摘要
目的 了解醋酸铅对人外周血淋巴细胞HPRT基因位点的影响 ,以探讨铅的致突变性。方法 采用多核细胞法体外研究醋酸铅对人外周血淋巴细胞HPRT基因位点的影响及突变频率。结果 0 2 5mmol/L醋酸铅即可诱发人外周血淋巴细胞HPRT基因位点突变 ,且突变频率随着染毒浓度的增加而升高 ,突变频率Y (1 0 -3 )与醋酸铅浓度C (mmol/L)之间存在一定的剂量 -效应关系 ,直线方程分别为 :Y =1 0 5 6 6 +0 1 6 6 1C (r=0 95 82 )和Y =1 0 76 2 +0 1 987C (r =0 94 34)。结论 在一定条件下 ,醋酸铅对人外周血淋巴细胞HPRT基因位点及突变频率有影响 。
Objective To study the mutational effect of PbAc 2 on HPRT gene of human peripheral lymphocytes in vitro.Methods A simple and sensitive method,cytokinesis block(CB) assay was used to detect the frequency of HPRT locus mutation in vitro induced by PbAc 2 and its dose effect relationship.Results The test showed that PbAc 2 could induce HPRT locus mutation in human lymphocyte and there existed a linear relationship between the concentration of the heavy metal saline and the mutation frequency(MF) of HPRT gene.Conclusions PbAc 2 could result in HPRT locus mutation in lymphocytes under this experimental conditions.It is expected that this assay might be used to study the environmental exposed population in future.
