AIM:To determine the molecular mechanisms of Shugan decoction(SGD) in the regulation of colonic motility and visceral hyperalgesia(VHL) in irritable bowel syndrome(IBS).METHODS:The chemical compounds contained in SGD ...AIM:To determine the molecular mechanisms of Shugan decoction(SGD) in the regulation of colonic motility and visceral hyperalgesia(VHL) in irritable bowel syndrome(IBS).METHODS:The chemical compounds contained in SGD were measured by high-performance liquid chromatography.A rat model of IBS was induced by chronic water avoidance stress(WAS).The number of fecal pellets was counted after WAS and the pain pressure threshold was measured by colorectal distension.Morphological changes in colonic mucosa were detected by hematoxylin-eosin staining.The contents of tumor necrosis factor(TNF)-αin colonic tissue and calcitonin-gene-related peptide(CGRP)in serum were measured by ELISA.The protein expression of serotonin[5-hydroxytryptamide(5-HT)],serotonin transporter(SERT),chromogranin A(Cg A)and CGRP incolon tissue was measured by immunohistochemistry.RESULTS:SGD inhibited colonic motility dysfunction and VHL in rats with IBS.Blockers of transient receptor potential(TRP)vanilloid 1(TRPV1)(Ruthenium Red)and TRP ankyrin-1(TRPA1)(HC-030031)and activator of protease-activated receptor(PAR)4 increased the pain pressure threshold,whereas activators of PAR2and TRPV4 decreased the pain pressure threshold in rats with IBS.The effect of SGD on pain pressure threshold in these rats was abolished by activators of TRPV1(capsaicin),TRPV4(RN1747),TRPA1(Polygodial)and PAR2(AC55541).In addition,CGRP levels in serum and colonic tissue were both increased in these rats.TNF-αlevel in colonic tissue was also significantly upregulated.However,the levels of 5-HT,SERT and Cg A in colonic tissue were decreased.All these pathological changes in rats with IBS were attenuated by SGD.CONCLUSION:SGD alleviated VHL and attenuated colon motility in IBS,partly by regulating TRPV1,TRPV4,TRPA1,PAR2,5-HT,Cg A and SERT,and reducing CGRP and TNF-αlevel.展开更多

目的观察内热针治疗早期股骨头缺血性坏死的临床效果。方法选取2017年1月-2018年12月在解放军总医院第一医学中心康复医学科就诊的Ficat分期为Ⅰ期和Ⅱ期的接受内热针治疗的股骨头缺血性坏死患者。内热针针刺部位为臀部和腹股沟,每个部...目的观察内热针治疗早期股骨头缺血性坏死的临床效果。方法选取2017年1月-2018年12月在解放军总医院第一医学中心康复医学科就诊的Ficat分期为Ⅰ期和Ⅱ期的接受内热针治疗的股骨头缺血性坏死患者。内热针针刺部位为臀部和腹股沟,每个部位治疗1次,间隔5 d,针刺后加热针体至43℃。记录治疗前、治疗后1个月、治疗后12个月时患者的视觉模拟评分(visual analogue scale,VAS)和Harris髋关节评分(Harris hip score,HHS)。结果21例股骨头缺血性坏死患者中男性15例,女性6例,平均年龄32.1(21~46)岁,Ficat分期Ⅰ期8例,Ⅱ期13例。治疗前、治疗后1个月、治疗后12个月,VAS评分分别为3.7±1.2、1.9±1.5、2.3±1.1,HHS总评分分别为50.6±13.8、73.7±8.6、69.7±13.3,治疗后1个月和治疗后12个月的VAS评分及HHS总评分与治疗前比较,差异有统计学意义(P均<0.01)。HSS评分中疼痛(19.05±8.3 vs 30.8±9.2)、髋关节功能(27.2±6.3 vs 32.9±7.0)、关节活动度(2.1±0.6 vs 3.0±1.1)三个子项目评分治疗后12个月较治疗前均显著提升,结果与总评分趋势一致(P均<0.01)。结论内热针疗法可改善FicatⅠ~Ⅱ期股骨头缺血性坏死患者的疼痛和下肢功能状态。展开更多
We investigated the effects of transient receptor potential M8(TRPM8)channel on the proliferation and motility of androgen-independent prostate cancer PC-3 cells.After being permanently transfected with an empty vecto...We investigated the effects of transient receptor potential M8(TRPM8)channel on the proliferation and motility of androgen-independent prostate cancer PC-3 cells.After being permanently transfected with an empty vector and cDNA encoding the TRPM8 protein,cells were analysed for cell cycle distribution and motility using flow cytometry and scratch assay.Immunocytochemistry and Ca^(2+)imaging analysis revealed the overexpression of functional TRPM8 channel on both endoplasmic reticulum and plasma membrane of PC-3-TRPM8 cells.Cell cycle distribution and scratch assay analysis revealed that TRPM8 induced cell cycle arrest at the G_(0)/G_(1)stage(P<0.05)and facilitated the cell apoptosis induced by starvation(P<0.05).Furthermore,TRPM8 inhibited the migration of PC-3-TRPM8 cells(P<0.01)through the inactivation of focal-adhesion kinase.It appears that TRPM8 was not essential for the survival of PC-3 cells;however,the overexpression of TRPM8 had negative effects on the proliferation and migration of PC-3 cells.Thus,TRPM8 and its agonists may serve as important targets for the treatment of prostate cancer.展开更多
Moxibustion is an important component part of Traditional Chinese Medicine(TCM).Among different kinds of moxibustion methods,thermal stimulation seems to be a pivotal impact factor to the theraputic efficacy.Based on ...Moxibustion is an important component part of Traditional Chinese Medicine(TCM).Among different kinds of moxibustion methods,thermal stimulation seems to be a pivotal impact factor to the theraputic efficacy.Based on its thermal characteristic and treated area-skin,we hypothesize that the thermosensitive TRPV channels may involve in the mechanism of moxibustion.This study,by referring to various experimental and clinical data,analyzes the properties and features of transient receptor potential vanilloid(TRPV)subfamily 1-4 and the impact of moxibustion on these channels.The factors impacting the efficacy of moxibustion treatment were analyzed on three levels:the independent ba-sic factors of moxibustion(temperature,space and time);moxibustion intensity(a compound factor achieved through comprehensive control of the three individual basic factors mentioned above);and moxibustion quantity(the amount of temperature stimulation applied within a certain unit of time,including the total amount of moxibustion treatment).The results from present study show that the effect of moxibustion therapy appears to be determined by the activation of TRPV1-4,mainly TRPV1 and TRPV2.Temperature(the degree of heat stimulation),time and area(how long the treatment lasts and how many TRPV1-4 channels are activated)affect the intensity of moxibustion treatment to form effective moxibustion quantity;this should be considered in clinical moxibustion application.展开更多
Acupuncture is a medical treatment that has been widely pra cticed in China for over 3000 years,yet the neural mechanisms of acupuncture are not fully understood.We hypothesized that neurons and astrocytes act indepen...Acupuncture is a medical treatment that has been widely pra cticed in China for over 3000 years,yet the neural mechanisms of acupuncture are not fully understood.We hypothesized that neurons and astrocytes act independently and synergistically under acupuncture stimulation.To investigate this,we used two-photon in vivo calcium reco rding to observe the effects of acupuncture stimulation at ST36(Zusanli)in mice.Acupuncture stimulation in peripheral acupoints potentiated calcium signals of pyramidal neurons and astrocytes in the somatosensory cortex and resulted in late-onset calcium transients in astrocytes.Chemogenetic inhibition of neurons augmented the astrocytic activity.These findings suggest that acupuncture activates neuronal and astrocytic activity in the somatosensory co rtex and provide evidence for the involvement of both neurons and astrocytes in acupuncture treatment.展开更多
Injury to the brain after intracerebral hemorrhage(ICH)results from numerous complex cellular mechanisms.At present,effective therapy for ICH is limited and a better understanding of the mechanisms of brain injury is ...Injury to the brain after intracerebral hemorrhage(ICH)results from numerous complex cellular mechanisms.At present,effective therapy for ICH is limited and a better understanding of the mechanisms of brain injury is necessary to improve prognosis.There is increasing evidence that ion channel dysregulation occurs at multiple stages in primary and secondary brain injury following ICH.Ion channels such as TWIK-related K+channel 1,sulfonylurea 1 transient receptor potential melastatin 4 and glutamate-gated channels affect ion homeostasis in ICH.They in turn participate in the formation of brain edema,disruption of the blood-brain barrier,and the generation of neurotoxicity.In this review,we summarize the interaction between ions and ion channels,the effects of ion channel dysregulation,and we discuss some therapeutics based on ion-channel modulation following ICH.展开更多

Here, we investigated the role of TRPC3 in undifferentiated mous...Transient receptor potential canonical subfamily member 3(TRPC3) is known to be important for neural development and the formation of neuronal networks. Here, we investigated the role of TRPC3 in undifferentiated mouse embryonic stem cells(mESCs) and during the differentiation of mESCs into neurons. CRISPR/Cas9-mediated knockout(KO) of TRPC3 induced apoptosis and the disruption of mitochondrial membrane potential both in undifferentiated mESCs and in those undergoing neural differentiation. In addition, TRPC3 KO impaired the pluripotency of mESCs. TRPC3 KO also dramatically repressed the neural differentiation of mESCs by inhibiting the expression of markers for neural progenitors, neurons, astrocytes and oligodendrocytes.Taken together, our new data demonstrate an important function of TRPC3 with regards to the survival, pluripotency and neural differentiation of mESCs.展开更多
Spodoptera frugiperda is a highly destructive pest that has become a global problem due to its robust reproductive and migratory capabilities.Transient receptor potential(TRP)channels,which constitute a vast ion chann...Spodoptera frugiperda is a highly destructive pest that has become a global problem due to its robust reproductive and migratory capabilities.Transient receptor potential(TRP)channels,which constitute a vast ion channel family,play pivotal roles in sensing the external environment and maintaining internal homeostasis in insects.TRP channels have been widely investigated for their critical roles in regulating various insect behaviors in recent years.In this study,we identified 15 TRP gene loci encoding 26 transcripts in the genome of S.frugiperda and analyzed their expression profiles at different developmental stages.The results revealed that S.frugiperda possesses four TRPC genes,six TRPA genes,one TRPM gene,two TRPV genes,one TRPN gene,and one TRPML gene,while a canonical TRPP is absent.Moreover,the SfruTRPA1 was functionally characterized using the Xenopus oocyte expression system.The results showed that SfruTRPA1 is activated by temperature increases from 20 to 45℃,and there is no significant desensitization after repeated stimuli within the same temperature range.Additionally,SfruTRPA1 is activated by certain natural chemicals,including allyl isothiocyanate(AITC)and cinnamaldehyde(CA).These findings provide valuable insights to the TRP genes in S.frugiperda.展开更多

Fo...The aim of this research was to assess the antinociceptive activity of the transient receptor potential (TRP) channel TRPV1, TRPM8, and TRPA1 antagonists in neurogenic, tonic, and neuropathic pain models in mice. For this purpose, TRP channel antagonists were administered into the dorsal surface of a hind paw 15 min before capsaicin, allyl isothiocyanate (AITC), or formalin. Their antiallodynic and antihyperalgesic efficacies after intraperitoneal ad- ministration were also assessed in a paclitaxel-induced neuropathic pain model. Motor coordination of paclitaxel- treated mice that received these TRP channel antagonists was investigated using the rotarod test. TRPV1 antagonists, capsazepine and SB-366791, attenuated capsaicin-induced nociceptive reaction in a concentration-dependent manner. At 8 pg/20 pl, this effect was 51% (P〈0.001) for capsazepine and 37% (P〈0.05) for SB-366791. A TRPA1 antagonist, A-967079, reduced pain reaction by 48% (P〈0.05) in the AITC test and by 54% (P〈0.001) in the early phase of the formalin test. The test compounds had no influence on the late phase of the formalin test. In paclitaxel-treated mice, they did not attenuate heat hyperalgesia but N-(3-aminopropyl)-2-{[(3-methylphenyl)methyl]oxy}-N-(2-thienylmethyl) benzamide hydrochloride salt (AMTB), a TRPM8 antagonist, reduced cold hyperalgesia and tactile allodynia by 31% (P〈0.05) and 51% (P〈0.01), respectively. HC-030031, a TRPA1 channel antagonist, attenuated tactile allodynia in the von Frey test (62%; P〈0.001). In conclusion, distinct members of TRP channel family are involved in different pain models in mice. Antagonists of TRP channels attenuate nocifensive responses of neurogenic, tonic, and neuropathic pain, but their efficacies strongly depend on the pain model used.展开更多
AIM: To explore the possibility of using the Noninvasive Micro-test Technique (NMT) to investigate the role of Transient Receptor Potential Canonical 1 (TRPC1) in regulating Ca^2+ influxes in HL-7702 cells, a no...AIM: To explore the possibility of using the Noninvasive Micro-test Technique (NMT) to investigate the role of Transient Receptor Potential Canonical 1 (TRPC1) in regulating Ca^2+ influxes in HL-7702 cells, a normal human liver cell line.METHODS: Net Ca^2+ fluxes were measured with NMT, a technology that can obtain dynamic information of specific/selective ionic/molecular activities on material surfaces, non-invasively. The expression levels of TRPCl were increased by liposomal transfection, whose effectiveness was evaluated by Western-blotting and single cell reverse transcription-polymerase chain reaction.RESULTS: Ca^2+ influxes could be elicited by adding 1 mmol/L CaCl2 to the test solution of HL-7702 cells. They were enhanced by addition of 20 μmol/L noradrenalin and inhibited by 100 μmol/L LaCl3 (a non-selective Ca^2+ channel blocker); 5 μmol/L nifedipine did not induce any change. Overexpression of TRPCl caused increased Ca^2+ influx. Five micromoles per liter nifedipine did not inhibit this elevation, whereas 100 μmol/L LaCI3 did.CONCLUSION: In HL-7702 cells, there is a type of TRPCl-dependent Ca^2+ channel, which could be detected v/a NMT and inhibited by La^3+.展开更多

Hypomagnesemia has recently been recognized a...Proton pump inhibitors (PPIs) are commonly used in clinical practice for the prevention and treatment of peptic ulcer, gastritis, esophagitis and gastroesophageal refux. Hypomagnesemia has recently been recognized as a side effect of PPIs. Low magnesium levels may cause symptoms from several systems, some of which being potentially serious, such as tetany, seizures and arrhythmias. It seems that PPIs affect the gastrointesti-nal absorption of magnesium. Clinicians should be vigi-lant in order to timely consider and prevent or reverse hypomagnesemia in patients who take PPIs, especially if they are prone to this electrolyte disorder.展开更多
Transient receptor potential(TRP)channels are strongly associated with colon cancer development and progression.This study leveraged a multivariate Cox regression model on publicly available datasets to construct a TR...Transient receptor potential(TRP)channels are strongly associated with colon cancer development and progression.This study leveraged a multivariate Cox regression model on publicly available datasets to construct a TRP channels-associated gene signature,with further validation of signature in real world samples from our hospital treated patient samples.Kaplan-Meier(K-M)survival analysis and receiver operating characteristic(ROC)curves were employed to evaluate this gene signature’s predictive accuracy and robustness in both training and testing cohorts,respectively.Additionally,the study utilized the CIBERSORT algorithm and single-sample gene set enrichment analysis to explore the signature’s immune infiltration landscape and underlying functional implications.The support vector machine algorithm was applied to evaluate the signature’s potential in predicting chemotherapy outcomes.The findings unveiled a novel three TRP channels-related gene signature(MCOLN1,TRPM5,and TRPV4)in colon adenocarcinoma(COAD).The ROC and K-M survival curves in the training dataset(AUC=0.761;p=1.58e-05)and testing dataset(AUC=0.699;p=0.004)showed the signature’s robust predictive capability for the overall survival of COAD patients.Analysis of the immune infiltration landscape associated with the signature revealed higher immune infiltration,especially an increased presence of M2 macrophages,in high-risk group patients compared to their low-risk counterparts.High-risk score patients also exhibited potential responsiveness to immune checkpoint inhibitor therapy,evident through increased CD86 and PD-1 expression profiles.Moreover,the TRPM5 gene within the signature was highly expressed in the chemoresistance group(p=0.00095)and associated with poor prognosis(p=0.036)in COAD patients,highlighting its role as a hub gene of chemoresistance.Ultimately,this signature emerged as an independent prognosis factor for COAD patients(p=6.48e-06)and expression of model gene are validated by public data and real-world patients.Overall,this bioi展开更多

文摘目的观察内热针治疗早期股骨头缺血性坏死的临床效果。方法选取2017年1月-2018年12月在解放军总医院第一医学中心康复医学科就诊的Ficat分期为Ⅰ期和Ⅱ期的接受内热针治疗的股骨头缺血性坏死患者。内热针针刺部位为臀部和腹股沟,每个部位治疗1次,间隔5 d,针刺后加热针体至43℃。记录治疗前、治疗后1个月、治疗后12个月时患者的视觉模拟评分(visual analogue scale,VAS)和Harris髋关节评分(Harris hip score,HHS)。结果21例股骨头缺血性坏死患者中男性15例,女性6例,平均年龄32.1(21~46)岁,Ficat分期Ⅰ期8例,Ⅱ期13例。治疗前、治疗后1个月、治疗后12个月,VAS评分分别为3.7±1.2、1.9±1.5、2.3±1.1,HHS总评分分别为50.6±13.8、73.7±8.6、69.7±13.3,治疗后1个月和治疗后12个月的VAS评分及HHS总评分与治疗前比较,差异有统计学意义(P均<0.01)。HSS评分中疼痛(19.05±8.3 vs 30.8±9.2)、髋关节功能(27.2±6.3 vs 32.9±7.0)、关节活动度(2.1±0.6 vs 3.0±1.1)三个子项目评分治疗后12个月较治疗前均显著提升,结果与总评分趋势一致(P均<0.01)。结论内热针疗法可改善FicatⅠ~Ⅱ期股骨头缺血性坏死患者的疼痛和下肢功能状态。

文摘We investigated the effects of transient receptor potential M8(TRPM8)channel on the proliferation and motility of androgen-independent prostate cancer PC-3 cells.After being permanently transfected with an empty vector and cDNA encoding the TRPM8 protein,cells were analysed for cell cycle distribution and motility using flow cytometry and scratch assay.Immunocytochemistry and Ca^(2+)imaging analysis revealed the overexpression of functional TRPM8 channel on both endoplasmic reticulum and plasma membrane of PC-3-TRPM8 cells.Cell cycle distribution and scratch assay analysis revealed that TRPM8 induced cell cycle arrest at the G_(0)/G_(1)stage(P<0.05)and facilitated the cell apoptosis induced by starvation(P<0.05).Furthermore,TRPM8 inhibited the migration of PC-3-TRPM8 cells(P<0.01)through the inactivation of focal-adhesion kinase.It appears that TRPM8 was not essential for the survival of PC-3 cells;however,the overexpression of TRPM8 had negative effects on the proliferation and migration of PC-3 cells.Thus,TRPM8 and its agonists may serve as important targets for the treatment of prostate cancer.

文摘Moxibustion is an important component part of Traditional Chinese Medicine(TCM).Among different kinds of moxibustion methods,thermal stimulation seems to be a pivotal impact factor to the theraputic efficacy.Based on its thermal characteristic and treated area-skin,we hypothesize that the thermosensitive TRPV channels may involve in the mechanism of moxibustion.This study,by referring to various experimental and clinical data,analyzes the properties and features of transient receptor potential vanilloid(TRPV)subfamily 1-4 and the impact of moxibustion on these channels.The factors impacting the efficacy of moxibustion treatment were analyzed on three levels:the independent ba-sic factors of moxibustion(temperature,space and time);moxibustion intensity(a compound factor achieved through comprehensive control of the three individual basic factors mentioned above);and moxibustion quantity(the amount of temperature stimulation applied within a certain unit of time,including the total amount of moxibustion treatment).The results from present study show that the effect of moxibustion therapy appears to be determined by the activation of TRPV1-4,mainly TRPV1 and TRPV2.Temperature(the degree of heat stimulation),time and area(how long the treatment lasts and how many TRPV1-4 channels are activated)affect the intensity of moxibustion treatment to form effective moxibustion quantity;this should be considered in clinical moxibustion application.

文摘Acupuncture is a medical treatment that has been widely pra cticed in China for over 3000 years,yet the neural mechanisms of acupuncture are not fully understood.We hypothesized that neurons and astrocytes act independently and synergistically under acupuncture stimulation.To investigate this,we used two-photon in vivo calcium reco rding to observe the effects of acupuncture stimulation at ST36(Zusanli)in mice.Acupuncture stimulation in peripheral acupoints potentiated calcium signals of pyramidal neurons and astrocytes in the somatosensory cortex and resulted in late-onset calcium transients in astrocytes.Chemogenetic inhibition of neurons augmented the astrocytic activity.These findings suggest that acupuncture activates neuronal and astrocytic activity in the somatosensory co rtex and provide evidence for the involvement of both neurons and astrocytes in acupuncture treatment.

文摘Injury to the brain after intracerebral hemorrhage(ICH)results from numerous complex cellular mechanisms.At present,effective therapy for ICH is limited and a better understanding of the mechanisms of brain injury is necessary to improve prognosis.There is increasing evidence that ion channel dysregulation occurs at multiple stages in primary and secondary brain injury following ICH.Ion channels such as TWIK-related K+channel 1,sulfonylurea 1 transient receptor potential melastatin 4 and glutamate-gated channels affect ion homeostasis in ICH.They in turn participate in the formation of brain edema,disruption of the blood-brain barrier,and the generation of neurotoxicity.In this review,we summarize the interaction between ions and ion channels,the effects of ion channel dysregulation,and we discuss some therapeutics based on ion-channel modulation following ICH.

文摘Transient receptor potential canonical subfamily member 3(TRPC3) is known to be important for neural development and the formation of neuronal networks. Here, we investigated the role of TRPC3 in undifferentiated mouse embryonic stem cells(mESCs) and during the differentiation of mESCs into neurons. CRISPR/Cas9-mediated knockout(KO) of TRPC3 induced apoptosis and the disruption of mitochondrial membrane potential both in undifferentiated mESCs and in those undergoing neural differentiation. In addition, TRPC3 KO impaired the pluripotency of mESCs. TRPC3 KO also dramatically repressed the neural differentiation of mESCs by inhibiting the expression of markers for neural progenitors, neurons, astrocytes and oligodendrocytes.Taken together, our new data demonstrate an important function of TRPC3 with regards to the survival, pluripotency and neural differentiation of mESCs.

文摘Spodoptera frugiperda is a highly destructive pest that has become a global problem due to its robust reproductive and migratory capabilities.Transient receptor potential(TRP)channels,which constitute a vast ion channel family,play pivotal roles in sensing the external environment and maintaining internal homeostasis in insects.TRP channels have been widely investigated for their critical roles in regulating various insect behaviors in recent years.In this study,we identified 15 TRP gene loci encoding 26 transcripts in the genome of S.frugiperda and analyzed their expression profiles at different developmental stages.The results revealed that S.frugiperda possesses four TRPC genes,six TRPA genes,one TRPM gene,two TRPV genes,one TRPN gene,and one TRPML gene,while a canonical TRPP is absent.Moreover,the SfruTRPA1 was functionally characterized using the Xenopus oocyte expression system.The results showed that SfruTRPA1 is activated by temperature increases from 20 to 45℃,and there is no significant desensitization after repeated stimuli within the same temperature range.Additionally,SfruTRPA1 is activated by certain natural chemicals,including allyl isothiocyanate(AITC)and cinnamaldehyde(CA).These findings provide valuable insights to the TRP genes in S.frugiperda.

文摘The aim of this research was to assess the antinociceptive activity of the transient receptor potential (TRP) channel TRPV1, TRPM8, and TRPA1 antagonists in neurogenic, tonic, and neuropathic pain models in mice. For this purpose, TRP channel antagonists were administered into the dorsal surface of a hind paw 15 min before capsaicin, allyl isothiocyanate (AITC), or formalin. Their antiallodynic and antihyperalgesic efficacies after intraperitoneal ad- ministration were also assessed in a paclitaxel-induced neuropathic pain model. Motor coordination of paclitaxel- treated mice that received these TRP channel antagonists was investigated using the rotarod test. TRPV1 antagonists, capsazepine and SB-366791, attenuated capsaicin-induced nociceptive reaction in a concentration-dependent manner. At 8 pg/20 pl, this effect was 51% (P〈0.001) for capsazepine and 37% (P〈0.05) for SB-366791. A TRPA1 antagonist, A-967079, reduced pain reaction by 48% (P〈0.05) in the AITC test and by 54% (P〈0.001) in the early phase of the formalin test. The test compounds had no influence on the late phase of the formalin test. In paclitaxel-treated mice, they did not attenuate heat hyperalgesia but N-(3-aminopropyl)-2-{[(3-methylphenyl)methyl]oxy}-N-(2-thienylmethyl) benzamide hydrochloride salt (AMTB), a TRPM8 antagonist, reduced cold hyperalgesia and tactile allodynia by 31% (P〈0.05) and 51% (P〈0.01), respectively. HC-030031, a TRPA1 channel antagonist, attenuated tactile allodynia in the von Frey test (62%; P〈0.001). In conclusion, distinct members of TRP channel family are involved in different pain models in mice. Antagonists of TRP channels attenuate nocifensive responses of neurogenic, tonic, and neuropathic pain, but their efficacies strongly depend on the pain model used.

文摘AIM: To explore the possibility of using the Noninvasive Micro-test Technique (NMT) to investigate the role of Transient Receptor Potential Canonical 1 (TRPC1) in regulating Ca^2+ influxes in HL-7702 cells, a normal human liver cell line.METHODS: Net Ca^2+ fluxes were measured with NMT, a technology that can obtain dynamic information of specific/selective ionic/molecular activities on material surfaces, non-invasively. The expression levels of TRPCl were increased by liposomal transfection, whose effectiveness was evaluated by Western-blotting and single cell reverse transcription-polymerase chain reaction.RESULTS: Ca^2+ influxes could be elicited by adding 1 mmol/L CaCl2 to the test solution of HL-7702 cells. They were enhanced by addition of 20 μmol/L noradrenalin and inhibited by 100 μmol/L LaCl3 (a non-selective Ca^2+ channel blocker); 5 μmol/L nifedipine did not induce any change. Overexpression of TRPCl caused increased Ca^2+ influx. Five micromoles per liter nifedipine did not inhibit this elevation, whereas 100 μmol/L LaCI3 did.CONCLUSION: In HL-7702 cells, there is a type of TRPCl-dependent Ca^2+ channel, which could be detected v/a NMT and inhibited by La^3+.
文摘Proton pump inhibitors (PPIs) are commonly used in clinical practice for the prevention and treatment of peptic ulcer, gastritis, esophagitis and gastroesophageal refux. Hypomagnesemia has recently been recognized as a side effect of PPIs. Low magnesium levels may cause symptoms from several systems, some of which being potentially serious, such as tetany, seizures and arrhythmias. It seems that PPIs affect the gastrointesti-nal absorption of magnesium. Clinicians should be vigi-lant in order to timely consider and prevent or reverse hypomagnesemia in patients who take PPIs, especially if they are prone to this electrolyte disorder.

文摘Transient receptor potential(TRP)channels are strongly associated with colon cancer development and progression.This study leveraged a multivariate Cox regression model on publicly available datasets to construct a TRP channels-associated gene signature,with further validation of signature in real world samples from our hospital treated patient samples.Kaplan-Meier(K-M)survival analysis and receiver operating characteristic(ROC)curves were employed to evaluate this gene signature’s predictive accuracy and robustness in both training and testing cohorts,respectively.Additionally,the study utilized the CIBERSORT algorithm and single-sample gene set enrichment analysis to explore the signature’s immune infiltration landscape and underlying functional implications.The support vector machine algorithm was applied to evaluate the signature’s potential in predicting chemotherapy outcomes.The findings unveiled a novel three TRP channels-related gene signature(MCOLN1,TRPM5,and TRPV4)in colon adenocarcinoma(COAD).The ROC and K-M survival curves in the training dataset(AUC=0.761;p=1.58e-05)and testing dataset(AUC=0.699;p=0.004)showed the signature’s robust predictive capability for the overall survival of COAD patients.Analysis of the immune infiltration landscape associated with the signature revealed higher immune infiltration,especially an increased presence of M2 macrophages,in high-risk group patients compared to their low-risk counterparts.High-risk score patients also exhibited potential responsiveness to immune checkpoint inhibitor therapy,evident through increased CD86 and PD-1 expression profiles.Moreover,the TRPM5 gene within the signature was highly expressed in the chemoresistance group(p=0.00095)and associated with poor prognosis(p=0.036)in COAD patients,highlighting its role as a hub gene of chemoresistance.Ultimately,this signature emerged as an independent prognosis factor for COAD patients(p=6.48e-06)and expression of model gene are validated by public data and real-world patients.Overall,this bioi
