmol·L^(-1), whereas monodesmosidic saponins with a free carboxyl group at C-28(1 and 2) showed potent inhibitory activities with IC_(50) values being 12.7 and 8.3 μmol·L^(-1), respectively. 展开更多
