A combined method of high performance liquid chromatograph-elecrtrospray-ionization mass spectrometer(HPLC-ESI-MS/MS) coupled with a photodiode array detector(HPLC-DAD) and principal component analysis(PCA) was ...A combined method of high performance liquid chromatograph-elecrtrospray-ionization mass spectrometer(HPLC-ESI-MS/MS) coupled with a photodiode array detector(HPLC-DAD) and principal component analysis(PCA) was applied to the qualitative and quantitative analyses of alkaloids in Cortex Phellodendri(CP) samples, and to the differentiation of two species of CP, Cortex Phellodendri Chinensis(CPC) and Cortex Phellodendri Amurensis(CPA). Twenty-two peaks appeared in the HPLC-MS base peak chromatogram of CP detected by the HPLC-ESI-MS/MS analysis, and the alkaloids were identified according to the MSn data, the known MS fragmentation rules and the literature data. Five alkaloids including berberine, palmatine, jatrorrhizine, phellodendrine and magnoflorine were simultaneously determinated by the HPLC-DAD. Berberine was the primary component in all CP samples, and the contents of berberine and palmatine were exploited to be two critical parameters for effective discrimination between the two species of CP. The average content of berberine in CPC(58.75 mg/g) was higher than that in CPA(9.16 mg/g), while the content of palmatine was less, only 0.25 mg/g in CPC and 4.19 mg/g in CPA. With the use of PCA, samples datasets were separated successfully into two different clusters corresponding to the two species, and berberine, pahnatine, phellodendrine and magnoflorine contribute most to the above mentioned calssifying . The proposed method oroved to be a useful tool in the aualitv control of Chinese herbal medicines.展开更多
Objective: To investigate whether the dried root of Phellodendron amurense Ruprecht(Phellodendri cortex; PC) extract improves arthritic symptoms through anti-inflammatory and immune-modulatory effects in collagen-i...Objective: To investigate whether the dried root of Phellodendron amurense Ruprecht(Phellodendri cortex; PC) extract improves arthritic symptoms through anti-inflammatory and immune-modulatory effects in collagen-induced arthritis in mice. Methods: Rheumatoid arthritis(RA) was induced in male DBA/1 mice by immunization with type Ⅱ collagen(ColⅡ). CIA mice were divided into 5 groups(n=10 per a group) with normal, CIA control, PC extract(50 mg/kg and 100 mg/kg)-treated, and meloxicam(50 mg/kg)-treated as the reference drug. The PC extract or meloxicam were administered orally in CIA mice once a day for 14 days after arthritis induction. Arthritic score, levels of anti-ColⅡ IgG2a antibody, prostaglandin E2(PGE2), tumor necrosis factor(TNF)-α, and interleukin(IL)-17 in the sera of CIA mice were measured. Histopathological changes in the ankle joints of CIA mice were also analyzed by staining with hematoxylin and eosin(H and E), safranin-O and immunohistochemistry using anti-TNF-α and anti-IL-17 antibodies. Results: The arthritic score was increased in CIA mice in a time-dependent manner, as were the serum levels of anti-ColⅡ IgG2a antibody, PGE2, TNF-α, and IL-17. However, the oral administration of PC extract at 50 and 100 mg/kg in CIA mice significantly decreased the arthritic scores, and the serum levels of anti-ColⅡ IgG2a, PGE2, TNF-α, and IL-17 compared with those in the CIA group(P〈0.05 or P〈0.01). Furthermore, histopathological improvement of the joint architecture in CIA mice was observed after administration of PC extract. PC extract also significantly inhibited the expression of TNF-α and IL-17 in the joints of CIA mice by suppressing the expression of their m RNA and proteins. Conclusion: PC extract may improve the pathological progression of RA through the inhibition of joint destruction by synovial inflammation and immune-stimulation, therefore, it would be a potential anti-arthritic agent in RA.展开更多
Cervical eropion,a pathological change associated with chronic cervicitis,is a common condition that is difficult to cure.M any pa tients particularly those with mild or medium infection and those preparing for pregna...Cervical eropion,a pathological change associated with chronic cervicitis,is a common condition that is difficult to cure.M any pa tients particularly those with mild or medium infection and those preparing for pregnancy require a simple but effective medication.In this study,extracts of an empirical herbal prescription composed of six Chinese traditional drugs viz Cortex Phellodendri,Rhizoma Coptidis,Olibanum,Myrrha,borneol and catechu were formulated to facilitate intravaginal administration and improve efficacy.An extract of the first four components was formulated with borneol as a thermosensitive gel(TG)while an extract of catechu used to prepare a regular gel(CG)because of a chemical incompatibility.The optimized TG was prepared using poloxamer 407 and poloxamer 188.The CG was prepared using glycerin,carbopol and triethanolamine.The gels were characterized in vitro in terms of release of berberine(TG)and total catechins(CG)and in vivo in a rat model of cervical erosion.Trea tment by once daily application of the TG for 7 days followed by once daily application of the CG for 3 days produced a restoration of normal tissues.Gel formulation of the empirical Chinese traditional remedy appears to provide a promising treatment for cervical erosion.展开更多

A rapid method for the simultaneous determination of berberine(BBR),matrine(MT)and oxymatrine(OMT)by nonaqueous capillary electrophoresis(NACE)was developed.Optimum separation of the analytes was obtained on a 50cm...A rapid method for the simultaneous determination of berberine(BBR),matrine(MT)and oxymatrine(OMT)by nonaqueous capillary electrophoresis(NACE)was developed.Optimum separation of the analytes was obtained on a 50cm×50μm i.d.fused-silica capillary using a non-aqueous buffer system of 70mM ammonium acetate,7.0% acetic acid and 10% acetonitrile at 25kV and 20℃.The relative standard deviations(R.S.D.)of the migration times and peak areas of the three active components were 0.06%-0.20% and 0.12%-3.41% for berberine,0.11%-0.60% and 0.74%-1.63% for matrine,0.15% and 0.45% for oxymatrine,respectively.Detection limits of berberine,matrine and oxymtrine were 0.18μg/mL,4.08μg/mL and 4.16μg/mL,respectively.In the tested concentration range,good linear relationships(0.9992 for berberine,0.9988 for matrine and 0.9988 for oxymatrine)were observed.The linear calibration ranges were 0.45-360.0μg/mL for berberine,8.16-408.0μg/mL for matrine and 20.8-416.0μg/mL for oxymatrine.This method has been successfully applied to the phytochemical analysis of alkaloids extracts from two commonly used traditional Chinese herbal drugs:Sophora flavescens Ait.(Kushen)and Cortex phellodendri chinensis(Huangbai)and their medicinal preparations.展开更多
Coptidis Rhizoma(Chinese:Huanglian)and Phellodendri Chinensis Cortex(Chinese:Huangbo)are widely used Traditional Chinese Medicine,and often used in combination because of their similar pharmacological effects in clini...Coptidis Rhizoma(Chinese:Huanglian)and Phellodendri Chinensis Cortex(Chinese:Huangbo)are widely used Traditional Chinese Medicine,and often used in combination because of their similar pharmacological effects in clinical practice.However,the quality control methods of the two drugs are different and complicated,which is time consuming and laborious in practical application.In this paper,rapid and simultaneous determination of moisture and berberine in Coptidis Rhizoma(CR)and Phellodendri Chinensis Cortex(PC)was realized by near-infrared spectroscopy(NIRs)combined with global models.Competitive adaptive reweighted sampling(CARS)and successive projection algorithm(SPA)method were applied for variable selection.Principal component analysis(PCA)and partial least squares regression method(PLSR)were applied for qualitative and quantitative analysis,respectively.The characteristic variables of berberine showed similarity and consistency in distribution,providing basis for the global models.For moisture content,the global model had relative standard error of prediction set(RSEP)value of 3.04%and 2.53%for CR and PC,respectively.For berberine content,the global model had RSEP value of 5.41%and 3.97%for CR and PC,respectively.These results indicated the global models based on CARS-PLS method achieved satisfactory prediction for moisture and berberine content,improving the determination e±ciency.Furthermore,the greater range and larger number of samples enhanced the reliance of the global model.The NIRs combined with global models could be a powerful tool for quality control of CR and PC.展开更多
Background:Compound cortex phellodendri liquid(CPL)is a kind of classical compound preparation,which has potential curative effect in treating inflammatory diseases.Increasing evidences support that inflammation plays...Background:Compound cortex phellodendri liquid(CPL)is a kind of classical compound preparation,which has potential curative effect in treating inflammatory diseases.Increasing evidences support that inflammation plays important roles in the pathogenesis of myeloproliferative neoplasms(MPN).This study aims to preliminarily clarify the therapeutic potential and molecular mechanisms of CPL for MPN based on network pharmacology and molecular docking techniques.Methods:The active components and corresponding action targets of CPL were searched by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),while MPN-related targets were searched through GeneCards,DisGeNET,OMIM,DrugBank and TTD databases respectively.Protein-Protein Interaction(PPI)Networks of potential targets were constructed using STRING 11.5 and analyzed visually with Cytoscape 3.9.1.In addition,Metascape platform was used for GO and KEGG analysis that were subsequently visualized with Cytoscape 3.9.1 built-in plug-ins CluoGO or SangerBox platform.Finally,Autodock Vina was used for molecular docking of potential targets and main active ingredients,which were visualized with Pymol software.Experimentally,we used in vitro mouse primary cells culture system to evaluate the effect of CPL on the erythroid and megakaryocytes differentiation that are excessively driven in MPN respectively.Results:The active components of CPL in the treatment of MPN are mainly flavonoids.The core proteins of CPL for MPN intervention are correlated to TP53,AKT1,JUN,CASP3,EGFR,TNF,MYC,IL6.Multiple signaling pathways were closely related to the treatment of MPN intervened by CPL,including PI3K-Akt signaling,TNF-αsignaling,JAK-STAT signaling and NF-κB signaling pathways.These potential targets had good conformation with the core active ingredients of CPL.In line with above findings,we demonstrated that CPL significantly inhibits the proliferation of differentiation of erythrocytes and megakaryocytes in vitro,further supporting the therapeutic poten展开更多
Objective:To compare the therapeutic effects of three different external wet compress methods on facial hormone dependent dermatitis(FCAD),and to provide reference for clinical treatment of FCAD.Methods:From June 2016...Objective:To compare the therapeutic effects of three different external wet compress methods on facial hormone dependent dermatitis(FCAD),and to provide reference for clinical treatment of FCAD.Methods:From June 2016 to June 2019,160 FCAD patients in our dermatology clinic were selected and divided into three groups according to random number table:the first group was treated with Compound Cortex Phellodendri liquid coating,the second group was treated with collagen paste dressing,and the third group was treated with 0.9%sodium chloride injection solution.Apply 8 layers of gauze or cold compress directly to the skin lesions for 4 weeks in each group.Results:After 4 weeks of treatment,no adverse reactions were found.The effective rates of the first group,the second group,and the third group were 72.22%,62.26%and 41.51%,respectively.Compared with the third group,the effective rate of the first group and the second group was significantly higher,and there was statistical significance between the groups(x2=10.294,x2=4.573,P<0.05);but there was no significant statistical difference between the first group and the second group(x2=1.205,P>0.05).Conclusion:Compound Cortex Phellodendri coating is suitable for FCAD patients with severe inflammation,collagen paste dressing is suitable for FCAD patients with mild to moderate inflammatory reaction and pigmentation,and 0.9%sodium chloride injection is suitable for patients with mild facial flushing and swelling.Clinical treatment should be selected according to the patient's condition and rash characteristics.展开更多
Traditional Chinese medicine(TCM)Phellodendri Chinensis Cortex(PCC)has been used for the treatment of human hepatocellular carcinoma,but the underlying mechanisms are still unclear.In this study,15 active compositions...Traditional Chinese medicine(TCM)Phellodendri Chinensis Cortex(PCC)has been used for the treatment of human hepatocellular carcinoma,but the underlying mechanisms are still unclear.In this study,15 active compositions of PCC were obtained from Traditional Chinese Medicine Systems Pharmacology Database(TCMSP),and 505 putative identified targets of PCC were screened by Swiss Target Prediction server.Next,HCC data was downloaded from Drugbank and GeneCards databases.Furtherly,45 common targets were revealed.The network diagrams of the active component-target network,protein-protein interaction(PPI)network and active component-target-pathway network were constructed using Cytoscape software.The analysis of the network results showed that the active ingredients of PCC,such as berberine,obacunone,rutaecarpine,candletoxin A,palmatine,isocorypalmine,quercetin,and(S)-Canadine,had a good binding activity with more targets.Additionally,Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses revealed that common targets were significantly enriched in Ras signaling pathway,ErbB signaling pathway,and Mammalian target of rapamycin(mTOR)signaling pathway.Altogether,the multi-component,multi-target,and multi-pathway characteristics of PCC provided a reference for the in-depth study of the mechanism of PCC in the treatment of HCC.展开更多

文摘Background:Compound cortex phellodendri liquid(CPL)is a kind of classical compound preparation,which has potential curative effect in treating inflammatory diseases.Increasing evidences support that inflammation plays important roles in the pathogenesis of myeloproliferative neoplasms(MPN).This study aims to preliminarily clarify the therapeutic potential and molecular mechanisms of CPL for MPN based on network pharmacology and molecular docking techniques.Methods:The active components and corresponding action targets of CPL were searched by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),while MPN-related targets were searched through GeneCards,DisGeNET,OMIM,DrugBank and TTD databases respectively.Protein-Protein Interaction(PPI)Networks of potential targets were constructed using STRING 11.5 and analyzed visually with Cytoscape 3.9.1.In addition,Metascape platform was used for GO and KEGG analysis that were subsequently visualized with Cytoscape 3.9.1 built-in plug-ins CluoGO or SangerBox platform.Finally,Autodock Vina was used for molecular docking of potential targets and main active ingredients,which were visualized with Pymol software.Experimentally,we used in vitro mouse primary cells culture system to evaluate the effect of CPL on the erythroid and megakaryocytes differentiation that are excessively driven in MPN respectively.Results:The active components of CPL in the treatment of MPN are mainly flavonoids.The core proteins of CPL for MPN intervention are correlated to TP53,AKT1,JUN,CASP3,EGFR,TNF,MYC,IL6.Multiple signaling pathways were closely related to the treatment of MPN intervened by CPL,including PI3K-Akt signaling,TNF-αsignaling,JAK-STAT signaling and NF-κB signaling pathways.These potential targets had good conformation with the core active ingredients of CPL.In line with above findings,we demonstrated that CPL significantly inhibits the proliferation of differentiation of erythrocytes and megakaryocytes in vitro,further supporting the therapeutic poten
文摘Objective:To compare the therapeutic effects of three different external wet compress methods on facial hormone dependent dermatitis(FCAD),and to provide reference for clinical treatment of FCAD.Methods:From June 2016 to June 2019,160 FCAD patients in our dermatology clinic were selected and divided into three groups according to random number table:the first group was treated with Compound Cortex Phellodendri liquid coating,the second group was treated with collagen paste dressing,and the third group was treated with 0.9%sodium chloride injection solution.Apply 8 layers of gauze or cold compress directly to the skin lesions for 4 weeks in each group.Results:After 4 weeks of treatment,no adverse reactions were found.The effective rates of the first group,the second group,and the third group were 72.22%,62.26%and 41.51%,respectively.Compared with the third group,the effective rate of the first group and the second group was significantly higher,and there was statistical significance between the groups(x2=10.294,x2=4.573,P<0.05);but there was no significant statistical difference between the first group and the second group(x2=1.205,P>0.05).Conclusion:Compound Cortex Phellodendri coating is suitable for FCAD patients with severe inflammation,collagen paste dressing is suitable for FCAD patients with mild to moderate inflammatory reaction and pigmentation,and 0.9%sodium chloride injection is suitable for patients with mild facial flushing and swelling.Clinical treatment should be selected according to the patient's condition and rash characteristics.
文摘Traditional Chinese medicine(TCM)Phellodendri Chinensis Cortex(PCC)has been used for the treatment of human hepatocellular carcinoma,but the underlying mechanisms are still unclear.In this study,15 active compositions of PCC were obtained from Traditional Chinese Medicine Systems Pharmacology Database(TCMSP),and 505 putative identified targets of PCC were screened by Swiss Target Prediction server.Next,HCC data was downloaded from Drugbank and GeneCards databases.Furtherly,45 common targets were revealed.The network diagrams of the active component-target network,protein-protein interaction(PPI)network and active component-target-pathway network were constructed using Cytoscape software.The analysis of the network results showed that the active ingredients of PCC,such as berberine,obacunone,rutaecarpine,candletoxin A,palmatine,isocorypalmine,quercetin,and(S)-Canadine,had a good binding activity with more targets.Additionally,Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses revealed that common targets were significantly enriched in Ras signaling pathway,ErbB signaling pathway,and Mammalian target of rapamycin(mTOR)signaling pathway.Altogether,the multi-component,multi-target,and multi-pathway characteristics of PCC provided a reference for the in-depth study of the mechanism of PCC in the treatment of HCC.
