摘要
本文报告了正常雄性家兔小肠粘膜细胞核提取液,经 38%饱和废硫酸铵沉淀、DEAE-纤维素层析、DNA-纤维素层析、羟基磷灰石层析、Sephadex G—200凝胶过滤,最终得到纯化5200倍的DHCC受体40μg,产率为9%。
The technique in use in the purification of the rabbit intestinal receptor for 1, 25 (OH2) D3 included ammonium sulfate precipitation, DEAE-cellulose, DNA - cellulose, hydroxylapatite and Sephadex G -200 gel-exclusion Chromatography. This kind of purification scheme resulted in purification of the receptor protein by 5200 - fold with 9% yield.On analysis, the purified protein on polyacrylamide gel electrophoresis contains sodium dodecyl sulfate. Isoelectric focusing and isotachophoresis suggested that the protein is homogeneous and characterized by having a molecular weight of approximately 65 000 and a PI of 6.1.
