4种思茅松总DNA提取方法的比较 被引量:17

摘要 以思茅松(Pinuskesiyavarlangbianensis)针叶为材料,分别采取了简易提取法、高盐沉淀法、CTAB沉淀法和高盐低pH法提取基因组DNA,并通过琼脂糖凝胶电泳、限制性内切酶处理和RAPD3种方法对所提取的DNA样品进行检测,将它们在DNA产量、质量等方面的优缺点进行总结。 Different methods,i.e. basic method,high salt precipitation,CTAB precipitation,low pH extraction medium with high salt,were used to extract genomic DNA from the needle leaves of Pinus kesiya var langbianensis.The DNA samples obtained by the above methods were tested by agarose gel electrophoresis,restriction digests and PCR.The difference in yield,quality of the DNA samples was compared,and high salt precipitation was considered to be a fast,inexpensive and reliable technique among the four methods.
