摘要
目的 :研究α-硫辛酸对脑缺血再灌注大鼠凋亡调控基因的表达及炎性因子的影响,为临床防治提供参考。方法 :选择清洁级的108只健康SD雄性大鼠作为研究对象,根据随机数表法分为模型组(脑缺血再灌注组)、假手术组、干预组(α-硫辛酸预干预组),各36只。采用线栓法制定大脑中动脉闭塞2h后的再灌注模型,并在再灌注后的不同时间分成6、12、24、48h后应用TTC染色法对脑梗死体积进行检测,并开展神经功能评分,应用原位杂交及免疫印迹法对各组大鼠的凋亡调控基因Bax mRNA、JNK3表达水平进行检测,利用免疫组织化学法、原位杂交法对各组大鼠的NF-k B、TNF-α表达水平进行检测。结果 :干预组大鼠再灌注后6h、12h、24h、48h的神经功能缺损评分均低于模型组,差异有统计学意义;干预组大鼠再灌注后6h、12h、24h、48h的脑梗死体积均低于模型组,差异有统计学意义;假手术组大鼠再灌注后6h、12h、24h、48h的细胞凋亡计数均低于模型组、干预组,且干预组低于模型组,差异有统计学意义;假手术组大鼠再灌注后6h、12h、24h、48h的凋亡调控基因Bax mRNA、JNK3表达水平均低于模型组、干预组,且干预组低于模型组,差异有统计学意义;假手术组大鼠再灌注后6h、12h、24h、48h的炎性因子NF-k B、TNF-α表达水平均低于模型组、干预组,且干预组低于模型组,差异有统计学意义。结论 :α-硫辛酸能对大鼠脑缺血再灌注后出现的神经元凋亡进行有效抑制,且能对大鼠脑缺血再灌注损伤进行保护,进一步减轻脑缺血再灌注损伤。
Objective To study the effect of α-lipoic acid on the expression of apoptosis-regulating genes and inflammatory factors in cerebral ischemia-reperfusion rats, and to provide reference for clinical prevention and treatment. Methods 108 cases of healthy SD male rats were randomly divided into model group(cerebral ischemia reperfusion group), sham operation group and intervention group(α-lipoic acid pre-intervention group) according to the random number table method, each group of 36 cases. The model of reperfusion was established after 2 h of middle cerebral artery occlusion. The infarct volume was measured by TTC staining at 6, 12, 24, 48 h after reperfusion at different time points, and the neurological function score was performed. The expressions of Bax mRNA and JNK3 were detected by in situ hybridization and immunoblotting. The expression of Bax mRNA and JNK3 in each group was detected by immunohistochemistry and in situ hybridization. The expressions of NF-Expression levels were tested. Results The scores of neurological deficits were significantly lower in the intervention group than those in the model group at 6 h, 12 h, 24 h and 48 h after reperfusion. At 6 h, 12 h, 24 h after reperfusion. The apoptotic count of the sham operation group was lower than that of the model group at 6 h, 12 h, 24 h and 48 h after reperfusion, and the intervention group was significantly lower than that of the model group. The expression of Bax mRNA and JNK3 in the sham operation group at 6 h, 12 h, 24 h and 48 h after reperfusion were lower than those in the model group, and the difference was statistically significant. The expression of NF-k B and TNF-α in the sham operation group at 6 h, 12 h, 24 h and 48 h after reperfusion were significantly lower than those in the intervention group. The expression level of the intervention group was lower than that of the model group and the intervention group, and the intervention group was lower than the model group, the difference was statistically significant.
