Investigations of their daily living lead to the conclusion that the “New Unemployed” has gone through a “system withdrawal” process. They are now facing an environment of “system vacuum” that none of other unemployed groups has ever encountered before. The author emphasizes in the paper a strategic employment principle of “For Future Generations” so that focused attention and care can be provided and this toughest challenge in China can be dealt with.展开更多

Methods： In this study, 205 cases of PMOP were divided into the PMOP Shen-yin deficiency group （Group A）, PMOP Shen-yang deficiency group （Group B）, PMOP without Shen deficiency group （Group C）, and control group （Group N）. Real-time polymerase chain reaction （RT-PCR） and Western blot techniques were used to observe the effects of LDP treatment on the cardiotrophin-like cytokine factor 1 （CLCF1）, ankyrin repeat and SOCS box containing 1 （ASB1）, and proldneticin 2 （PROK2） genes and the Janus kinase/signal transducer and activator of transcription （JAK/STAT） signaling pathway. Results： The mRNA （P〈0.05） and protein （P〈0.01） expression levels of the CLCF1 gone in Group A were significantly lower than the corresponding levels in Group N. After LDP treatment for 3 months, the mRNA expression levels of the CLCF1 gone were obviously up-regulated （P〈0.01）. After 6-month treatment, the expression levels of CLCF1 mRNA and protein were significantly up-regulated （both P〈0.01）, and the average bone density of the top femur had significantly increased （P〈0.05）. In vitro, CLCF1 overexpression resulted in a significant increase in the total protein and phosphorylated protein levels of JAK2 and STAT3. Conclusions： The CLCF1 gone is an important gone associated with PMOP Shen-yin deficiency and the therapeutic effects of LDP may be mediated by up-regulation of CLCF1 gone expression and activation of the JAK/STAT signaling pathway.展开更多

Embryos of KM strain exhibited 2-cell block, whereas B6C3F1 embryos, which are regarded as a nonblocking strain, proceeded to the 4-cell stage in our culture condition. It is often assumed that the block of early development is due to the failure of/ygotic gene activation (ZGA) in cultured embryos. In this study we examined protein synthesis patterns by two-dimensional gel eleetrophoresis of [35 S] methionine radiolabeled 2-cell embryos. Embryos from the blocking strain and the nonblocking strain were compared in their development both in vitro and in vivo. The detection of TRC expression, a marker of ZGA. at 42 h post hCG in KM embryos developed in vitro suggested that ZGA was also initiated even in the 2-cell arrested embryos. Nevertheless, a significant delay of ZGA was observed in KM strain as compared with normally developed B6C3F1 embryos. At the very beginning of major ZGA as early as 36 h post hCG, TRC has already been expressed in B6C3F1 embryos developed in vitro and KM embryos developed in vivo. But for 2-cell blocked KM embryos, TRC was still not detectable even at 38 h post hCG. These evidences suggest that 2-cell-blocked embryos do initiate ZGA, and that 2-cell block phenomenon is due not to the disability in initiating ZGA. but to a delay of ZGA.展开更多

A num- ber of different mechanisms of resistance have been reported, some of which are associated with plasmid-mediated acquisition of genes. Therefore, studies on plasmids in A. haumannii have been a hot issue lately. We have performed complete genome sequencing of A. haumannii MDR-TJ, which is a multidrug-resistant isolate. Finalizing the remaining large scaffold of the previous assem- bly, we found a new plasmid pABTJ2, which carries many phage-like elements. The plasmid pAB- TJ2 is a circular double-stranded DNA molecule, which is 110,967 bp in length. We annotated 125 CDSs from pABTJ2 using IMG ER and ZCURVE_V, accounting lbr 88.28% of the whole plasmid sequence. Many phage-like elements and a tRNA-coding gene were detected in pABTJ2, which is rarely reported among A. haumannii. The tRNA gene is specific for asparagine codon GTT, which may be a small chromosomal sequence picked up through incorrect excision during plasmid forma- tion. The phage-like elements may have been acquired during the integration process, as the GC content of the region carrying phage-like elements was higher than that of the adjacent regions. The finding of phage-like elements and tRNA-coding gene in pABTJ2 may provide a novel insight into the study of A. haumannii pan-plasmidome.展开更多

This study sought to evaluate the genotypes and allele frequencies of the -94 G/A polymorphism in the dopamine D1 receptor gone by real-time PCR using TaqMan fluorescent probes. One hundred and sixty-two patients with schizophrenia and 101 healthy controls living in Shandong province of China were evaluated. Experimental results showed that the G/A genotype distribution was significantly higher in the schizophrenia patients than in healthy controls. The frequencies of G allele and A allele were not significantly different between the schizophrenia patients and the controls. Thus, the -94 G/A polymorphism in the dopamine D1 receptor gone was found to be associated with schizophrenia in a Chinese Han population from Shandong province.展开更多

They could transmit a great variety of serious animal and human pathogens, which are great threats to the health of ...Hyalomma anatolicum, Hyalomma asiaticum and Hyalomma detritum are wide-spread tick vectors in China. They could transmit a great variety of serious animal and human pathogens, which are great threats to the health of human beings and the safety of stockbreeding. Most of them are distributed in Xinjiang and Inner Mongolia, and they share similar morphologies. This study is to establish a method for identifying H. anatolicum, H. asiaticum and H. detritum with molecular markers and to revealing the phylogenetic relationship of these ticks. Ticks were collected from domestic animals in Xinjiang and Inner Mongolia autonomous regions and classified by morphological characters. 16S rRNA and mitachondrial cytochrome oxidase subunit I gone （CO1） of ticks were amplified by PCR and subsequently sequenced. Phylogenetic trees were constructed by MEGA 5.0 and Mrbayes 3.2. On the 16S rRNA-based phylogenetic tree, H. anatolicum and H. aisaticum were clustered together with their respective classes. In a...This experiment sought to observe the migration and distribution of bone mesenchymal stem cells transfected with the cytosine deaminase gone （BMSCs-CD/eGFP） after transplantation in vivo through three pathways. In addition, we examined the tropism of these cells to glioma. Intracranial C6 glioma models were established in Sprague-Dawley rats using an intracranial stereotactic inoculation method. When tumors were 7 days old, rats were inoculated with lx106 BMSCs-CD/eGFP cells via the tumor-bearing internal carotid artery, the contralateral hemisphere and the tumor-bearing glioma. Fluorescence microscopy revealed that BMSCs-CD/eGFP exhibited a strong capacity for migration to tumors. BMSCs-CD/eGFP transplanted via the tumor-bearing intemal carotid artery were observed to distribute in glioma tissues. BMSCs-CD/eGFP inoculated via the ipsilateral glioma mainly located within and at the edge of glioma tissues. BMSCs-CD/eGFP inoculated via the contralateral hemisphere mainly distributed at the proximal end of the tumor at the incubation site.展开更多
