【目的】探讨COI基因作为标准DNA条形码技术鉴定外形差异较小的地方鸡品种的可行性。【方法】以华南地区9种优质地方鸡(怀乡鸡、清远麻鸡、惠阳胡须鸡、中山沙栏鸡、阳山鸡、杏花鸡、五华三黄鸡、文昌鸡和广西三黄鸡)和国外引进品种隐...【目的】探讨COI基因作为标准DNA条形码技术鉴定外形差异较小的地方鸡品种的可行性。【方法】以华南地区9种优质地方鸡(怀乡鸡、清远麻鸡、惠阳胡须鸡、中山沙栏鸡、阳山鸡、杏花鸡、五华三黄鸡、文昌鸡和广西三黄鸡)和国外引进品种隐性白羽鸡为试验材料,测定标准的DNA条形码技术的线粒体细胞色素C氧化酶亚基I(cytochrome C oxidase subunit I,COI),同时下载已发表的31条家鸡和原鸡及绿头鸭的COI基因序列,分析品种遗传多样性与遗传距离,构建单倍型中介网络图和系统发生邻接树,界定区分品种特异的单倍型。【结果】除去PCR引物序列,获得了695 bp COI基因片段。根据标准的DNA条形码序列,截取648 bp线粒体COI基因序列进行分析。10个鸡品种203个个体共检测到110个变异位点,占分析位点的16.98%,其中90个单一位点突变,20个简约信息位点。平均核苷酸多样性为0.00394(0.00349—0.00560),平均单倍型多样性为0.832(0.763—0.905),其中五华三黄鸡最高,中山沙栏鸡次之,文昌鸡最低。定义了84种单倍型,单倍型1为9个地方鸡种所共享,出现频率为64次;单倍型9和5为家鸡和隐性白羽鸡共享,出现频率分别为29次和19次;每个鸡品种均有品种特异的单倍型。广西三黄鸡、五华三黄鸡与中山沙栏鸡的单倍型数最多,为13个,隐性白羽鸡与清远麻鸡的最少,为8个。不同品种的单倍型分布差异较大,如杏花鸡的单倍型主要分布在1,清远麻鸡主要分布在1和9,惠阳胡须鸡主要分布在1、5和9,隐性白羽鸡主要分布在9和79。10个鸡种品种间遗传距离范围为0.003—0.006,净遗传距离为0—0.003;鸡品种间的遗传距离一般大于鸡品种内的遗传距离;绿头鸭与鸡品种间的遗传距离大于0.2。中介网络图将84个单倍型分为3条进化枝,呈现出一定的品种特异性,如以单倍型9为起点的进化枝没有广西三黄鸡和文昌鸡分布,但另外�展开更多
Meat quality traits of four genotypes of Chinese indigenous chicken [Ninghai chicken(NC),frizzle chicken(FC),Ninghai xiang chicken(XC),and Zhenning loquat chicken(LC)] and one genotype of commercial broiler [Arbor Acr...Meat quality traits of four genotypes of Chinese indigenous chicken [Ninghai chicken(NC),frizzle chicken(FC),Ninghai xiang chicken(XC),and Zhenning loquat chicken(LC)] and one genotype of commercial broiler [Arbor Acres plus broiler(AAB)] were analyzed.The indigenous chickens were raised before the commercial chickens in order to achieve the same final processed days.Indigenous chickens of NC,FC,XC,and LC showed significantly higher inosine-5′-monophosphate(IMP) content,shorter fiber diameter,and lower shear force than those of AAB(P<0.05).In the indigenous genotypes,NC and FC had significantly shorter fiber diameters and lower shear forces than XC and LC(P<0.05),and NC and XC had a higher IMP content than FC and LC(P<0.05).Moreover,the indigenous genotype of LC significantly displayed the highest protein content(P<0.05) in the five genotypes of birds,and no significant differences of protein content were found between the other genotypes of NC,FC,XC,and AAB(P>0.05).The indigenous chickens from FC displayed the highest total lipid content in the five bird genotypes(P<0.05).Significant differences of pH,color values of L* and a*,and drip loss for the five genotypes of birds were also observed.In conclusion,there were significant differences in the meat quality traits of the bird breeds selected in this study,and the indigenous chickens,especially the NC genotype,produced better quality meat as far as the IMP content,fiber diameters,and shear forces were concerned.展开更多
The growth and development regularity and genetic parameters were described in three types of indigenous chicken in China, Shaobo, Huaixiang and Youxi Chicken, by 3 nonlinear mathematics models, Logistics, Gompertz an...The growth and development regularity and genetic parameters were described in three types of indigenous chicken in China, Shaobo, Huaixiang and Youxi Chicken, by 3 nonlinear mathematics models, Logistics, Gompertz and Bertalanfy, according to the data of Body Weight from 1 week to 10 weeks. The results showed that the growth process of the three breeds could be illustrated well by three models, and the indices of fitness were very high (more than 0.99). Among the 3 models, Gompertz model was the best for its less bias from practice. The inflexions of growth were 5.98, 5.11 and 6.16 weeks of age, and body weights were 2115.77 g, 1499.08 g and 1409.62 g, respectively in Shaobo Chicken, Huaixiang Chicken and Youxi Chicken.展开更多
A total of 587 individuals from 12 indigenous chicken breeds from South China and two commercial breeds were genotyped for 26 microsatellites to investigate the genetic diversity and population structure.All microsate...A total of 587 individuals from 12 indigenous chicken breeds from South China and two commercial breeds were genotyped for 26 microsatellites to investigate the genetic diversity and population structure.All microsatellites were found to be polymorphic.The number of alleles per locus ranged from 5 to 36,with an average of12.10±7.00(SE).All breeds,except White Recessive Rock,had high allelic polymorphism(>0.5).Higher genetic diversity was revealed in the indigenous chicken breeds rather than in the commercial breeds.Potential introgression from the commercial breeds into the indigenous chickens was also detected.The population structure of these indigenous chicken breeds could be explained by their geographical distribution,which suggested the presence of independent history of breed formation.Data generated in this study will provide valuable information to the conservation for indigenous chicken breeds in future.展开更多

[ Method ] Using ELISA assay and virus isolation method, epidemiological investigation of ALV-AB and ALV-J avian leukemia of 10 indigenous chicken breeds were conducted. ALV dynamics were monitored in F2 generation of four chicken lines. ALV pollution of attenuated live vaccines used in raising process was also inspected through sampling method. [ Result] The positive rate of ALV-P27 antigen was 0 -62.1% ; the positive rate of ALV-AB antibody was 0 -25.0% ; the positive rate of ALV-J antibody was 0 - 59.0% ; the positive rate of virus isolation was 0 - 22.0%. The positive rate of ALV-P'27 antigen in 1 -day-old chick mecunium of four lines was 6.0% - 67.0%. The positive rate of virus isolation in 6-week-old chickens was 2.0% - 34.3%. Two kinds of vaccines from two hatches pro- duced by a manufacturer were polluted by ALV. [ Conclusion] Most indigenous chickens were infected by ALV. There were great differences among different breeds of indigenous chicken, which might be related to ALV genetic resistance of different indigenous chickens. The ALV positive rates of F: chicks were slightly enhanced in some lines, which might be related to vaccine pollution.展开更多
