摘要
目的探讨组织工程骨的体内血管化情况和修复大节段骨缺损的能力。方法采用骨髓穿刺、密度梯度离心的方法获取兔骨髓基质细胞,经体外诱导分化,接种到聚磷酸钙纤维/L-聚乳酸/胶原(CPPf/PLLA/Collagen)支架材料上,植入自体尺骨中下段1.5cm长的骨膜骨缺损区,对照组为缺损区单纯植入CPPf/PLLA/Collagen,术后观察12周。结果术后4周缺损区为新生骨样组织充填,血循环丰富,12周时形态结构已接近正常骨组织;随修复时间延长,修复组织的力学性能逐渐增强。结论骨髓基质细胞与CPPf/PLLA/Collagen复合物具有较强的成骨能力,有望成为修复骨缺损的治疗方法。
Objective To evaluate the vascularization and capability of MSCs CPPf/PLLA/ Collagen composites in repair of large segmental bone defects of rabbits. Methods The MSCs of rabbits were harvested and cultured in osteogenic conditions. The MSCs were collected and seeded on CPPf/PLLA/ Collagen scaffold. The composites of MSCs CPPf/PLLA/ Collagen were implanted into the segmental bong defects of rabbit ulna 1.5cm in length. Results The radiographic and histological examination showed that all the defects healed at 12 weeks. The vascularization and biomechanical examination showed that the behavior of new bone was similar to that of normal bone. Conclusion Since the MSCs CPPf/PLLA/Collagen can repair segmental bone defects, it can be used in the clinical treatment of bone defects.
