Zinc sulfate and quercetin induced and inhibited the expression of HSP72, respectively. Intraperitoneal injections of zinc sulfate, SP600125 (c-Jun N-terminal kinase inhibitor), or quercetin were performed on retinal ganglion cells in a Wistar rat model of chronic ocular hypertension. Our results showed that compared with the control group, the expression of HSP72 in retinal ganglion cells and the lateral geniculate body was increased after the injection of zinc sulfate, but was decreased after the injection of quercetin. The expression of phosphorylated c-Jun N-terminal kinases and phosphorylated c-Jun were visible 3 days after injection in the control group, and reached apeak at 7 days. Zinc sulfate and SP600125 significantly decreased the expression of p-c-Jun, whereas quercetin significantly enhanced the expression of this protein. These results suggest that HSP72 protects retinal ganglion cells and lateral geniculate body in a rat model of chronic ocular hypertension from injury by blocking the activation of the stress-activated kinase/c-Jun N-terminal kinase apoptotic pathway.展开更多

Pathogenic mutations in TP53 gene have only been reported in 50% of colorectal cancer(CRC) patients. A single nucleotide polymorphism(SNP) in the TP53 gene resulting in the presence of either arginine(Arg)or proline(Pro) or both at codon 72 was shown to alter TP53 tumor-suppressor properties. This SNP has been investigated as a risk factor for numerous cancers,including CRC. In this study we analyzed TP53 codon 72 polymorphism distribution in 461 IBD,181 primary sclerosing cholangitis patients and 62 healthy controls. Genotyping of TP53 was performed by sequencing and restriction fragment length polymorphism analysis of genomic DNA extracted from peripheral blood. RESULTS: The most frequent TP53 genotype in IBD patients was Arg/Arg occurring in 54%-64% of cases(and in only 32% of controls). Arg/Pro was the most prevalent genotype in controls(53%) and less common in patients(31%-40%). Pro/Pro frequency was not significantly different between controls and IBD patients. CONCLUSION: The data suggests that the TP53 codon 72 Arg/Arg genotype is associated with increased risk for IBD development.展开更多
This article, based on the field work took place on the zone of Tuomuer (托木尔) Peak, western Tian Shan (天山 ) during the period May 2008 to September 2009, obtained the spatial distribution of debris layer on t...This article, based on the field work took place on the zone of Tuomuer (托木尔) Peak, western Tian Shan (天山 ) during the period May 2008 to September 2009, obtained the spatial distribution of debris layer on the reference glaciers (Glacier No. 72, Glacier No. 74, Tuomuer Glacier) by detailed measurements of debris thickness and ablation rates on glacier and further by Spot-5 (5 m, 2005) high-resolution satellitic image applying remote sensing and geographic information systems approach to research the spatial distribution of debris layer on the zone of Tuomuer Peak. Specifically, the results indicate a sharp in ablation with debris cover thickness increasing from 0-4 cm followed by a decrease in ablation with debris thickness increasing beyond 4 cm for the glaciers No. 72 and No. 74. Spatial distributions of debris layer on the three reference glaciers have the same characteristics, the overall distribution from the vertical, the maximum thickness of debris in the glacier terminal, and the thickness of debris is constantly thinning since the end of the glacier increases with altitude. For the overall distribution from the horizontal, the regular pattern of debris thickness from both sides to the middle is diminishing. The debris on the zone of Tuomuer Peak mostly covered the glacier tongue and is mainly distributed below the altitude of 4 000 m; the area of debris covered approximate accounted for 14.9% on the entire glacier area in this region. Spatial distribution of debris layer on the zone of Tuomuer Peak is mainly affected by the elevation of the glacier terminal, followed by the slopes orientation, the sizes, and so on.展开更多

METHODS: One-hundred and nine controls an...AIM: To evaluate the potential association between p53 codon 72 polymorphism and sporadic colorectal adenocarcinoma development, and human papillornavirus (HPV) infection. METHODS: One-hundred and nine controls and 53 patients with colon cancer from the city of La Plata, Argentina were analyzed, p53 codon 72 genotypes and HPV infection were identified using allele-specific polymerase chain reaction and nested polymerase chain reaction, respectively. RESULTS: The differences in the distribution ofp53 codon 72 polymorphisrn between the cases and controls were statistically significant. The arginine allele had a prevalence of 0.65 in controls and 0.77 in cases. The corresponding odds ratio for the homozygous arginine genotype was 2.08 (95% CI, 1.06-4.05; P〈0.05). Lack of association was found between ,053 polymorphism and HPV infection in the set of adenocarcinomas. CONCLUSION: The findings of the present study indicate that p53 codon 72 arginine homozygous genotype may represent a genetic predisposing factor for colon cancer development. However, further studies are needed in order to elucidate the role of p53 codon 72 polymorphism in colorectal cancer.展开更多

This study aimed to test the hypothesis that ...Background Nitric oxide (NO) is a biologically active molecule which has been reported to protect the heart against ischemia and reperfusion injury in different species. This study aimed to test the hypothesis that nitric oxide may induce the expression of heat shock protein 72 (HSP72) which may protect the heart against ischemia.Methods Rabbits were given intravenous saline or S-nitroso-N-acetylpenicillamine (SNAP), a nitric oxide donor, or Zaprinast, an inhibitor of cyclic guanosine monophosphate (GMP)-phosphodiesterase, which may increase myocardial cyclic GMP content. Twenty-four hours later, the rabbits were either sampled to measure HSP72, or induced with a 30-minute coronary occlusion followed by a 120-minute reperfusion, and then the infarct size was measured. Meanwhile, chelerythrine (CHE, an inhibitor of protein kinase C) was given intravenously 5 minutes before SNAP injection and the effect on HSP72 expression and infarct size was determined.Results Twenty-four hours after pretreatment, immunoblotting showed HSP72 expression increased in the SNAP group compared with control groups, and this was blocked by CHE. Myocardial infarct size in the SNAP group was smaller than that of the control group ((32.4±5.8)% vs (51.1±4.7)%, P 〈0.05). Pretreated with CHE abolished the infarct size-limiting effect of SNAP ((46.0±5.1)%). Pretreatment with Zaprinast neither induced HSP72 expression nor reduced infarct size ((55.4±5.4)%).Conclusion NO induced HSP72 expression and a delayed protection to the heart via the activities of protein kinase C by a cyclic GMP-independent pathway.展开更多
