摘要
以引进的速生型刺槐的复叶为实验材料 ,进行离体培养 ,研究生长调节物质对刺槐复叶的小叶和叶轴再生的影响。以MS为基本培养基 ,对IAA、IBA、NAA、2 ,4 D 4种生长素 ,及BA、KT、ZT 3种细胞分裂素进行单因子试验 ,对比并筛选出对刺槐小叶和叶轴再生最好的生长素和细胞分裂素为NAA、BA。采用 32 析因设计 ,进行NAA、BA配合试验 ,确定叶轴不定芽分化的最佳生长调节物质配比为NAA 0 5mg L +BA 2 0mg L ,此时不定芽分化率 73 47% ;小叶的不定芽分化的最佳配方为MS +NAA 1 0mg L +BA 2 0mg L ,不定芽分化率为 2 0 5 %。
The effects of growth regulators on regeneration of compound leaves of fast growing Robinia pseudoacacia were studied. Base on MS medium, four kinds of auxin IAA,IBA,NAA,2,4 D and three kinds of cytokinin BA,KT,ZT were arranged in monofactoral experiments. NAA and BA were selected to be the best regulators for regeneration of folioles and raches of the compound leaf. Then a 3 2 factorial design was used to determine the best combination of NAA and BA with different concentration. The results indicated that MS+NAA 0 5mg/L+BA 2 0mg/L was the best medium for adventitious bud regeneration of the raches with the differentiation rate of 73 43%, and to the folioles, it was MS+NAA 1 0mg/L+BA 2 0mg/L with the differentiation rate of 20 5%.
