摘要
西红花苷属于脱辅基类胡萝卜素化合物,是中国传统珍贵药材西红花(藏红花)的主要活性成分。CCDs(类胡萝卜素裂解双加氧酶)是催化玉米黄质形成西红花苷等脱辅基类胡萝卜素的重要酶,为了解西红花CCD家族基因的差异,本研究从NCBI数据库中检索获得11个西红花CCD家族基因,对基因序列及表达特性进行分析。结果表明,西红花CsCCD家族基因包括CCD1、CCD4、CCD7、CCD8 4个亚基因家族,各个亚家族的基因序列较为保守、编码的蛋白质三维结构存在差异。通过表达分析发现不同CsCCD基因在不同花器官中的表达特性存在明显差异,这一结果与通过转录组测序得到的CsCCD基因表达水平相符。推测不同CsCCD基因在西红花苷合成过程中的功能存在差异。
Crocin is an apocarotenoid compound, the major active ingredient of saffron (Crocus sativus) which has been cultivated and used as spice and medicinal plant for thousands of years.Carotenoid cleavage dioxygenases (CCDs) are important enzymes that catalyze zeaxantin to generate crocin in Crocus.To understand the difference of CCD genes in Crocus, 11 CsCCD genes searched from NCBI were analyzed for the sequence, structure and expression.The 11 CsCCD genes were divided into four subfamilies(CCD1, CCD4, CCD7, and CCD8) which shared similar conserved domains and different 3D structure of encoded protein.The expressions of CsCCD genes differed in different floral organs (stigma, petal and stamen), which were consistent with the expression patterns of CsCCD gene obtained by the transcription sequencing.It is speculated that different CsCCD genes plays different roles in crocin biosynthesis.
