Tradit...Epstein-Barr virus (EBV) encoded latent membrane protein 1 (LMP1) is considered to be the major oncogenic protein of EBV encoded proteins, and also it has always been the core of the oncogenic mechanism of EBV. Traditional receptor theory demonstrates that cell surface receptors exert biological functions on the membrane, which neither enter into the nucleus nor directly affect the transcription of the target genes. But, advanced studies on nuclear transloca-tion of the epidermal growth factor receptor (EGFR) family have greatly developed our knowl-edge of the biological function of cell surface receptors. In this study, we used Tet-on LMP1 HNE2 cell line as a cell model, which is a dual-stable LMP1 integrated NPC cell line and the ex-pression of LMP1 in which could be regulated by Tet system. We found that LMP1 could regulate the nuclear translocation of EGFR in a dose-dependent manner from both quantitative and qualitative levels through the Western blot analysis and the immunofluorescent analysis with a laser scanning confocal microscope. We further demonstrated that the nuclear localization se-quence of EGFR played some roles in the location of the protein within the nucleus under LMP1 regulation, and the nuclear accumulation of EGFR regulated by LMP1 was in a ligand-independent manner. These findings provide a novel view that the regulation of LMP1 on the nuclear translocation of EGFR is critical for the process of nasopharyngeal carcinoma.展开更多

Infectious identification related to cancer may lead to better treatment for both the prevention and targeting of cancer therapy. Although nearly 20% of all cancers are caused by an infection of a microbe, the amount of evidence and information regarding the mechanisms associated with oncogenesis varies dramatically from one organism to the next. This review cannot be exhaustive because we are not aware of all infections worldwide in addition to their potential mechanisms for oncogenesis. More research is required for all of the species mentioned in this review.展开更多

Studies proved the role of EBV in tumor-genesis and linked it to prognosis and therapy of patients. Objectives: To determine the frequency of EBV in non-Hodgkin lymphomas using EBV latent membrane protein 1 (EBV-LMP1) immunohistochemical stain. Methods: This cross-sectional study was conducted at radio-isotope centre of Khartoum (2012-2014). A total of 75 cases of non-Hodgkin lymphoma paraffin embedded sections were stained for EBV LMP1 antibody. Data were analyzed by SPSS 16 and statistical cross linking of the results of immune staining with other data was done. Results: Out of 75 patients of non Hodgkin’s lymphoma (74.7%) were males. EBV-LMP1 Immune-staining was positive in (17.3%) with predominance of Burkitt’s lymphoma (33.3%), followed by diffuse large B cell lymphoma (17.9%). Conclusion: Burkitt’s lymphoma expressed the highest percentage of non-Hodgkin’s lymphoma positive cases (46.2%) out of the total (17.3%) positive cases. Different methods need to be used in studying Burkitt’s lymphoma expression of EBV and its effects on the treatment and prognosis of cases.展开更多
目的比较原发性鼻腔非霍奇金淋巴瘤(non-Hodgkin’s lymphomas,NHL)和慢性鼻-鼻窦炎中Epstein-Bar(EB)病毒缺失型潜伏膜蛋白1(latent membrane protein 1,LMP1)基因的表达,探讨原发性鼻腔非霍奇金淋巴瘤LMP1缺失型与预后的关系。方法采...目的比较原发性鼻腔非霍奇金淋巴瘤(non-Hodgkin’s lymphomas,NHL)和慢性鼻-鼻窦炎中Epstein-Bar(EB)病毒缺失型潜伏膜蛋白1(latent membrane protein 1,LMP1)基因的表达,探讨原发性鼻腔非霍奇金淋巴瘤LMP1缺失型与预后的关系。方法采用免疫组织化学方法检测55例原发性鼻腔NHL和25例慢性鼻-鼻窦炎LMP1,采用聚合酶链式反应(polymersechain reaction,PCR)检测LMP1基因,结合随访资料分析LMP1缺失型鼻腔NHL与预后的关系。结果鼻腔NHL中免疫组化LMP1蛋白阳性率为29/55(53%),LMP1基因阳性率为33/55(60%),其中缺失型LMP1基因者26/33(78.8%);慢性鼻-鼻窦炎中LMP1基因阳性率为14/25(56%),其中缺失型LMP1基因者5/14(35.7%)。鼻腔NHL患者和慢性鼻—鼻窦炎患者缺失型LMP1基因的表达差异有统计学意义(P<0.05)。LMP1基因阳性的鼻腔NHL患者5年总生存率为52.4%,其中LMP1缺失型和LMP1野生型鼻腔NHL的5年生存率分别为36.0%和85.7%,二者生存曲线差异有统计学意义,P值为0.04(log-rank检验)。结论提示缺失型LMP1在原发性鼻腔NHL的发病和病情进展中可能起重要作用。展开更多
文摘目的 研究非洲淋巴细胞瘤病毒LMP1(EBV LMP1)、巨细胞病毒pp65(CMVpp65)在肾脏疾病患者肾组织中的表达情况,探讨病毒感染与人类肾脏疾病发生、发展的关系.方法 临床资料齐全的肾穿刺活检标本435例,免疫组化法检测EBV LMP1和CMVpp65在肾组织中的表达情况.结果 (1)175例(40.2%)肾组织中EBV LMP1表达阳性,狼疮性肾炎(LN)组及IgA肾病(IgAN)组与非免疫性肾病组及原发性肾炎(PGN)组比较,差异有统计学意义;170例(39.1%)肾组织中CMVpp65表达阳性,LN组与非免疫性肾病组及PGN组比较,差异有统计学意义;(2)PGN组初发患者与复发患者肾组织EBV LMP1、CMVpp65阳性率比较差异有统计学意义,其他病种则无;(3)合并感染与无合并感染患者EBV LMP1、CMVpp65阳性率比较差异均无统计学意义;(4)肾组织EBV LMP1表达阳性的LN患者血清anti-Sm-Ab阳性率高于阴性组(P〈0.05,OR=6.00,95%CI:1.200~29.998);而CMVpp65表达阳性者血清anti-Sm-Ab阳性率低于阴性组(P〈0.01,OR=0.156,95%CI:0.038~0.643).结论 肾组织EBV及CMV感染可能参与LN及IgAN的发病,其中EBV感染可能以分子拟态方式诱发系统性红斑狼疮.关键词:爱泼斯坦-巴尔病毒;人类巨细胞病毒;肾脏疾病;肾组织分类号:R737.1 文献标识码:A文章编号:1001-7399(2011)03-0257-04EBV LMP1 and CMVpp65 in renal tissue of patients with renal diseasesYU Xiao-xia LIU Hua-feng CHEN Xiao-wen TAO Jing-li YANG Chen LI
