硫酸软骨素在小鼠烟曲霉菌性角膜炎中的抗炎作用

摘要 硫酸软骨素(chondroitin sulfate, CS)是一种天然存在的糖胺聚糖,主要存在于软骨,皮肤,血管,韧带和肌腱中。目前研究发现CS具有抗炎、免疫调节、抗氧化、抗肿瘤等多种药理学活性。但是在真菌性角膜炎中尚未有研究,本文就CS在小鼠烟曲霉菌性角膜炎中的治疗作用进行相关研究。方法:体外实验用烟曲霉菌感染巨噬细胞后,用CS处理,用PCR方法检测TLR-2、Dectin-1、LOX-1的mRNA的表达。体内实验用烟曲霉菌孢子感染C57BL/6小鼠角膜制造烟曲霉菌角膜炎模型,然后使用CS或PBS处理,用PCR方法检测TLR-2、Dectin-1、LOX-1的mRNA的表达。结果:CS降低了TLR-2、Dectin-1、LOX-1的mRNA表达,减轻了烟曲霉菌性角膜炎的炎症反应。结论:CS通过抑制模式识别受体的表达减轻小鼠烟曲霉菌性角膜炎的炎症反应。 Chondroitin sulfate (CS) is a naturally occurring glycosaminoglycan, which is mainly found in cartilage and skin, BLOOD vessels, ligaments and tendons. Studies have found that chondroitin sulfate has anti-inflammatory, immune regulation, anti-oxidation, anti-tumor and other pharmacological activities. However, it has not been studied in fungal keratitis. This article aims to study the therapeutic effect of chondroitin sulfate in mice with Aspergillus fumigatus keratitis. Methods: In vitro experiments, macrophages were infected with Aspergillus fumigatus and treated with chondroitin sulfate. PCR was used to detect the mRNA expression of TLR-2, Dectin-1, and LOX-1. In vivo experiments, the cornea of C57BL/6 mice were infected with Aspergillus fumigatus spores to make Aspergillus fumigatus keratitis model, then treated with chondroitin sulfate or PBS. PCR was used to detect the mRNA expression of TLR-2, Dectin-1, and LOX-1. Results: Chondroitin sulfate reduced the inflammatory response of keratitis by reducing the mRNA of TLR-2, Dectin-1, and LOX-1. Conclusion: Chondroitin sulfate reduces the inflammatory response of mouse keratitis by inhibitingthe expression of pattern recognition receptors.
