铁死亡是氧化细胞性死亡的一种形式,其特征是积累的铁和脂质过氧化产生ROS,从而触发死亡。铁死亡的发生和执行受到多种因素的影响,包括铁代谢、氨基酸代谢、脂质代谢、谷胱甘肽代谢和甲戊酸途径。在多种疾病过程中,如缺血再灌注损伤、退行性病变和肿瘤等,铁死亡被发现参与其中。研究表明,铁死亡在妇科恶性肿瘤的发生和发展中也扮演关键角色,并且利用铁死亡可以增强放疗、化疗、靶向治疗和免疫治疗的敏感性。因此,靶向铁死亡可能成为治疗肿瘤的一种新兴治疗策略。本文综述了铁死亡的发生机制以及在妇科恶性肿瘤研究方面的最新进展。
Ferroptosis is a form of oxidative cellular death characterised by the generation of ROS from perox-idation of accumulated iron and lipids, which triggers death. The onset and execution of ferroptosis is influenced by multiple factors, including iron metabolism, amino acid metabolism, lipid metabo-lism, glutathione metabolism, and the mevalonate pathway. Ferroptosis has been found to be in-volved in a variety of disease processes such as ischaemia-reperfusion injury, degenerative lesions and tumours. Studies have shown that ferroptosis also plays a key role in the development and progression of gynaecological malignancies, and that the use of ferroptosis enhances the sensitivity of radiotherapy, chemotherapy, targeted therapy and immunotherapy. Therefore, targeting fer-roptosis may become an emerging therapeutic strategy for the treatment of tumours. This article reviews the mechanisms of ferroptosis and recent advances in the study of gynaecological malig-nancies.
