RVA and Farinograph Properties Study on Blends of Resistant Starch and Wheat Flour 被引量:18

Rapid visco anal... Resistant starch (RS) is the undigested starch that passes through the small intestine to the large intestine. As a functional low calorie additive, it has special applications in the food industry. Rapid visco analysis (RVA) and the Brabender farinograph were used to study the pasting properties and the viscoelasticity of blends of RS (RS3 and RS2) and three wheat flours. The wheat flours represented strong gluten wheat (SGW), intermediate gluten wheat (IGW), and weak gluten wheat (WGW) flours, at different levels of RS substitution (0, 5, 10, 15, and 20%). The influence of RS3 on the control wheat flours and RS-wheat flour blends were consistent with those of RS2. The peak, trough, and final viscosities of RS3-wheat flour blends were higher than those of the corresponding RS2-wheat flour blends. The peak, trough, breakdown, final, and setback viscosities ofwheat-RS blends decreased with an increase in resistant starch contents from 0 to 20% in the blends. The 0-20% RS-wheat flour blends were all able to form doughs. The dough development times, dough stabilities, dough breakdown times, and farinograph quality numbers for the RS-wheat flour blends decreased as the RS proportion in the blends increased. The values for RS-SGW flour blends were the highest, followed by RS-IGW and then RS-WGW flour blends. The water absorption values for RS-wheat flour blends and the mixing tolerance index for RS-WGW flour blends were found to increase significantly with an increasing proportion of RS from 0 to 20%, but the mixing tolerance index for RS-SGW and RS-IGW flour blends showed no significant differences amongst the different ratios. Correlation analysis showed that the Farinograph quality number was highly positively correlated with dough breakdown time, dough stability, and dough development time (r= 1.000, 0.958, 0.894), and highly negatively correlated with the mixing tolerance index (r =-0.890). A set of near-isogenic lines (NILs) of rice which has single resistance gene for each line was developed by use of LTH as recurrent parent and Kusabue,Tsuyuake, K1, PiNo. 4, K60 and BL1, which are from Kiyosawa's differential varieties(KDV), as donor parents. Differentiating ability of these NILs to Philippine isolates was compared with that of their donor parents, KDV and IRRI' s NILs. The results of comparative studies were as follows: (1) Differentiating ability of Chinese NILs to Philippine isolates was much stronger than that of their donor parents and that of KDV. (2) Chinese NILs had similar ability to differentiate races to IRRI's NILs or higher differentiating ability than them. But IRRI's NILs almost had no differentiating ability to isolates from japonica rice-growing regions. They can not be used to differentiate races of isolates from japonica ricegrowing regions. Whereas, Chinese NILs can be used to differentiate races of isolates from both japonica ricegrowing regions and indica rice-growing regions. Dehydroascorbate reductase (DHAR) plays a critical role in the ascorbate-glutathione recycling reaction for most higher plants. To date, studies on DHAR in higher plants have focused largely on Arabidopsis and agricultural plants, and there is virtually no information on the molecular characteristics of DHAR in gymnosperms. The present study reports the cloning and characteristics of a DHAR (PbDHAR) from a pine, Pinus bungeana Zucc. ex Endl. The PbDHAR gene encodes a protein of 215 amino acid residues with a calculated molecular mass of 24.26 kDa. The predicted 3-D structure of PbDHAR showed a typical glutathione S-transferase fold. Reverse transcripUon-polymerase chain reaction revealed that the PbDHAR was a constitutive expression gene in P. bungeana. The expression level of PbDHAR mRNA in P. bungeana seedlings did not show significant change under high temperature stress. The recombinant PbDHAR was overexpressed in Escherichia coil following purification with affinity chromatography. The recombinant PbDHAR exhibited enzymatic activity (19.84 i.mnol/min per mg) and high affinity (a Krn of 0.08 mM) towards the substrates dehydroascorbate (DHA). Moreover, the recombinant PbDHAR was a thermostable enzyme, and retained 77% of its initial activity at 55℃. To evaluate the possible genetic interrelationships between flour components and the sedimentation volume(SD),a doubled haploid(DH) population comprising 168 lines were used to identify the conditional quantitative trait loci(QTLs) for SD in three environments.Ten additive QTLs and 15 pairs of epistatic QTLs were detected for SD through unconditional and conditional QTL mapping.Three major additive QTLs were detected for SD conditioned on the seven quality traits.Two additive QTLs were found to be independent of these traits.Three additive QTLs were suppressed by three of the seven traits because of non-detection in unconditional mapping.Three pairs of epistatic QTLs were completely affected by the seven traits because of detection in unconditional mapping but no-detection in conditional mapping.Twelve pairs of epistatic QTLs were detected in conditional mapping.Our results indicated that conditional mapping could contribute to a better understanding of the interdependence of different and closely correlated traits at the QTL molecular level,especially some minor QTLs were found.The conditional mapping approach provides new insights that will make it possible to avoid the disadvantages of different traits by breeding through molecular design. 展开更多

BACKGROUND Difficulty in obtaining tetracycline,increased adverse reactions,and relatively complicated medication methods have limited the clinical application of the classic bismuth quadruple therapy.Therefore,the search for new alternative drugs has become one of the research hotspots.In recent years,minocycline,as a semisynthetic tetracycline,has demonstrated good potential for eradicating Helicobacter pylori(H.pylori)infection,but the systematic evaluation of its role remains lacking.AIM To explore the efficacy,safety,and compliance of minocycline in eradicating H.pylori infection.METHODS We comprehensively retrieved the electronic databases of PubMed,Embase,Web of Science,China National Knowledge Infrastructure,SinoMed,and Wanfang database as of October 30,2023,and finally included 22 research reports on H.pylori eradication with minocycline-containing regimens as per the inclusion and exclusion criteria.The eradication rates of H.pylori were calculated using a fixed or a random effect model,and the heterogeneity and publication bias of the studies were measured.RESULTS The single-arm meta-analysis revealed that the minocycline-containing regimens achieved good overall H.pylori eradication rates,reaching 82.3%[95%confidence interval(CI):79.7%-85.1%]in the intention-to-treat analysis and 90.0%(95%CI:87.7%-92.4%)in the per-protocol analysis.The overall safety and compliance of the minocycline-containing regimens were good,demonstrating an overall incidence of adverse reactions of 36.5%(95%CI:31.5%-42.2%).Further by traditional meta-analysis,the results showed that the minocycline-containing regimens were not statistically different from other commonly used eradication regimens in eradication rate and incidence of adverse effects.Most of the adverse reactions were mild to moderate and well-tolerated,and dizziness was relatively prominent in the minocycline-containing regimens(16%).CONCLUSION The minocycline-containing regimens demonstrated good efficacy,safety,and compliance in H.pylori eradication.Minocycline 展开更多

Background: Triclosan [5-chloro-2-(2,4-dichlorophenoxy) phenol, TCS], a common antimicrobial additive in many personal care and health care products, is frequently detected in human blood and urine. Therefore, it has been considered an emerging and potentially toxic pollutant in recent years. Long-term exposure to TCS has been suggested to exert endocrine disruption effects, and promote liver fibrogenesis and tumorigenesis. This study was aimed at clarifying the underlying cellular and molecular mechanisms of hepatotoxicity effect of TCS at the initiation stage.Methods: C57BL/6 mice were exposed to different dosages of TCS for 2 weeks and the organ toxicity was evaluated by various measurements including complete blood count, histological analysis and TCS quantification. Single cell RNA sequencing(scRNA-seq) was then carried out on TCS-or mock-treated mice livers to delineate the TCS-induced hepatotoxicity. The acquired single-cell transcriptomic data were analyzed from different aspects including differential gene expression, transcription factor(TF) regulatory network, pseudotime trajectory, and cellular communication, to systematically dissect the cellular and molecular events after TCS exposure. To verify the TCS-induced liver fibrosis,the expression levels of key fibrogenic proteins were examined by Western blotting, immunofluorescence, Masson’s trichrome and Sirius red stainings. In addition, normal hepatocyte cell MIHA and hepatic stellate cell LX-2 were used as in vitro cell models to experimentally validate the effects of TCS by immunological, proteomic and metabolomic technologies.Results: We established a relatively short term TCS exposure murine model and found the TCS mainly accumulated in the liver. The scRNA-seq performed on the livers of the TCS-treated and control groups profiled the gene expressions of > 76,000 cells belonging to 13 major cell types. Gibberellic acid(GA3)is a vital plant growth hormone widely used in agriculture.Currently,GA3 production relies on liquid fermentation by the filamentous fungus Fusarium fujikuroi.However,the lack of an effective selection marker recycling system hampers the application of metabolic engineering technology in F.fujikuroi,as multiple-gene editing and positive-strain screening still rely on a limited number of antibiotics.In this study,we developed a strategy using pyr4-blaster and CRISPR/Cas9 tools for recycling orotidine-5'-phosphate decarboxylase(Pyr4)selection markers.We demonstrated the effectiveness of this method for iterative gene integration and large gene-cluster deletion.We also successfully improved GA3 titers by overexpressing geranylgeranyl pyrophosphate synthase and truncated 3-hydroxy-3-methyl glutaryl coenzyme A reductase,which rewired the GA3 biosynthesis pathway.These results highlight the efficiency of our established system in recycling selection markers during iterative gene editing events.Moreover,the selection marker recycling system lays the foundation for further research on metabolic engineering for GA3 industrial production. 展开更多

Objective:To assess the clinical outcomes of fertility-sparing treatments in young patients with epithelial ovarian carcinoma (EOC).Methods:A retrospective study of young EOC inpatients (≤40 years old) was performed during January 1994 and December 2010 in eight institutions.Results:Data were analyzed from 94 patients treated with fertility-sparing surgery with a median follow-up time of 58.7 months.As histologic grade increased,overall survival (OS) and disease-free survival (DFS) of patients receiving fertility-sparing surgery declined.Neither staging surgery nor laparoscopy of early stage EOC with conservative surgery had a significant effect on OS or DFS.Normal menstruation recommenced after chemotherapy in 89% of the fertility-sparing group.Seventeen pregnancies among twelve patients were achieved by the end of the follow-ups.Conclusions:Fertility-sparing treatment for patients with EOC Stage I Grade 1 could be cautiously considered for young patients.The surgical procedure and surgical route might not significantly influence the prognosis.Standard chemotherapy is not likely to have an evident impact on ovarian function or fertility in young patients. 展开更多

A quinoline derivative N-(2-hydroxyl-naphthylmethyl)-N-(quinol-8-yl)amine(2) was synthesized and characterized, which can selectively recognize Cu^2+ over other metal ions such as Zn^2+, Cd^2+, Sn^2+, Pb^2+, Hg^2+, Ni^2+, Mg^2+, Cr^3+, Mn^2+, Sr^2+, K^+, Ca^2+, Na^+, Ba^2+ in acetonitrile/water(volume ratio 99:1). Cu^2+ induced a new absorption peak at 464 nm in the absorption spectrum(ε=4.66×10^2 L·mol^-1·cm^-1) and quenched the fluorescence emission of quinoline derivative 2. The binding of quinoline derivative 2 to Cu^2+ was in 1 : 1 model and stable constant was esti- mated to be 2.48×10^5 L·mol^-1. 展开更多

Structural investigations of a wheat tau GST (TaGSTU4) show two subunit interactions: a hydrogen bond between the Tyr93 and Pro65 from another subunit of the dimer, and two salt bridges between residues Glu78 and side chains of Arg95 and Arg99 in the opposite subunit. By investigating enzyme activities, kinetic parameters and structural characterizations, this study showed the following results: (i) the hydrogen bond interaction between the Tyr93 and Pro65 was not essential for dimerization, but contributed to the enzyme's catalytic activity, thermal stability and affinity towards substrates glutathione and 1-chloro-2, 4-dinitrobenzene; and (ii) two salt bridges mainly contributed to the protein structure stability and catalysis. The results of this study form a structural and functional basis for rational design of more selective and environmentally friendly herbicides. 展开更多

<strong>Background:</strong> In the present study, autophagy-related long non-coding RNAs (lncRNAs) in lung adenocarcinoma (LUAD) were screened for diagnosis and prognosis, and the molecular mechanisms of ... <strong>Background:</strong> In the present study, autophagy-related long non-coding RNAs (lncRNAs) in lung adenocarcinoma (LUAD) were screened for diagnosis and prognosis, and the molecular mechanisms of LUAD at the genetic level were investigated. <strong>Methods:</strong> From The Cancer Genome Atlas (TCGA) database, 497 gene expression data and 436 clinical data of LUAD cases were collected for analysis. In addition, 232 autophagy-related genes (ARGs) were extracted from the Human Autophagy Database (HADb). Spearman rank correlation test and the Akaike information criterion (AIC) were performed to screen the data. After filtering, a survival model including three autophagy-related lncRNAs was generated. Based on the following formula: risk score = ΣCoef gene i×Gene i expression, the risk score of all LUAD patients could be calculated. LUAD patients were divided into two groups based on risk score for survival curve using Kaplan-Meier survival analysis. Both univariate and multivariate survival analyses were used to determine whether the three lncRNAs were independent prognostic factors using the survival package in R. Furthermore, the receiver operating characteristic (ROC) curves of clinical data were created to assess the stability of the survival model. Finally, the Gene Set Enrichment Analysis (GSEA) was used for analysis of related pathways. <strong>Results:</strong> A prognostic model consisting of three lncRNAs (AC011477.2, AC099850.3, and TRG-AS1) was generated for analysis. The 5-year survival rate in the high-risk group was 26.51% (95% CI: 0.1842 - 0.382), which was statistically lower than in the low-risk group 41.6% (95% CI: 0.307 - 0.563, P < 0.05). The area under the ROC curve (AUC) of risk score was 0.700, indicating a higher diagnostic accuracy of risk score. Objective:To explore the impacts of bone marrow mesenchymal stem cells(BMSCs)intervention on the neuronal withering and autophagy in rats after subarachnoid hemorrhage(SAH)in rats and its process.Methods:forty-eight SD rats(males),were randomly assigned to the followings:sham surgery(Sham)group,modeling(SAH)group,modeling group+bone marrow mesenchymal stem cells(BMSCs)group,the modeling group+BMSCs+AMPK inhibitor(Compound C)group,twelve rats in each group.Except Sham group,other groups formed SAH samples with intravascular points.Separate groups Compound C and BMSCs,respectively Compound C and 10μL of normal saline were injected into the left lateral ventricle of rats using a stereoscope brain machine 30 minutes before modeling,and 2μL concentration is 1×10^(8)/mL cell suspension was injected into the ventricles of the brain 1 hour after the model was established.Observe whether the rats have severe brain swelling.Garcia were used to test the neural function of rats.TUNEL staining was used to monitor neuronal apoptosis in the rat hippocampal gyrus.Immunohistochemical fluorescence reflected the expression of two proteins,LC3-II and Beclin-1,in rat hippocampal gyrus.Western blotting is applied to measure the expressions of autophagy-associated proteins in the AMPK pathway.Results:Compared with the group undergoing sham-surgery,brain edema worsened in the model group,neuronal apoptosis rate was increased,neural function was weakened,Protein granules decreased(P<0.05)and expression levels of p-AMPK and other proteins decreased(P<0.05);brain swelling and neuronal apoptosis were reduced in the BMSCs group by comparison with the modeling group’s,with substantial elevation in the expression of proteins comprising LC3-Ⅱ,Beclin-1,and p-AMPK.And the standard of p-mTOR protein expression was considerably lessened(P<0.05);rats belonging to group compound C showed increased brain swelling which is relative to that of BMSCs group,increased neuronal apoptosis,decreased neuronal function(P<0.05),increased p-AMPK protein ex 展开更多

Background Reoperation for total colonic aganglionosis(TCA)may be required for residual aganglionosis after an initial radical operation.We aimed to investigate the symptoms,management,and outcomes of patients who required a redo pullthrough(Redo PT).Methods Nine TCA patients underwent Redo PT at our center between 2007 and 2017.Their medical records were reviewed.Parental telephone interviews that included disease-specific clinical outcomes were conducted,and post-operative complications and long-termoutcomes(including height-for-age/weight-for-age and bowel-function score)were compared to those of single-pull-through(Single PT)patients(n=21).Results All the nine Redo PT patients suffered obstruction within 1 month after the initial operation that could not be alleviated by conservative treatment.All abdominal X-ray/contrast barium enemas showed proximal bowel dilatation,indicating residual aganglionosis.The median ages at the initial operation and Redo PT were 200 and 509 days,respectively.Reoperation consisted of an intraoperative frozen biopsy and a modified laparotomic Soave procedure in all patients.Postoperative complications included perianal excoriation(n=3),intestinal obstruction(n=2),enterocolitis(n=2),and rectovestibular fistula(n=1).Seven Redo PT patients were followed up for a mean time of 7.162.3 years;six(85.7%)had good growth and four(57.1%)had good bowel-function recovery.Post-operative complications and long-term outcomes were almost equal between the Redo PT and Single PT groups(all P>0.05).Conclusion TCA patients with recurrent obstructive symptoms and dilated proximal bowel may have residual aganglionosis after an initial operation.Redo PT is effective and provides good long-termoutcomes comparable to those of patients who benefited from Single PT. 展开更多

Objective:To study the effect of intense pulsed light autopulse combined with oral metronidazole on serum inflammatory mediators in patients with mild and moderate acne. Methods: A total of 84 patients who were diagnosed with mild-to-moderate acne in our hospital between May 2013 and December 2015 were selected and randomly divided into laser group and metronidazole group who received intense pulsed light autopulse combined with oral metronidazole therapy and oral metronidazole monotherapy respectively. Before treatment as well as 1 week and 2 weeks after treatment, the serum was collected, fluorescent quantitative PCR kits were used to determine the expression of TLR molecules and NLRP inflammasome, and enzyme-linked immunosorbent assay kits were used to determine the levels of inflammatory cytokines.Results: 2 weeks and 4 weeks after treatment, serum IL-1β, IL-8, IL-12 and IL-18 levels as well as TLR2, TLR4, NLRP3, ASC and Caspase-1 mRNA expression of both groups of patients were significantly lower than those before treatment, and serum IL-1β, IL-8, IL-12 and IL-18 levels as well as TLR2, TLR4, NLRP3, ASC and Caspase-1 mRNA expression of laser group were significantly lower than those of metronidazole group.Conclusion: Intense pulsed light autopulse combined with oral metronidazole has significant inhibitory effect on the inflammatory response mediated by pattern recognition receptors TLR2, TLR4 and NLRP3 in patients with mild and moderate acne. 展开更多

Objective To investigate the aberrant expressions of TNF-α and TGF-β1 in peripheral blood mononuclear cells (PBMCs) and placental tissues in patients with early spontaneous abortion Methods Using the technique of semi-quantitative reverse transcript-polymerase chain reaction (RT-PCR), TNF-α mRNA and TGF-β1 mRNA in PBMCs were measured in spontaneous abortion group (30 cases), normal pregnancy group (25 cases) and nonpregnant group (25 cases). The expressive intension of TNF-α protein and TGF-β1 protein in placental tissues was also identified by immunohistochemistry. Results Both levels of TNF-α mRNA and TGF-β1 mRNA expressed in PBMCs were significantly different between the three groups respectively (P〈0.05). Levels of TNF-α in syncytiotrophoblastic and cytotrophoblastic cells of the two aborted groups were substantially higher than those of the nonregnant group (P〈0.01), but the levels of TGF-β1 in syncytiotrophoblastic cells of the two aborted groups were markedly lower than those of the non-pregnant group (P〈0.01). Conclusion There is potential relation between TGF-β at the fetomaternal interface and spontaneous abortion. TGF-β1 may contribute to the maintenance of pregnancy, and low-level expression of TGF-β1 may be associated with pregnancy failure. 展开更多

In order to provide a scientific basis for laying out a unified standard of the reference value (Wintrobe) of Chinese people's erythrocyte sedimentation rate (ESR). this paper studies the relationship between the reference values (Wintrobe) of Chinese healthy people's ESR, and the test is done according to the Wintrobe Laws and five geographical factors. It is found that the altitude is the modt important factor affecting the reference value (Wintrobe) of Chinese people's ESR. As altitude increases gradually, the reference value (Wintrobe) of people's ESR decreases accordingly, the correlation is quite obvious. By using the method of regression analysis, two multivariate regression equations and ten univariate regression equations are deduced If the geographical index values in a particular area are known, the reference value (Wintrobe) of Chinese people's ESR of this region can be established by means of the regression equations. An active miniature inverted-repeat transposable element(MITE),MCL as-A,was previously identified from'Candidatus Liberibacter asiaticus'known to be associated with citrus Huanglongbing(HL B,yellow shoot disease).To explore the recent transposition status of MCLas-A,389'Ca.L.asiaticus'strains collected from nine regions in China were amplifed using a specific primer set and three representative'Ca.L.asiaticus'strains were analyzed by next-generation sequencing(NGS)approach.PCR and genomic analysis showed that the entire MCLas A was only present in 1.80%(7/389)and the jumping-out type of the MITE was predominant(8123%)in samples tested,suggesting high frequency transposition occurred in'Ca.L.asiaticus'strains recently collected from China.Biological roles of transposition of the active MITE remain to be determined. 展开更多

An outbreak of powerful tornadoes tore through multiple states in the central and southern United States from 10 to 11 December 2021.It is claimed the deadliest tornado outbreak that has taken place on December days.The National Oceanic and Atmospheric Administration had confirmed 66 tornadoes as of 21 December,producing at least 90 fatalities.Most tornadoes occurred at night and thus they were difficult to be visually located,which directly increases the risk for local residents.Two violent nighttime tornadoes were rated category 4 on the enhanced Fujita scale(EF4).Although a high death toll was caused during this event,the operational service actually presented an excellent performance.This tornado outbreak has aroused extensive discussion from both the public and the research community in China.This paper presents a brief discussion on the formation environment and warning services of the tornado outbreak.Recall the deadliest violent tornado in the past 45 years in China,the radar-based tornadic vortex signatures at the locations with EF4 damages show a comparable strength with those in the current cases.Some views on the tornado warning issuance and receiving and damage surveys in China are also presented. 展开更多

Objective:To explore the value of serum amyloid A content in the diagnosis of gastric cancer in patients with gastric cancer.Methods:The serum of 120 gastric cancer patients and 30 healthy physical examination patients admitted to Hainan Provincial Hospital of Traditional Chinese Medicine from September 2015 to December 2017 were selected as experimental subjects.Results:The expression of SAA1 in serum of patients with gastric cancer was significantly higher than healthy subjects(42.38±10.12)μg/mL vs.(22.70±8.98)μg/mL,and the difference was statistically significant(t=-9.725,P<0.001).ROC curve analysis showed that the AUC of SAA1 diagnosis of gastric cancer was 0.907(95%CI:0.838-0.976),the detection threshold was 29.02μg/mL,and the diagnostic sensitivity and specificity were 93.3%and 86.7%,respectively.The expression level of serum SAA1 was not correlated with gender(r=0.056,P>0.05),age(r=-0.032,P>0.05)and tumor metastasis(r=-0.173,P>0.05)of gastric cancer patients,and correlated with tumor size(r=-0.228,P<0.05)and invasion degree(r=-0.409,P<0.05),and increased with the increase of tumor invasion degree.Conclusion:The expression level of serum SAA1 in gastric cancer patients is significantly increased,which may be a new potential marker for the diagnosis of gastric cancer. 展开更多
