Andrographolide sulfonate ameliorates lipopolysaccharide-induced acute lung injury in mice by down-regulating MAPK and NF-κB pathways 被引量:39

Acute lung injury(ALI) or acute respiratory distress syndrome(ARDS) is a severe, lifethreatening medical condition characterized by widespread inflammation in the lungs, and is a significant source of morbidity and mortality in the patient population. New therapies for the treatment of ALI are desperately needed. In the present study, we examined the effect of andrographolide sulfonate, a water-soluble form of andrographolide(trade name: Xi-Yan-Ping Injection), on lipopolysaccharide(LPS)-induced ALI and inflammation. Andrographolide sulfonate was administered by intraperitoneal injection to mice with LPS-induced ALI. LPS-induced airway inflammatory cell recruitment and lung histological alterations were significantly ameliorated by andrographolide sulfonate. Protein levels of pro-inflammatory cytokines in bronchoalveolar lavage fluid(BALF) and serum were reduced by andrographolide sulfonate administration. m RNA levels of pro-inflammatory cytokines in lung tissue were also suppressed. Moreover, andrographolide sulfonate markedly suppressed the activation of mitogen-activated protein kinase(MAPK) as well as p65 subunit of nuclear factor-κB(NF-κB). In summary, these results suggest that andrographolide sulfonate ameliorated LPS-induced ALI in mice by inhibiting NF-κB and MAPK-mediated inflammatory responses. Our study shows that water-soluble andrographolide sulfonate may represent a new therapeutic approach for treating inflammatory lung disorders. 展开更多

Anionic surfactant sodium dodecyl benzene sulfonate(SDBS)at varying concentrations was selected to investigate the influence on the wettability of Zhaozhuang Coal by molecular dynamics simulation.Six groups of water/surfactant/coal systems with different concentrations were constructed.The influence of surfactant with different concentrations on the wettability of coal was concluded by analyzing various properties from the energetic behaviors to the dynamic characteristics.The results show that the interfacial tension decreases sharply and then rises slowly with the increase of SDBS surfactant concentration,obtaining that surfactants can obviously reduce the interfacial tension.The surfactant molecules could be detected at the water/coal interface through analyzing the system’s relative concentration distribution.In addition,the difference in the wettability of surfactants on coal surfaces is caused by the spatial distribution differences of alkyl chains and the benzene ring of the surfactant molecules.And the negative interaction energy between SDBS and the coal surface indicates that adsorption process is spontaneous.Furthermore,it is of great practical significance for improving the dust reduction effect and reducing the disaster of coal dust by exploring the effects of surfactant molecules on the wettability of coal. 展开更多

Methods: Randomized contro... Objective: To systematically evaluate the effectiveness and safety of Sodium Tanshinone ⅡA Sulfonate Injection(STS) as one adjuvant therapy for treating unstable angina pectoris(UAP). Methods: Randomized controlled trials(RCTs) of UAP treated by STS were searched in the China National Knowledge Infrastructure Database(CNKI), VIP Database for Chinese Technical Periodicals(VIP), Wanfang Database, the Chinese Biomedical Literature Database(CBM), Web of Science, the Cochrane Library, Embase, and Pub Med, which from inception to January, 2016. The Cochrane Risk Assessment Tool was used to evaluate the methodological quality of the RCTs. The Review Manager 5.3 software was used to conduct the metaanalysis. Results: The results showed that 17 RCTs involving 1,372 patients were included. The meta-analysis indicated that the combined use of STS and Western medicine(WM) in the treatment of UAP can obviously improve the total effective rate [risk ratio(RR)=1.31, 95% confidence interval(CI)(1.24,1.39), P〈0.0001], and the total effective rate of electrocardiogram [RR=1.43, 95% CI(1.30,1.56), P〈0.0001], decrease the level of CRP [mean difference(MD)=–3.06, 95%CI(–3.85, –2.27), P〈0.00001], fibrinogen [MD=–1.03, 95% CI(–1.16, –0.89), P〈0.00001], and whole blood high shear viscosity [MD=–0.70, 95% CI(–0.92, –0.49), P〈0.00001]. Additionally, the occurrence of adverse drug reaction of the experimental group was significantly higher than that of the control group [RR=3.57, 95% CI(1.28, 9.94), P〈0.05]. Ecological Behavior of Linear Alkylbenzene Sulfonate (LAS) in Soil-Plant Systems 被引量:17

More and more linear alkylbenzene sulfonate (LAS) has contaminated the water and soil ma pollution discharge, making it important to identify the ecological behavior and toxicity of LAS so as to carry out measures that will reduce its negative effects on the ecosystem. The ecological behavior of LAS. including degradation, migration, and plant uptake, in both soil-paddy rice and soil-soybean systems was studied. Reduction of LAS in pot and field plots followed the first order reaction kinetics with degradation half-lives of 35-50 days with LAS decreasing to very low concentrations after a season of crop growth. Strong migration ability for LAS was found and the breakthrough time in a 1.5 in soil monolith was significantly shortened to 23 days by preferential flow. Leachate volumes of soil-paddy and soil-soybean systems at preferential breakthrough were much different, while the leachate volumes at equilibrium governed by soil adsorption/desorption processes were very similar. Significant uptake of LAS in both paddy rice and sovbeans was observed in pot and field experiments (P < 0.05). In aquatic culture, 20 μg mL-1 and above of LAS significantly inhibited the growth of paddy seedlings (P < 0.05). The critical concentration for LAS in soil inhibiting the growth and yield of paddy was 160 μg g-1; when higher, there was a strong negative influence, with decreases in height, spike length, and production, when lower than 80 μg g-1, paddy growth was stimulated. There was little effect of LAS on soybeans. 展开更多

Adsorption experiments were carried out to investigate the sorption behaviors of naphthalene and phenanthrene in six different soils and to determine the effects of temperature, linear alkylbenzene sulfonate (LAS) and cetylrimethyl ammonium bromide (CTAB) on sorption. The results show that for a given sorbent phenanthrene exhibited greater nonlinear and stronger sorption than naphthalene. There was a strong negative correlation for the Koc values with organic carbon content (foc). The increase of temperature was not favorable to sorption. Sorption decreased along with the increasing aqueous LAS concentration from 0 to 1000 mg/L. At low CTAB concentration (〈 100 mg/L), the adsorption increased as CTAB hemimicelles formed on the soil surface. At high concentration, CTAB decreased the adsorption by occupying active hydrophobic adsorption sites and solubilization of naphthalene and phenanthrene. 展开更多

Cardiac dysfunction, a common consequence of sepsis, is the major contribution to morbidity and mortality in patients. Sodium tanshinone IIA sulfonate(STS) is a water-soluble derivative of Tanshinone IIA(TA), a main active component of Salvia miltiorrhiza Bunge, which has been widely used in China for the treatment of cardiovascular and cerebral system diseases. In the present study, the effect of STS on sepsis-induced cardiac dysfunction was investigated and its effect on survival rate of rats with sepsis was also evaluated. STS treatment could significantly decrease the serum levels of C-reactive protein(CRP), procalcitonin(PCT), cardiac troponin Ⅰ(cTn-Ⅰ), cardiac troponin T(cTn-T), and brain natriuretic peptide(BNP) in cecal ligation and puncture(CLP)-induced) septic rats and improve left ventricular function, particularly at 48 and 72 h after CLP. As the pathogenesis of septic myocardial dysfunction is attributable to dysregulated systemic inflammatory responses, several key cytokines, including tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β), interleukin-6(IL-6), interleukin-10(IL-10) and high mobility group protein B1(HMGB1), were detected to reveal the possible mechanism of attenuation of septic myocardial dysfunction after being treated by STS. Our study showed that STS, especially at a high dose(15 mg×kg–1), could efficiently suppress inflammatory responses in myocardium and reduce myocardial necrosis through markedly reducing production of myocardial TNF-α, IL-6 and HMGB1. STS significantly improved the 18-day survival rate of rats with sepsis from 0% to 30%(P < 0.05). Therefore, STS could suppress inflammatory responses and improve left ventricular function in rats with sepsis, suggesting that it may be developed for the treatment of sepsis. 展开更多

The sorption capacity was doubled when the temperature increased from 5 to 35°C, and thermodynamics parameters △G0 was calculated to be –7.11 to –5.04 kJ/mol, △H0 was 14.2 kJ/mol, and △S 0 was 69.5 J/(mol·K), indicating that the sorption was a spontaneous, endothermic, and entropy driven process. Desorption hysteresis occurred at all studied temperatures which suggested that humic acid may be an important sink of PFOS in the environment. 展开更多

This study developed a population pharmacokinetic model for sodium tanshinone IIA sulfonate(STS) in healthy volunteers and coronary heart disease(CHD) patients in order to identify significant covariates for the pharmacokinetics of STS. Blood samples were obtained by intense sampling approach from 10 healthy volunteers and sparse sampling from 25 CHD patients, and a population pharmacokinetic analysis was performed by nonlinear mixed-effect modeling. The final model was evaluated by bootstrap and visual predictive check. A total of 230 plasma concentrations were included, 137 from healthy volunteers and 93 from CHD patients. It was a two-compartment model with first-order elimination. The typical value of the apparent clearance(CL) of STS in CHD patients with total bilirubin(TBIL) level of 10 μmol×L^(–1) was 48.7 L×h^(–1) with inter individual variability of 27.4%, whereas that in healthy volunteers with the same TBIL level was 63.1 L×h^(–1). Residual variability was described by a proportional error model and estimated at 5.2%. The CL of STS in CHD patients was lower than that in healthy volunteers and decreased when TBIL levels increased. The bootstrap and visual predictive check confirmed the stability and validity of the final model. These results suggested that STS dosage adjustment might be considered based on TBIL levels in CHD patients. 展开更多

Andrographolide total ester sulfonate(ATES) injection is one of the products of traditional Chinese medicine(TCM) currently used against viral infection in China.ATES injection was approved for manufacturing and marketing in January 2002.It is indicated for acute respiratory infections,tonsillitis,chronic obstructive pulmonary disease,influenza,foot and mouth disease,bronchiolitis,herpangina,mumps,infectious mononucleosis and psychosis.However,its usage also carries risk.We investigated the use of ATES at the Wuhan Union Hospital from January 2014 to December 2014 and evaluated its real-world clinical application using the hospital centralized monitoring method.A total of 848 cases were enrolled in this study.In these cases,it was mainly used for postoperative anti-inflammation and treating upper respiratory infection,pneumonia and bronchitis.Among them,39.86% were contraindicated.Irregular medication of adults and children accounted for 1.91% and 23.38%,respectively.Improper choice of solvent accounted for 3.18%.The choice of intravenous drip versus aerosol inhalation was reasonable.A case of adverse events(AEs) was observed in the monitoring period,and the incidence of adverse drug reaction(ADR) of ATES injection was 0.12%.ATES injection in our hospital is relatively safe with a low incidence of adverse reactions.The study assesses the clinical usage and adverse reactions of ATES injection,and provides suggestions for rational use in clinical practice. 展开更多

Aim: To quantitatively study the histological changes of the testis and epididymis as a result of a drastic reduction of testosterone secretion. Methods: Fourteen adult Sprague-Dawley rats were injected intraperitoneally with ethane dimethane sulfonate (EDS, 75 mg/kg) and the same number of animals were injected with normal saline as a control. At days 7 and 12 (after treatment), respectively, half of the animals from each group were killed. The testes and epididymides were removed and tissue blocks embedded in methacrylate resin. The cell number per testis was estimated using the stereological optical disector and some other parameters were obtained using other morphometric methods. Results: The EDS treatment resulted in an almost complete elimination of Leydig cells but had no effect on the numbers of Sertoli cells per testis. At day 7 after EDS treatment, many elongated spermatids were retained in the seminiferous epithelium and many round spermatids could be seen in the epididymal ducts. At day 12, a looser arrangement of spermatids and spermatocytes became evident, with apparent narrow empty spaces being formed between germ cells in an approximately radial direction towards the tubule lumen; the numbers (per testis) of non-type B spermatogonia and spermatocytes were similar to controls, whereas that of type B spermatogonia increased by 59%, and that of early round, elongating and late elongated spermatids decreased by 37%, 72% and 52%, respectively. Conclusion: The primary spermatogenic lesions following EDS administration were (i) spermiation failure and (ii) detachment of spermatids and spermatocytes associated with impairment in spermiogenesis and meiosis. 展开更多
