摘要
铁死亡是指通过铁过载产生活性氧,使脂质、核酸和蛋白质受到损坏最终导致细胞程序性死亡的过程,作为一种新型的调节性细胞死亡(RCD)模式,近年来受到许多关注。肝细胞癌(hepatocellutar carcinoma, HCC)是世界范围内的预后相对较差的恶性肿瘤之一,我国又是肝癌高发国。近年来有研究发现HCC细胞与正常细胞存在更多的生理性差异,并且发现这些差异都是参与铁死亡的关键调控因素,可见铁死亡过程是影响HCC的发生与发展的重要因素。本文汇总近年来铁死亡发生机制及其在HCC中的作用研究进展,并发散思维,试图为HCC的治疗探索新的干预靶点。
Ferroptosis is a newly identified iron dependent regulatory cell death (RCD) mode, relies on reac-tive oxygen species produced by iron overload to damage lipids, nucleic acids, and proteins, result-ing in programmed cell death. Hepatocellular carcinoma (HCC) is recognized as one of the most ma-lignant tumors with high degree of malignancy and relatively poor prognosis. Recent studies have found that intracellular iron metabolism disorder, lipid peroxide accumulation, and amino acid an-tioxidant system imbalance are all physiological differences between HCC cells and normal cells, and these differences are key regulatory factors of iron death, so iron death plays an important role in the occurrence and development of HCC. This article reviews the mechanism of iron death and its role in HCC, in an attempt to explore new intervention targets for the treatment of HCC.
