摘要 目的 探讨髓样分化蛋白 - 2 (myeloiddifferentiationprotein - 2 ,MD - 2 )在内毒素(LPS)对内皮细胞核因子 -кB(NF -кB)激活中的作用。 方法 以ECV30 4细胞为模型 ,逆转录聚合酶链反应 (RT -PCR)方法检测内皮细胞MD - 2mRNA的表达 ;转染MD - 2功能突变体 ,观察MD - 2在内毒素对内皮细胞NF -кB激活和内皮细胞分泌IL - 8中的作用。 结果 内皮细胞有MD - 2mRNA的表达 ,转染MD - 2的突变体能明显抑制内毒素对内皮细胞NF -кB的激活和内皮细胞IL - 8的分泌。 结论 MD - 2在内毒素对内皮细胞NF -κB激活中发挥着不可缺少的作用 ,且在内毒素对内皮细胞激活 /损伤效应中占有重要的地位。 Objective To investigate the role of myeloid differentiation protein-2 (MD-2) in lipopolysaccharide (LPS) inducing NF-кB activation of endothelial cells. Methods ECV 304 cells were used as example models and the expression of MD-2 mRNA analyzed with RT-PCR. The role of MD-2 in LPS inducing NF-кB activation of the endothelial cells and in endothelial cell secreting IL-8 was explored by using mutant MD-2 cDNA with gene transfection. Results MD-2 mRNA was expressed in human endothelial cells. Transfection of mutant MD-2 cDNA could obviously inhibit LPS inducing NF-кB activation of endothelial cells and endothelial cells secretingIL-8 from the endothelial cells. Conclusion MD-2 may play a crucial role in LPS inducing NF-кB activation of endothelial cells as well as in LPS activating/injuring endothelial cells.
