Helicobacter pylori(H.pylori)is a gram-negative bacterium that infects approximately 4.4 billion individuals worldwide.However,its prevalence varies among different geographic areas,and is influenced by several factors.The infection can be acquired by means of oral-oral or fecal-oral transmission,and the pathogen possesses various mechanisms that improve its capacity of mobility,adherence and manipulation of the gastric microenvironment,making possible the colonization of an organ with a highly acidic lumen.In addition,H.pylori presents a large variety of virulence factors that improve its pathogenicity,of which we highlight cytotoxin associated antigen A,vacuolating cytotoxin,duodenal ulcer promoting gene A protein,outer inflammatory protein and gamma-glutamyl transpeptidase.The host immune system,mainly by means of a Th1-polarized response,also plays a crucial role in the infection course.Although most H.pylori-positive individuals remain asymptomatic,the infection predisposes the development of various clinical conditions as peptic ulcers,gastric adenocarcinomas and mucosa-associated lymphoid tissue lymphomas.Invasive and non-invasive diagnostic methods,each of them with their related advantages and limitations,have been applied in H.pylori detection.Moreover,bacterial resistance to antimicrobial therapy is a major challenge in the treatment of this infection,and new therapy alternatives are being tested to improve H.pylori eradication.Last but not least,the development of effective vaccines against H.pylori infection have been the aim of several research studies. 展开更多

干旱、高盐及低温等非生物胁迫是限制植物生长发育的主要环境因子。这些环境胁迫因子通常导致植物体内生理代谢改变,并参与非生物胁迫调控转录因子的差异表达。植物抵御上述非生物逆境的能力与转录因子调控逆境相关功能基因的表达密不... 干旱、高盐及低温等非生物胁迫是限制植物生长发育的主要环境因子。这些环境胁迫因子通常导致植物体内生理代谢改变,并参与非生物胁迫调控转录因子的差异表达。植物抵御上述非生物逆境的能力与转录因子调控逆境相关功能基因的表达密不可分。近年来,发掘植物非生物胁迫相关转录因子的功能及揭示转录因子介导植物非生物胁迫响应的调控机制,已成为植物营养分子生物学关注的热点之一。因此,了解植物非生物胁迫下的生理应答及转录因子参与的调控机制,对建立植物适应性改良途径具有重要科学意义。本文从干旱、高盐和低温三方面阐述了非生物胁迫下植物生理生化的适应性变化,概述了MYB、b ZIP、AP2/EREBP、WRKY和NAC五类与植物抗逆相关的转录因子的结构与功能特征,着重论述了转录因子介导植物抵御非生物胁迫的分子调控机制。植物遭遇非生物胁迫时,通常表现为生长速率、叶面积和叶片数量下降,蒸腾及光合速率降低。同时,植物体内活性氧逐渐累积,使细胞膜脂过氧化程度加剧,造成细胞损伤。为适应不利环境,在生理上植物表现为体内抗氧化酶活性增强,渗透调节物数量增多;在分子水平上,植物对非生物胁迫适应性的增强,通常与转录因子识别抗逆基因启动子特异性元件及调控逆境防御基因的转录有关。本文对于深入阐明干旱、高盐及低温胁迫下植物生理生化应答与转录因子的分子调控机制提供了全新的科学启示。 展开更多

AIM: The existence and properties of alpha-fetoprotein (AFP) receptor on the surface of NIH 3T3 cells and the effects of AFP on cellular signal transduction pathway were investigated. METHODS: The effect of AFP on the proliferation of NIH 3T3 cells was measured by incorporation of 3H-TdR. Receptor-binding assay of 125I-AFP was performed to detect the properties of AFP receptor in NIH 3T3 cells. The influences of AFP on the [cAMP]i and the activities of protein kinase A (PKA) were determined. Western blot was used to detect the change of K-ras P21 protein expression. RESULTS: The proliferation of NIH 3T3 cells treated with 0-80 mg/L of AFP was significantly enhanced. The Scatchard analysis indicated that there were two classes of binding sites with KD of 2.722 x 10(-9)M (Bmax=12810 sites per cell) and 8.931 x 10(-8)M (Bmax=119700 sites per cell) respectively. In the presence of AFP (20 mg/L), the content of cAMP and activities of PKA were significantly elevated . The level of K-ras P21 protein was upregulated by AFP at the concentration of 20 mg/L. The monoclonal antibody against AFP could reverse the effects of AFP on the cAMP content, PKA activity and the expression of K-ras p21 gene. CONCLUSION: The effect of AFP on the cell proliferation was achieved by binding its receptor to trigger the signal transduction pathway of cAMP-PKA and alter the expression of K- ras p21 gene. 展开更多

目的:探究血府逐瘀汤联合常规治疗对急性脑梗死患者炎症反应、氧化应激、内皮及相关因子的影响。方法:将本院收治的162例急性脑梗死患者随机分为对照组及观察组各81例。对照组采用降颅内压、抗凝、抗氧化及调脂等常规对症治疗,观察组在... 目的:探究血府逐瘀汤联合常规治疗对急性脑梗死患者炎症反应、氧化应激、内皮及相关因子的影响。方法:将本院收治的162例急性脑梗死患者随机分为对照组及观察组各81例。对照组采用降颅内压、抗凝、抗氧化及调脂等常规对症治疗,观察组在对照组基础上联合血府逐瘀汤治疗。比较分析两组炎症反应、氧化应激、内皮及相关因子的变化。结果:治疗前,两组肿瘤坏死因子-α(TNF-α)、C反应蛋白(CRP)、超氧化物歧化酶(SOD)、丙二醛(MDA)、晚期蛋白氧化产物(AOPPS)、内皮素-1(ET-1)、一氧化氮(NO)、基质金属蛋白酶(MMP-9)、血小板活化因子(PAF)、胰岛素样生长因子-1(IGF-1)水平比较,差异无统计学意义(P>0.05)。治疗后,两组TNF-α、CRP明显低于治疗前(P<0.05),且观察组TNF-α、CRP水平较对照组显著降低(P<0.05);两组MDA、AOPPS显著低于治疗前,而SOD显著高于治疗前(P<0.05),且观察组MDA、AOPPS显著低于对照组,而SOD明显高于对照组(P<0.05);两组ET-1明显低于治疗前,而NO明显高于治疗前(P<0.05),且观察组ET-1明显低于对照组,而NO明显高于对照组(P<0.05);两组MMP-9、PAF明显低于治疗前,而IGF-1显著高于治疗前(P<0.05),且观察组MMP-9、PAF明显低于对照组,而IGF-1明显高于对照组(P<0.05)。结论:血府逐瘀汤联合常规治疗急性脑梗死,可有效降低患者炎症及氧化应激反应,改善其血管内皮功能及神经功能,显著减轻脑损伤程度,具有临床推广意义。 展开更多

目的:探讨超声气压弹道和钬激光技术用于经皮肾镜下碎石清石治疗复杂性肾结石的疗效及血清学指标差异。方法:收集在本院进行手术治疗的复杂性肾结石患者78例,回顾其手术方式及检查结果,分为接受超声气压弹道术治疗的观察组34例、接受钬... 目的:探讨超声气压弹道和钬激光技术用于经皮肾镜下碎石清石治疗复杂性肾结石的疗效及血清学指标差异。方法:收集在本院进行手术治疗的复杂性肾结石患者78例,回顾其手术方式及检查结果,分为接受超声气压弹道术治疗的观察组34例、接受钬激光治疗的对照组44例。记录两组的Ⅰ期结石取净率;采集血清中并采用肌氨酸氧化酶法检测血清肾功能指标,采用ELISA法检测炎症因子、应激激素含量。结果:观察组的平均取石时间短于对照组(P<0.05);两组Ⅰ期结石取净率比较,差异无统计学意义(P>0.05)。术后3d,观察组血清肾功能指标Scr、BUN、CysC水平低于对照组(P<0.05);观察组血清炎症因子PCT、IL-1β、IL-22、IL-13含量低于对照组,IL-4含量高于对照组(P<0.05);观察组血清中应激激素AngⅠ、AngⅡ、Adr、NE含量低于对照组(P<0.05)。结论:超声气压弹道和钬激光技术均可有效清除复杂性肾结石且超声气压弹道技术具有操作快、创伤小的优势。 展开更多

AIM: The transcription factor EGR-1 (early growth response gene-1) plays an important role in cell growth, differentiation and development. It has identified that EGR-1 has significant transformation suppression activity in some neoplasms, such as fibrosarcoma, breast carcinoma. This experiment was designed to investigate the role of egr-1 in the cancerous process of hepatocellular carcinoma (HCC) and esophageal carcinoma (EC), and then to appraise the effects of EGR-1 on the growth of these tumor cells. METHODS: Firstly, the transcription and expression of egr-1 in HCC and EC, paracancerous tissues and their normal counterpart parts were detected by in situ hybridization and immunohistochemistry, with normal human breast and mouse brain tissues as positive controls. Egr-1 gene was then transfected into HCC (HHCC, SMMC7721) and EC (ECa109) cell lines in which no egr-1 transcription and expression were present. The cell growth speed, FCM cell cycle, plate clone formation and tumorigenicity in nude mice were observed and the controls were the cell lines transfected with vector only. RESULTS: Little or no egr-1 transcription and expression were detected in HCC, EC and normal liver tissues. The expression of egr-1 were found higher in hepatocellular paracancerous tissue (transcription level P=0.000; expression level P=0.143, probably because fewer in number of cases) and dysplastic tissue of esophageal cancer (transcription level P=0.000; expression level P=0.001). The growth rate of egr-1-transfected HHCC (HCC cell line) cells and ECa109 (EC cell line) cells was much slower than that of the controls. The proportion of S phase cell, clone formation and tumorigenicity were significantly lower than these of the controls' (decreased 45.5% in HHCC cells and 34.1% in ECa109 cells; 46.6% and 41.8%; 80.4% and 72.6% respectively). There were no obvious differences between SMMC7721 (HCC) egr-1-transfected cells and the controls with regard to the above items. To investigate whether the expression of exogenous heme oxygenase-1 (HO-1) gene within vascular smooth muscle cells (VSMC) could protect the cells from free radical attack and inhibit cell proliferation, we establishe... To investigate whether the expression of exogenous heme oxygenase-1 (HO-1) gene within vascular smooth muscle cells (VSMC) could protect the cells from free radical attack and inhibit cell proliferation, we established an in vitro transfection of human HO-1 gene into rat VSMC mediated by a retroviral vector. The results showed that the profound expression of HO-1 protein as well as HO activity was 1.8- and 2.0-fold increased respectively in the transfected cells compared to the non-transfected ones. The treatment of VSMC with different concentrations of H2O2 led to the remarkable cell damage as indicated by survival rate and LDH leakage. However, the resistance of the HO-1 transfected VSMC against H2O2 was significantly raised. This protective effect was dramatically diminished when the transfected VSMC were pretreated with ZnPP-IX, a specific inhibitor of HO, for 24 h. In addition, we found that the growth potential of the transfected cells was significantly inhibited directly by increased activity of HO-1, and this effect might be related to decreased phosphorylation of MAPK. These results suggest that the overexpression of introduced hHO-1 is potentially able to reduce the risk factors of atherosclerosis, partially due to its cellular protection against oxidative injury and to its inhibitory effect on cellular proliferation. 展开更多

在南北样带温带区选择11个栎属树种的核心分布区,对各区栎属建群树种的功能性状(叶片干物质含量、比叶重、叶片养分含量、叶片光合速率以及种子性状)进行了测定,并对功能性状与气象因子的关系进行了分析。结果表明:首先,在南北样带温带... 在南北样带温带区选择11个栎属树种的核心分布区,对各区栎属建群树种的功能性状(叶片干物质含量、比叶重、叶片养分含量、叶片光合速率以及种子性状)进行了测定,并对功能性状与气象因子的关系进行了分析。结果表明:首先,在南北样带温带区,当外界环境变化时,栎属建群树种的功能性状也会随之变化。由南向北随着温度的下降,年均日照时数增加,栎属树种的生长季缩短,而基于叶片面积和重量的光合能力(Aarea、Amass)增加,这使得它们在有限时间内提高光合生产来维持其自身的生理活动。同时,种子去皮干重、种皮干重均随着温度的降低、年日照时数的增加而增加。当降水条件变化时,栎属树种还会通过调节其叶片干物质含量、比叶重来适应其生境。其次,功能性状间也存在一定的相关关系;此外,随着研究尺度的变化(种间尺度至种内尺度),功能性状与气象因子间关系也随之变化。 展开更多

AIM: To observe the growth suppression effect of exogenous introduction of early growth response gene-1 (Egr-1 gene) on esophageal carcinoma tissue as well as on esophageal carcinoma cell line Eca109 and to explore the potential application of Egr-1 gene in gene therapy of tumor. METHODS: Eukaryotic expression vector of PCMV-Egr-1 plasmid was introduced into Eca109 cell line which expressed no Egr-1 protein originally with lipofectamine transfection method. The introduction and expression of PCMV-Egr-1 plasmid into Eca109 cell line was confirmed by G418 selection culture, PCR amplification of neogene contained in the vector, Western blot analysis and immunocytochemical analysis. The cell growth curve, soft agar colony formation rate and tumorigenicity in SCID mice were examined to demonstrate the growth suppression effect of exogenous Egr-1 gene on Eca109 cell line. The Egr-1 mRNA and Egr-1 protein were also detected in 50 surgical specimens of esophageal carcinoma by in situ hybridization and immunohistochemistry. RESULTS: Exogenous Egr-1 gene was introduced successfully into Eca109 cell line and expressed Egr-1 protein stably. The transfected Eca109 cell line grew more slowly than control Eca109 as shown by cell growth curves, the soft agar colony formation rate (4.0% vs 6.9%, P 【 0.01) and the average growth rate of tumor in SCID mice (35.5 +/- 7.6 vs 65.8 +/- 7.6, P 【 0.05). The expression level of Egr-1 mRNA and protein significantly increased in dysplastic epithelia adjacent to cancer rather than in cancer tissues (65.8% vs 20.0% by ISH and 57.9% vs 0.01). CONCLUSION: Exogenous Egr-1 gene shows the strong effect of growth inhibition in Eca109 cell line. Egr-1 in the cancer tissue shows down-regulated expression that supports the inhibited function of Egr-1 in cancer growth and suggests Egr-1 may have an important role in gene therapy of esophageal carcinoma. 展开更多

Helicobacter pylori(H. pylori) infection is the most common bacterial infection worldwide. Persistent infection of the gastric mucosa leads to inflammatory processes and may remain silent for decades or progress causing more severe diseases, such as gastric adenocarcinoma. The clinical consequences of H. pylori infection are determined by multiple factors, including host genetic predisposition, gene regulation, environmental factors and heterogeneity of H. pylori virulence factors. After decades of studies of this successful relationship between pathogen and human host, various mechanisms have been elucidated. In this review, we have made an introduction on H. pylori infection and its virulence factors, and focused mainly on modulation of host immune response triggered by bacteria, changes in the pattern of gene expression in H. pylori-infected gastric mucosa, with activation of gene transcription involved in defense mechanisms, inflammatory and immunological response, cell proliferation and apoptosis. We also highlighted the role of bacteria eradication on gene expression levels. In addition, we addressed the recent involvement of different microRNAs in precancerous lesions, gastric cancer, and inflammatory processes induced by bacteria. New discoveries in this field may allow a better understanding of the role of major factors involved in the pathogenic mechanisms of H. pylori. 展开更多

针对农药雾滴难以穿透果树冠层,雾滴在叶背面附着率低等问题,提出风送喷雾与静电喷雾技术相结合的方法,为阐明各因素对喷雾覆盖率影响的显著性。采用4因素3水平的中心组合设计与响应面分析方法,研究感应电压、风机频率、喷雾距离和喷雾... 针对农药雾滴难以穿透果树冠层,雾滴在叶背面附着率低等问题,提出风送喷雾与静电喷雾技术相结合的方法,为阐明各因素对喷雾覆盖率影响的显著性。采用4因素3水平的中心组合设计与响应面分析方法,研究感应电压、风机频率、喷雾距离和喷雾压力对雾滴覆盖率的影响,以正面、反面雾滴覆盖率及其比值为响应值分别建立了二次回归模型。模型的决定系数分别为95.14%、93.68%、93.95%,相对误差分别小于5%、10%和15%。各因素对背正面覆盖率比影响的显著性排序为:充电电压>风机频率>喷雾距离>喷雾压力;对反面覆盖率影响的显著性排序为:充电电压>喷雾距离>喷雾压力>风机频率;对正面覆盖率影响的显著性排序为:喷雾距离>喷雾压力>风机频率>充电电压;荷电雾滴主要受气流曳力和静电力作用,风机频率、感应电压、喷雾距离之间的交互作用对响应面模型有显著性影响,因此合理匹配工作参数有利于提高反面的雾滴覆盖率。该研究为风送静电喷雾机设计与工作参数选择提供依据。 展开更多

目的调查胃镜检查中的应激反应,探讨可能的影响因素及预防对策。方法对200例胃镜检查者监测检查过程心血管反应及生理反应,观察应激反应与性别、年龄、基础疾病、插管反应、操作水平、检查时间、次数之间的关系。结果心血管反应发生率62... 目的调查胃镜检查中的应激反应,探讨可能的影响因素及预防对策。方法对200例胃镜检查者监测检查过程心血管反应及生理反应,观察应激反应与性别、年龄、基础疾病、插管反应、操作水平、检查时间、次数之间的关系。结果心血管反应发生率62.0%,收缩压与舒张压增高12.5%、14.6%,心率加快16.0%;出现缺血性ST-T及T波改变、心律失常18例;生理反应发生率为60.5%。影响因素:女性、年龄大、伴基础疾病、插管反应明显、检查时间长、内镜医师操作水平低等,与检查次数的多少无关。结论胃镜检查中,可发生不同程度的血液动力学变化及生理反应;提高内镜医生操作水平,缩短检查时间;同时对存在危险因素者进行心理干预、施行心电监护,进行无痛检查或应用恩丹西酮,可减轻应激反应。 展开更多

古近纪气候演化是由"温室气候"向"冰室气候"转变的过程,大致可划分为3个阶段:古新世-始新世早期的升温期,始新世中期-始新世末期的稳定降温期及渐新世-中新世早期的冰室气候期。在此期间发生了3次显著的气候事件,... 古近纪气候演化是由"温室气候"向"冰室气候"转变的过程,大致可划分为3个阶段:古新世-始新世早期的升温期,始新世中期-始新世末期的稳定降温期及渐新世-中新世早期的冰室气候期。在此期间发生了3次显著的气候事件,分别为PETM极热事件、Oi-1骤冷事件和Mi-1降温事件。这3次大的气候事件引起了生物圈的巨大波动:PETM期间发生了全球底栖大有孔虫灭绝事件及浮游有孔虫的辐射演化,并且是哺乳动物演化的关键时期;Oi-1期间全球植物群落由热带型密集雨林向寒冷干燥的森林草原过渡,在欧亚大陆出现了哺乳动物种群大规模改变的Grande Coupure和Mongolian Remodelling事件,也发生热水型浮游及底栖有孔虫的灭绝事件;Mi-1前后,浮游有孔虫部分属种绝灭,钙质超微化石也出现暖水种数量的急剧减少,现代植物群落在这一时期开始形成。古近纪气候的大幅波动和转折由诸多因素控制,主流观点认为PETM事件与天然气水合物的大量释放有关,但其喷发机制尚不明确;降温事件则与区域性构造隆升、洋流格局变化等因素有关。 展开更多

目的分析慢性心力衰竭患者心脏再同步化治疗(cardiac resynchronization therapy,CRT)无应答的危险因素。方法回顾性分析行CRT治疗的慢性心力衰竭患者109例,73例术后6个月左心室收缩末期容积增加≥10%为应答组,36例左心室收缩末期容积增... 目的分析慢性心力衰竭患者心脏再同步化治疗(cardiac resynchronization therapy,CRT)无应答的危险因素。方法回顾性分析行CRT治疗的慢性心力衰竭患者109例,73例术后6个月左心室收缩末期容积增加≥10%为应答组,36例左心室收缩末期容积增加<10%为无应答组,收集患者术前一般临床资料、常规超声参数、同步化参数及左心室起搏电极位置等指标,采用Logistic回归分析影响CRT无应答的危险因素。结果 2组应答率为66.97%;Logistic回归分析结果显示,心功能差、缺血性心肌病、合并室性心动过速、左心室收缩末期容积增大为CRT无应答的危险因素(P<0.01);心室间延迟、合并左束支传导阻滞为CRT无应答的保护因素(P<0.01)。结论心室间延迟、合并左束支传导阻滞可提高CRT应答率,心功能差、缺血性心肌病、合并室性心动过速、较大的左心室收缩末期容积是无应答的危险因素。 展开更多
