Using China New Generation Weather Radar (CINRAD) level-Ⅱ data, the original algorithms for removing isol-ated non-meteorological echoes and ground clutters in radar data, which have been applied to Weather Surveil...Using China New Generation Weather Radar (CINRAD) level-Ⅱ data, the original algorithms for removing isol-ated non-meteorological echoes and ground clutters in radar data, which have been applied to Weather Surveillance Radar-1988 Doppler (WSR-88D) in the USA and Severe Weather Automatic Nowcast (SWAN) system in China, are modified and improved. To remove isolated non-meteorological echoes, the new algorithm introduces a constraint parameter (Po) to distinguish whether a window of 5 x 5 points is isolated as external echoes. A statistical analysis of 150 radar scans (5 cases, with each case comprising 30 scans) under three different echo types (small-scale convec-tion, typhoon, and large-scale synoptic system) shows that the constraint parameter Po ≤ 0.167 is suitable for remov- ing isolated non-meteorological echoes while preserving the edge of meteorological echoes. A new parameter, NDZ, which promotes the ability of the algorithm to identify the ground clutters appearing at two adjacent elevation angles, is constructed based on the vertical continuity of reflectivity. These improved algorithms are tested for four cases (three cases of isolated non-meteorological echoes and one case of ground clutters). Based on the statistics of 232 volume scans of radar data (on a temporal resolution of 1 h) measured at Nanchang station from 0000 UTC 5 to 1600 UTC 14 March 2015, it is found that the improved algorithms not only eliminate most (over 95% under clear-sky conditions) of the isolated non-meteorological echoes and ground clutters (including those appearing at two adjacent elevation angles), but also well preserve the structure of meteorological echoes (storms).展开更多
Radiotherapy remains one of the major treatments for non-small cell lung cancer(NSCLC)patients;whereas intrinsic or acquired radioresistance limits its efficacy.Nevertheless,most studies so far have only focused on ac...Radiotherapy remains one of the major treatments for non-small cell lung cancer(NSCLC)patients;whereas intrinsic or acquired radioresistance limits its efficacy.Nevertheless,most studies so far have only focused on acquired resistance.The exact mechanisms of intrinsic radioresistance in NSCLC are still unclear.A few studies have suggested that epithelial–mesenchymal transition(EMT)is associated with radioresistance in NSCLC.However,little is known about whether the abnormal expression of specific microRNAs induces both EMT and radioresistance.We previously found that miR-410 has multiple roles as an oncomiRNA in NSCLC.In this study,we revealed that miR-410 overexpression promoted EMT and radioresistance,accompanied by enhanced DNA damage repair both in vitro and in vivo.Conversely,knockdown of miR-410 showed the opposite effects.We further demonstrated that PTEN was a direct target of miR-410 by using bioinformatic tools and dual-luciferase reporter assays,and the miR-410-induced EMT and radioresistance were reversed by PI3K,Akt,and mTOR inhibitors or by restoring the expression of PTEN in NSCLC cells.In addition,we preliminarily found that the expression of miR-410 was positively correlated with EMT and negatively associated with the expression of PTEN in NSCLC specimens.In summary,these results demonstrated that miR-410 is an important regulator on enhancing both NSCLC EMT and radioresistance by targeting the PTEN/PI3K/mTOR axis.The findings suggest that miR-410-induced EMT might significantly contribute to the enhanced radioresistance.Therefore,miR-410 may serve as a potential biomarker or therapeutic target for NSCLC radiotherapy.展开更多
FK228 is an FDA-approved anticancer drug naturally produced by Chromobacterium violaceum No.968 up to 19 mg/L in a pilot industry-scale batch fermentation.Here we report a genomics-guided discovery of Burkholderia tha...FK228 is an FDA-approved anticancer drug naturally produced by Chromobacterium violaceum No.968 up to 19 mg/L in a pilot industry-scale batch fermentation.Here we report a genomics-guided discovery of Burkholderia thailandensis MSMB43 as a new and significantly better source of FK228.The genome of B.thailandensis MSMB43 was found to contain a functional biosynthetic gene cluster highly homologous to that of FK228 in C.violaceum No.968,and the bacterium indeed produces authentic FK228.By simple fermentation in shaking flasks in a preferred M8 medium,B.thailandensis MSMB43 produced FK228 up to 67.7 mg/L;by fedbatch fermentation in a 20-L fermentor in M8 medium,B.thailandensis MSMB43 produced FK228 up to 115.9 mg/L,which is 95 fold higher than that of C.violaceum No.968 under the same laboratory fermentation conditions.RT-PCR analysis indicated that the high FK228 yield of B.thailandensis MSMB43 was due to high expression of biosynthetic genes,represented by Bth_depA,during the fermentation process.Further genetic manipulation resulted in a recombinant strain,B.thailandensis MSMB43/pBMTL3-tdpR,which harbors a broad host-range vector expressing the thailandepsin biosynthetic pathway regulatory gene tdpR.This engineered strain produced up to 168.5 mg/L of FK228 in fed-batch fermentation in a 20-L fermentor in M8 medium.Therefore,the wild-type B.thailandensis MSMB43 or its engineered derivative could potentially be a good starting point for an industrial process to improve FK228 production for its expanding use in therapy.展开更多

In this behavioral genetic study on conformity,we designed a social episode to investigate the genetic and environmental origin of individual differences in conforming behavior during psychosocial development.Specifically,107 monozygotic(MZ) and 74 dizygotic(DZ) twin pairs(age 7-19) were asked to choose a pen from a group of pens either with the majority color or with the minority color.We found that the resemblance between MZ twins in selecting the pen with the majority color was not significantly higher than that between DZ twins,suggesting that individual differences in conformity are due almost exclusively to the environment.Moreover,biometric model fitting revealed that the shared environmental component played a sizable role in shaping individuals' conforming behavior,and the influence increased with age(from 16% to 26%).Taken together,our study suggests that the common family and cultural environment has important ramifications for conformity.展开更多
