In this report, a novel strategy called step- wise multiple ion monitoring-enhanced product ions (stepwise MIM-EPI) was developed to construct the MS2T library, in which stepwise MIM was used as survey scans to trigger the acquisition of EPI. A total number of 698 (almost) non- redundant metabolites with MS2 spectra were obtained, of which 135 metabolites were identified/annotated. Integrating the data gathered from our MS2T library and other available multiple reaction monitoring (MRM) information, a widely targeted metabolomics method was developed to quantify 277 metabolites, including some phytohormones. Evaluation of the dehydration responses and natural variations of these metabolites in rice leaf not only suggested the coordinated regulation of abscisic acid (ABA) with metabolites such as serotonin derivative(s), polyamine conjugates under drought stress, but also revealed some C-glycosylated flavones as the potential markers for the discrimination of indica and japonica rice subspecies. The new MS2T library construction and widely targeted metabolomics strategy could be used as a tool for rice functional genomics.展开更多

METHODS: Using microarra...AIM: To investigate alternative or subordinate pathways involved in colorectal tumorigenesis and tumor growth, possibly determining at-risk populations and predicting responses to treatment. METHODS: Using microarray gene-expression analysis, we analyzed patterns of gene expression relative to canonical molecular changes and clinicopathological features in 84 sporadic colorectal cancer patients, standardized by tumor location. Subsets of differentially expressed genes were confirmed by real-time reverse-transcript polymerase chain reaction (RT-PCR). RESULTS: The largest number of genes identified as being differentially expressed was by tumor location, and the next largest number by lymphovascular or neural invasion of tumor cells and by mismatch repair (NMR) defects. Amongst biological processes, the immune response was significantly implicated in entire molecular changes observed during colorectal tumorigenesis (P 〈 0.001). Amongst 47 differentially expressed genes, seven (PISD, NIBP, BAI2, STOML1, MRPL21, MRPL16, and MKKS) were newly found to correlate with tumorigenesis and tumor growth. Most location-associated molecular changes had distinct effects on gene expression, but the effects of the latter were sometimes contradictory. CONCLUSION: We show that several differentially expressed genes were associated with canonical molecular changes in sporadic colorectal cancers, possibly constituting alternative or subordinate pathways of tumorigenesis. The upper lithosphere is the crust, having a thickness of 32-34km. The middle lithosphere is horizontal layering. 25-40 km thick. The lower lithosphere juxtaposed steeply declined high velocity and low velocity rock series, which might be split open under extension. Hot mantle flow upwelled to the level of the Moho then turned to horizontal direction and formed middle lithosphere. The shape of these kinds of lithosphere is somewhat like mushroom cloud in nuclear explosion. There is no causality between crustal structure of the Qinling orogen and its upper mantle structure nowadays. The Qinling orogen might be an intracontinental orogen of Indosinian-Yanshanian period, formed by wedging of the Yangtze crust into the middle crust of North China craton. and by reformation of extension in E-W direction in postorogenic period.展开更多

After the passage ...In situ observations from Argo profiling floats combined with satellite retrieved SST and rain rate are used to investigate an upper ocean response to Typhoon Bolaven from 20 through 29 August 2012. After the passage of Typhoon Bolaven, the deepening of mixed layer depth(MLD), and the cooling of mixed layer temperature(MLT) were observed. The changes in mixed layer salinity(MLS) showed an equivalent number of increasing and decreasing because the typhoon-induced salinity changes in the mixed layer were influenced by precipitation, evaporation, turbulent mixing and upwelling of thermocline water. The deepening of the MLD and the cooling of the MLT indicated a significant rightward bias, whereas the MLS was freshened to the left side of the typhoon track and increased on the other side. Intensive temperature and salinity profiles observed by Iridium floats make it possible to view response processes in the upper ocean after the passage of a typhoon. The cooling in the near-surface and the warming in the subsurface were observed by two Iridium floats located to the left side of the cyclonic track during the development stage of the storm, beyond the radius of maximum winds relative to the typhoon center. Water salinity increases at the base of the mixed layer and the top of the thermocline were the most obvious change observed by those two floats. On the right side of the track and near the typhoon center when the typhoon was intensified, the significant cooling from sea surface to a depth of 200×10^4 Pa, with the exception of the water at the top of the thermocline, was observed by the other Iridium float. Owing to the enhanced upwelling near the typhoon center, the water salinity in the near-surface increased noticeably. The heat pumping from the mixed layer into the thermocline induced by downwelling and the upwelling induced by the positive wind stress curl are the main causes for the different temperature and salinity variations on the different sides of the track. However, most studies of the underlying mechanism are at the single gene level; there are few reports regarding the mechanism at the whole-genome level. Using a rat genomic gene-chip, we profiled hippocampal gene expression changes in rats after electroacupuncture therapy. Electroacupuncture therapy alleviated depres- sion-related manifestations in the model rats. Using gene-chip analysis, we demonstrated that electroacupuncture at Baihui (DU20) and Yintang (EX-HN3) regulates the expression of 21 genes. Real-time PCR showed that the genes Vgf, lgf2, Trnp32, Loc500373, Hifla, Folrl, Nrnb, and Rtn were upregulated or downregulated in depression and that their expression tended to nor- malize after electroacupuncture therapy. These results indicate that electroacupuncture at Baihui and Yintang modulates depression by regulating the expression of particular genes.展开更多

Strong contrasting dipping layered reflections (4—11 s, TWT),which dominate the lower crust of the northern part of the profile, are interpreted as the underplating of the basalt related to extensional tectonism. In the south of the profile, the Yangtze craton is characterized by strong reflections in the middle crust, showing a distinct two-layer crustal structure. Over the region of the Tongling uplift, there appear the complex arc shape reflections, suggesting folded, faulted and intruded structures, and the transparent zone below them reveals the existence of batholith. The south dipping strong reflections between the upper crust and lower crust (4—7 s, TWT) suggest a detachment between them. The detachment provided space for the magma intrusion, and caused the formation of the batholith. The Yangtze craton has a clear Moho re-flection, while the Tongling uplift has a weak Moho, whereas below the reflective lower crust in the northern part of the profile, there are sub-Moho reflections. The abrupt variation of the Moho characteristics within a short distance indicates the complexity of magmatic activity.展开更多

Defects in apoptotic machinery are one of the main mechanisms that cells employ to evade cell death and become cancerous. Targeting the apoptotic defects, either by direct inhibition of BCL-2 family proteins or through modulation of regulatory pathways, can restore cell sensitivity to cell death. This review will focus on the aspects of BCL-2 family proteins, their interactions with kinase pathways, and how novel targeted agents can help overcome the apoptotic blockades. Furthermore, functional assays, such as BH3 profiling, may help in predicting responses to chemotherapies and aid in the selection of combination therapies by determining the mitochondrial threshold for initiating cell death.展开更多

As an important information source from the demand side, Advanced Metering Infrastructure (AMI) has gained increasing popularity all over the world. By making full use of the data gathered by AMI, stakeholders of the electrical industry can have a better understanding of electrical consumption behavior. This is a significant strategy to improve operation efficiency and enhance power grid reliability. To implement this strategy, researchers have explored many data mining techniques for load profiling. This paper performs a state-of-the-art, comprehensive review of these data mining techniques from the perspectives of different technical approaches including direct clustering, indirect clustering, clustering evaluation criteria, and customer segmentation. On this basis, the prospects for implementing load profiling to demand response applications, price-based and incentivebased, are further summarized. Finally, challenges and opportunities of load profiling techniques in future power industry, especially in a demand response world, are discussed.展开更多

In this study, we screened Arabidopsis lines overexpressing rice full-length (FL) cDNAs (rice FOX Arabidopsis lines) using a gas chromatography-time-of-flight mass spectrometry (GC-TOF/MS)-based technique to identify rice genes that caused metabolic changes. This screening system allows fast and reliable identification of candi- date lines showing altered metabolite profiles. We performed metabolomic and transcriptomic analysis of a rice FOX Ara- bidopsis line that harbored the FL cDNA of the rice ortholog of the Lateral Organ Boundaries (LOB) Domain (LBD)/ Asymmetric Leaves2-1ike (ASL) gene of Arabidopsis, At-LBD37/ASL39. The investigated rice FOX Arabidopsis line showed prominent changes in the levels of metabolites related to nitrogen metabolism. The transcriptomic data as well as the results from the metabolite analysis of the Arabidopsis At-LBD37/ASL39-overexpressor plants were consistent with these findings. Furthermore, the metabolomic and transcriptomic analysis of the Os-LBD37/ASL39-overexpressing rice plants indicated that Os-LBD37/ASL39 is associated with processes related to nitrogen metabolism in rice. Thus, the combination of a metabolomics-based screening method and a gain-of-function approach is useful for rapid characterization of novel genes in both Arabidopsis and rice.展开更多
The present study was designed to develop and validate a sensitive and reliable ultra high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-QTOF/MS) method to separate an...The present study was designed to develop and validate a sensitive and reliable ultra high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-QTOF/MS) method to separate and identify the chemical constituents of Qixue Shuangbu Tincture(QXSBT), a classic traditional Chinese medicine(TCM) prescription. Under the optimized UPLC and QTOF/MS conditions, 56 components in QXSBT, including chalcones, triterpenoids, protopanaxatriol, flavones and flavanones were identified and tentatively characterized within a running time of 42 min. The components were identified by comparing the retention times, accurate mass, and mass spectrometric fragmentation characteristic ions, and matching empirical molecular formula with that of the published compounds. In conclusion, the established UPLC-QTOF/MS method was reliable for a rapid identification of complicated components in the TCM prescriptions.展开更多

文摘The present study was designed to develop and validate a sensitive and reliable ultra high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-QTOF/MS) method to separate and identify the chemical constituents of Qixue Shuangbu Tincture(QXSBT), a classic traditional Chinese medicine(TCM) prescription. Under the optimized UPLC and QTOF/MS conditions, 56 components in QXSBT, including chalcones, triterpenoids, protopanaxatriol, flavones and flavanones were identified and tentatively characterized within a running time of 42 min. The components were identified by comparing the retention times, accurate mass, and mass spectrometric fragmentation characteristic ions, and matching empirical molecular formula with that of the published compounds. In conclusion, the established UPLC-QTOF/MS method was reliable for a rapid identification of complicated components in the TCM prescriptions.
